Beneficial effects of Allulose for age-associated sarcopenia

ABSTRACT: Allulose has been shown to moderate obese related dysmetabolism. In previous study, we found that allulose increased the muscle weight in diet-induced obese mice. It is there for hypothesized that allulose alleviated sarcopenia. The 12 month old mice were fed AIN93G with allulose. Allulose supplementation improved the muscle loss in aging mice.

Luteolin protects against obese sarcopenia in mice with high-fat diet-induced obesity by ameliorating inflammation and protein degradation in muscle

Project description:Although there are studies on obesity and sarcopenia, our understanding of obesity-mediated sarcopenia is insufficient. Additionally, no studies have investigated the application of luteolin in sarcopenia. We attempted to determine the effect of luteolin on obese sarcopenia in mice with high-fat diet (HFD)-induced obesity. Luteolin exerted suppressive effects on obesity, inflammation, and protein degradation in HFD-fed obese mice. Luteolin inhibited lipid infiltration into the muscle and decreased p38 and JNK activity and mRNA expression of inflammatory factors, such as TNFα, Tlr2, Tlr4, MCP1, and MMP2, in the muscle. Suppression of muscle inflammation by luteolin led to the inhibition of myostatin, FoxO, atrogin, and MuRF expression, which reduced protein degradation and improved muscle function. This is the first study to demonstrate that luteolin exerts a protective effect against obese sarcopenia, as its anti-obesity and anti-inflammatory activities inhibited protein degradation. Therefore, luteolin may be a useful supplement for prevention of obese sarcopenia.

Epigenome analysis of muscle tissue samples from male participants of the Hertfordshire Sarcopenia Study (HSS) and Hertfordshire Sarcopenia Study extension (HSSe)

Project description:Genome wide DNA methylation profiling of muscle tissue genomic DNA. The Illumina Infinium MethylationEPIC array was used to obtain DNA methylation profiles across approximately 850,000 CpGs. Samples were male, taken from both the Hertfordshire Sarcopenia Study (HSS, n=34) and the Hertfordshire Sarcoepnia Study exension (HSSe, n=44), to determine DNA methylation associations with measures of muscle mass and function. We further aimed to test whether differences in the muscle methylome are associated with sarcopenia in older individuals. We found significant associations between DNA methylation in the muscle tissue and sarcopenia, as well as individual measures of muscle mass and function, enriched around EZH2 binding sites.

Project description:Purpose: Sustained muscle fiber-specific mTORC1 activity, through deletion of the mTORC1 upstream inhibitor Tsc1, drives progressive muscle wasting and weakness reminiscent of sarcopenia. We aimed to characterise gene expression changes coinciding with the development of sarcopenia-like features in TSCmKO mice. Methods: Extensorum digitorum longus (EDL) muscles from 3- (weak phenotype) and 9-month-old (severe phenotype) TSCmKO and littermate control mice (Tsc1 floxed, HSA-Cre negative) treated with either vehicle or rapamycin (8 mg.kg.min-1) for 3 days were processed and sequenced. Results: The principal component analysis showed two major gene expression patterns. The first pattern represents an aging-like pattern with gene expression remaining stable between 3 and 9 months in wild type mice, but changing between 3 and 9 months in TSCmKO mice. The second pattern seems to represent early alterations in TSCmKO mice compared to WT, which subside between 3 and 9 months in TSCmKO. Importantly, when comparing changes in the gene expression in TSCmKO mice with natural aging gene expression profiles, there was a significant overrepresentation of commonly increasing and decreasing genes and an underrepresentation of oppositely regulated genes. Likewise, many of the top enriched GO terms for natural aging gene expression profiles were also enriched for genes changing the expression in the premature aging TSCmKO mice. Conclusions: Sustained, muscle fiber-specific mTORC1 activity drives sarcopenia-like gene expression programs, and the hyperactive mTORC1 seen in sarcopenic muscle may therefore contribute to sarcopenia.

The transcriptome profile in skeletal muscle of young male mice and aging male mice fed with or without eicosapentaenoic acid (EPA)

Project description:Age-related sarcopenia is associated with a variety of changes in skeletal muscle. These changes are interrelated with each other and associated with systemic metabolism, the details of which, however, are largely unknown. Eicosapentaenoic acid (EPA) is a promising nutrient against sarcopenia and has multifaceted effects on systemic metabolism. Although several human studies have suggested that EPA supplementation protects against sarcopenia, the causal relationship of EPA supplementation and an increase of muscle strength has poor evidence in vivo. We demonstrated that aging skeletal muscle in male mice shows lower grip strength and fiber type changes, both of which can be inhibited by EPA supplementation irrespective of muscle mass alteration. We hypothesized that the aging process in skeletal muscle can be intervened by the administration of EPA, via transcriptomic changes in skeletal muscle. This analysis revealed fast-to-slow fiber type transition in aging muscle, which was partially inhibited by EPA.

Results: Muscle transcriptomic profiles demonstrate a prominent signature of mitochondrial dysfunction in sarcopenic patients. Conclusions: Our study supports the fundamental role of mitochondrial dysfunction in driving pathological muscle and mobility decline in the elderly.
