The role of hsa_circ_0000073 in Osteosarcoma

ABSTRACT: Osteosarcoma (OS) is one of the leading causes of cancer mortality in children and teenagers. Patients with OS have been shown to have a poor prognosis and may suffer from potential distant metastasis. Emerging evidence has revealed that circular RNAs (circRNAs) are involved in OS progression. In this paper, we aim to explore the function of hsa_circ_0000073 in the progression of OS. OS cells transfected with sh‐NC or sh-circ_0000073 were enrolled in this study.We used microarray analysis to identify the mRNA expression in the cells.

Microarray profiling analysis of whole transcriptome expression in osteosarcoma

Project description:Osteosarcoma (OS)is a rare primary malignant bone tumor in adolescents and children with a poor prognosis. Identification of prognostic genes lags far behind the advance of the treatments. We identified differential genes by microarray analysis from paired OS tissues. Hub genes, gene set enrichment analysis, and pathway analysis were performed to gain an insight into the pathway alterations of OS. These results showed CPE could be served as a prognostic factor in osteoblastic OS and should be further investigated as potential therapeutic target. The present study evaluated the whole transcriptome expression of osteosarcoma progression and provided novel therapeutic targets for advanced osteosarcoma.

Acquisition of Cancer Stem Cell Properties in Osteosarcoma Cells by Defined Factors

Project description:Cancer stem cells (CSCs) are proposed to be unique subsets of clones within a tumor, attributed with tumor progression, resistance to chemotherapy, and metastases. However, little is known about the molecular mechanisms regulating the acquisition and maintenance of CSC properties in cancers, especially in rare cancers, such as osteosarcoma (OS). In this study, we generated OS cells with CSC properties following transduction of defined factors. This method enabled us to obtain abundant OS CSC material. The results of our study could facilitate a better understanding of the mechanisms of OS progression, and aid in the development of new therapies targeting CSCs in patients with OS.

Project description:Osteosarcoma in dogs is a spontaneously occurring disease with a global tumor gene expression signature indistinguishable from human pediatric tumors and clinical progression is remarkably similar. Unlike human OS, canine OS is a highly heritable disease with some large and giant dog breeds at >10x increased risk. We did a genome wide association study of osteosarcoma using the Illumina CanineHD genotyping array in three breeds: greyhound (mortality from OS = 26%), rottweiler (17%) and Irish wolfhound (IWH, 21%) and identified 33 inherited risk loci explaining 55 to 85% of phenotype variance in each breed. Data created for a genome-wide association studio of heritable osteosarcoma risk factors in dogs, including 267 racing greyhounds (153 affected (A) + 114 unaffected (U)), 135 rottweilers (80 A + 55 U), 141 IWH (76 A + 65 U) and 19 AKC greyhounds using the Illumina CanineHD array (~170,000 SNPs genomewide)

Plasma profiling of miRNAs in mouse model of osteosarcoma.

Project description:Osteosarcoma (OS) is the primary bone tumor in children and young adults. Currently, there are no reliable, non-invasive biological markers to detect the presence or progression of disease, assess therapy response, or provide upfront prognostic insights. Using a qPCR-based platform that analyzes more than 750 miRNAs, we analyzed control and diseased-associated plasma from a genetically engineered mouse model of OS to identify a profile of four plasma miRNAs.
