Genome-wide RNAseq of liver tissue derived from humanized mice fed by CDA-HFD and treated with CLDN1 mAb or vehicle Control

Project description:Six-week old Fah−/−/Rag2−/−/Il2rg −/− (FRG) –NOD breeding mice were intravenously injected with 1.5 x 10^9 plaque forming units (pfu) of an adenoviral vector encoding the secreted form of the human urokinase-like plasminogen activator (Ad-uPA). Forty-eight hours later, 10^6 PHH were injected intrasplenically. Successfully transplanted mice were fed with CDA-HFD for 16 weeks and then treated with humanized CLDN1 specific mAb 500 µg or vehicle for additional 8 weeks. RNA was extracted from non-tumorous liver tissue of 4 NASH fibrosis mice treated with CLDN1 mAb, 3 vehicle Control treated NASH fibrosis mice as well as 3 humanized mice fed by regular diet. Libraries were sequenced on the Illumina HiSeq 4000 as Single-Read 50 base reads.

Project description:To identify genes that may regulate distinct or overlapping functions of IRF-3 and IRF-7, primary human macrophage preparations were transduced with adenoviral vectors: Ad-GFP (control), Ad-F3 (expressing the active form of IRF-3, IRF-3 5D), or Ad-F7 (expressing the active form of IRF-7, IRF-7 D247-467) and evaluated by microarray analysis. RNA was collected 24 hours post-transduction with Ad-GFP, Ad-F3 and Ad-F7 and subjected to microarray analysis. Submited tables show the average of 7 donors.

Experiment Overall Design: Presented are gene expression data from: Experiment Overall Design: Dataset 1: Mouse liver 48h after i.v. infection of p53-/- mice with Ad TRF2DBDM compared to Ad GFP (2x10^10 PFU) (GSM103546, GSM103547) and Experiment Overall Design: Dataset 2: p53-/- heptaocellular carcinoma (HCC) from mTERC+ HBs (GSM103548, GSM103549, GSM103553) compared to mTERC-, HBs (GSM103550, GSM103551, GSM103552) transgenic mice. Experiment Overall Design: All mice were in a C57BL/6J background. Experiment Overall Design: Dataset 1: Liver RNA was extracted 48h after adenoviral infection (n=3 mice/group), samples were pooled for hybridization on Affymetrix microarrays. Experiment Overall Design: Dataset 2: RNA was extracted from HCC of 12-15month old mTERC+ and mTERC- mice (n=3 mice/group). Individual tumors were hybridized on Affymetrix microarrays. Experiment Overall Design: Differentially expressed gene clusters were identified using Array Assist 4.0 (Stratagene).
