Methylation studies in yellow-bellied marmots

ABSTRACT: An Infinium microarray platform (GPL28271, HorvathMammalMethylChip40) was used to generate DNA methylation data from blood samples from yellow-bellied marmots (Marmota flaviventris). DNA methylation data from n=159 blood samples. All samples were collected as part of a long-term study of a free-living population of yellow-bellied marmots in the Gunnison National Forest, Colorado (USA), where marmots were captured and blood samples collected biweekly during the their active season (May to August). Genomic DNA was extracted with Qiagen DNeasy blood and tissue kit.

Project description:An Infinium microarray platform (GPL28271, HorvathMammalMethylChip40) was used to generate DNA methylation data from many tissues of the naked mole rat We generated DNA methylation data from n=289 tissues. All tissues except for skin biopsies and blood were obtained from frozen tissue collection that were euthanized for other studies. Skin biopsies (2 mm punch) were collected from the backs of the animals under local anesthesia. Blood samples were collected from the tails. The n=3 induced pluripotent stem cells from NMR were generated as described in (Tan et al 2017., PMID: 29107597). As control set for the iPS study, we used n=3 fibroblasts samples. Gene expression shifts in yellow-bellied marmots prior to natal dispersal

Project description:We evaluated genome wide expression patterns in wild yellow-bellied marmots (Marmota flaviventris) prior to either dispersing from or remaining philopatric to their natal colony site. We identified differential expression of genes involved in lipid metabolism and muscle generation. Further, we found a network of genes involved in extra-cellular immune function and MHC class II pathways that increased in dispersers. Our results provide novel insights into the fundamental molecular changes required for natal dispersal in vertebrates.

Both whole blood (96) and remote biopsy (24) samples were obtained from a captive population of zebras maintained in a semi-wild state. After eliminating samples with low confidence for individual identity and age, we retained 76 blood samples and 20 biopsy samples, totaling 96 zebra samples.
