Large scale analysis of leaf gene expression upon treatment of apple seedlings with acibenzolar-S-methyl or water

ABSTRACT: Investigation of whole genome gene expression level changes in the youngest expanded leaves of open-pollinated Golden Delicious seedlings either sprayed with reverse osmosis water or acibenzolar-S-methyl (ASM). Treatment was performed by spraying sprayed to runoff (with a pressurized hand sprayer) with the commercial product Bion 50 WG (Syngenta, Basel, Switzerland; 50% of ASM) prepared in reverse osmosis water at a final concentration of 0.4 g/L. The youngest developped leaf of each seedling was sampled 3 days after the treatment.

Defense signaling in potato

Project description:Potato cultivar Russet Norkotah was grown in a growth chamber at 16 hour light/8 hour dark at 22°C fertilized with Osmocote. Plants were from tissue culture, transplanted to soil and used approximately 4 weeks after transplanting. RNA was extracted from leaves using Trizol. Plants were sprayed with one of four treatments, either 1mM salicylic acid, 100 µM methyl jasmonate, 1 mg/ml arachidonic acid or 100 µM methyl jasmonate plus 1 mM salicylic acid together. Samples were harvested at 3, 24 and 72 hours after spraying. Each salicylic acid or arachidonic acid treatment has 3 biological replicates done in parallel, using leaves pooled from 2 plants. The control for SA or AA treated plants were water sprayed plants, from which leaves of 3 different plants were pooled at each timepoint, done side-by-side with the treated samples. Methyl jasmonate or MJ+SA treatments had 2 biological replicates each, using leaves pooled from 2 plants. A separate set of water sprayed plants (from the SA and AA control treatments) were used as controls, and pooled leaves from 3 plants were used at each time-point, and done in parallel with the treated samples. Gene expression analysis of Pseudomonas putida KT2440 Wild Type and its exopolysaccharide mutants under water stress

Project description:Alginate, a major exopolysaccharide (EPS) produced by P. putida, is known to create hydrated environments and alleviate the effect of water limitation. In addition to alginate, P. putida is capable of producing cellulose (bcs), putida exopolysaccharide a (pea), and putida exopolysaccharide b (peb). However, unlike alginate, not much is known about their roles under water limitation. Hence, in this study we examined the role of different EPS under water stress. To create environmentally realistic water stress conditions as observed in soil, we used Pressurized Porous Surface Model (PPSM). Our main hypothesis was that under water stress, absence of alginate would be compensated by the other EPS. To test our hypothesis, we investigated colony morphologies and whole genome transcriptomes of P. putida KT2440 WT and its mutants deficient in either alginate or all known EPS

Transcription profiling of human airway smooth muscle cells after treatment with glucocorticoids and Protein Kinase A

Project description:Glucocorticoids (GCs) and protein kinase A (PKA)-activating agents (beta-adrenergic receptor agonists) are mainstream asthma therapies based on their ability to prevent or reverse excessive airway smooth muscle (ASM) constriction. Their abilities to regulate another important feature of asthma - excessive ASM growth are poorly understood. Recent studies have suggested that GCs render agents of inflammation such as interleukin 1beta and tumor necrosis factor alpha mitogenic to ASM, via suppression of (antimitogenic) induced cyclooxygenase-2-dependent PKA activity. To further explore the mechanistic basis of these observations, we assessed the effects of epidermal growth factor and interleukin 1beta stimulation, and the modulatory effects of GC treatment and PKA inhibition, on the ASM transcriptome by microarray analysis. Experiment Overall Design: Human ASM cultures were generated from trachea of 4 unidentified donors and used in passage 5-8. These cells were used to generate cells stabely expressing a PKA inhibitor (PKI) or GFP (control), which were then grown to confluence, serum starved for 24h, and stimulated with IL-1b, EGF,or both (E+I) in presence or absence of Fluticasone (Flu, 30 min pretreatment). After 8h stimulation cells were washed and total RNA isolated using TRIzol as per manufacturer's protocol. Gene expression was analyzed in 4 different cultures derived from 4 different donors (for a total of 3-4 microarrays per condition).
