Ex vivo scRNA-Seq of CD4+ cells in T-bet deficiency and wild type

ABSTRACT: Starting from frozen PBMCs of a patient with T-bet deficiency and a wild type control, CD4+ cells were enriched by magnetic sorting. The cells were either activated with plate-bound anti-CD3/anti-CD28 monoclonal antibodies for 14 hours or left untreated. Single-cell RNA sequencing (scRNA-Seq) was performed to study differentially expressed genes in the T helper cell compartment in T-bet deficiency.

CD38-NAD+ Immuno-Metabolic Axis Regulates Potent Anti-Tumor T Response

Project description:Spleens from the B6 mice were isolated and single cell suspension was made. CD4 T cells were purified from the splenocytes using magnetic bead separation. Briefly, Splenocytes were incubated with biotinylated antibody cocktail consisting of antibodies (Biolegend) to CD19, B220, CD11b, CD11c, NK1.1, Gr1, CD25 CD8. After a wash step, cells were incubated with streptavidin conjugated magnetic particles (BD Biosciences). After washing, CD4 T cells were isolated by applying a magnetic field and removing the untouched cells. Purified CD4 T cells were then activated with plate-bound anti-CD3 plus anti-CD28 in presence of either Th1 or Th17 or Th1/17 polarizing condition for 3 days. Total RNA from the 3 days differentiated Th1, Th17 and Th1/17 cells was isolated using mirVana miRNA isolation Kit (Invitrogen).

Transcription profiling of human Parp-1 deficient and wild type T cells following CD3 and CD3/CD28 activation

Project description:Primary T cells were isolated from spleen of Parp-1-/- and wild-type mice by magnetic depletion of non-T cells using a MACS Pan-T Cell isolation kit, according to the manufacturer´s instruction (Mintenyi Biotec, Bergisch Gladbach, Germany). Purity was assessed by flow cytometry analysis using antibodies against CD3, CD4 and CD8 and all preparations were more than 98% pure of T cells. The cells were activated with plate-bound anti-mouse CD3 (clone 145-2C11) (5 microg/ml) in the absence or the presence of anti-mouse CD28 (clone 37.51) (5microg/ml) both from BD PharMingen (San Diego, CA) and culture for 3.5 h in RPMI 1640 medium (BioWhittaker) supplemented with 10% FCS, 2mM L-glutamine, 5x10-5 M 2-mercaptoethanol (Sigma), 2.5 microg/ml fungizone, 100 IU/ml penicillin, and 10 microg/ml streptomycin.

Differential effects of inhibition of bone morphogenic protein (BMP) signalling on T-cell activation and differentiation

Project description:Dorsomorphin is a small molecule inhibitor of type I bone morphogenic protein receptors (BMPRs). We have found that dorsomorphin affects a wide range of T cell function. In order to obtain the bigger picture of the effects of DM in T cell activation. transcriptomic analysis was performed using mouse primary CD25-CD4+ T cells with either DM (4 μM) or vehicle in the presence or absence of stimulation by anti-CD3 and -CD28 antibodies. Mouse CD4+ T cells were prepared and cultured with or without stimulation by plate bound anti-CD3 mAb and soluble anti-CD28 mAb for 1 hour. Subsequently, 4 μM DM or DMSO was added and cells were incubated for 6 hours. Total RNA was isolated using Qiagen RNeasy mini kits.
