Project description:Left cardiac ventricles and quadriceps femoris muscles were collected from young adult (16-17 wk) and early aging (86 wk) C57BL6/J mice (n=4, 2 male 2 female). Proteins were extracted in 200 µL RIPA buffer and Halt protease/phosphatase inhibitor with a hand-held sonicator followed by centrifugation, reduced with dithiothreitol, alkylated with iodoacetamide, then digested with modified sequencing grade trypsin overnight at 37degC using a filter-assisted sample preparation protocol. The peptides were fractionated using Thermo high-pH reversed phase columns into 8 fractions. Label-free bottom-up mass spectrometry was performed using data dependent acquisition on a Thermo Orbitrap Q-Exactive HF instrument using typical settings.

Project description:Analysis of angiotensin II effect on left ventricle at gene expression level. The hypothesis tested in the present study was that angiotensin II treatment may affect gene expression in left ventricle in a strain specific manner. Results provide important information about which genes respond to angiotensin II in C57Bl/6N male mice compared to their C57Bl/6J counterparts. Total RNA obtained from isolated left ventricles from C57Bl/6J and C57Bl/6N mice subjected to 48 hours of angiotensin II infusion, via osmotic mini-pumps (500ng/kg/h) implanted sub-cutaneously, compared to sham operated controls.

Effect of Tubeimoside I (TA) on the innate immunity in C57BL/6 mice

Project description:A saponin monomer Tubeimoside I (TA) was found to have potential adjuvant activity. Since the adaptive immune response is mediated by innate immunity, we took the immune injection site (quadriceps femoris muscle) as the local immune microenvironment, and injected TA (50 μg) into quadriceps muscle to prepare samples. Muscles injected with PBS were served as the negative control. RNA was extracted 4 hours after drug treatment, and samples were prepared for whole-gene transcriptome expression profile microarray detection.
