Transcriptional profiles of placentas from systemic lupus erythematosus and normal full-term pregnancies.

ABSTRACT: We report transcriptional profiles of placentas from systemic lupus erythematosus pregnancies and normal full-term pregnancies. We recruited 8 pregnant patients with SLE, and 8 age-matched healthy pregnant women as normal controls. RNA-seq strand-specific libraries were constructed using the VAHTS Total RNA-seq (H/M/R)Library Prep Kit. Differentially expressed genes were identified using Cuffdiff. The P-value significance threshold in multiple tests was set by the false discovery rate (FDR). The fold-changes were also estimated according to the FPKM in each sample.The differentially expressed genes were selected using the following filter criteria: FDR <0.05 and fold-change >2.

Project description:It has been well documented that pre-eclampsia and unexplained fetal growth restriction (FGR) has a common etiological background, but little is known about the linkage al the molecular level. we have performed global gene expression profiling by oligonucleotide microarrays for placentas from pre-eclamptic, unexplained FGR and normal pregnancies to further elucidate the mechanisms underlying the development of these two disorders. The total number of samples used was 8 from pre-eclampsia, 8 from normotensive pregnancies with FGR and 8 from those without FGR.

Transcription profiling of decidua basalis from women with normal pregnancies or preeclampsia and/or fetal growth restriction

Project description:The aim of the study was to compare gene expression profiles in decidua basalis from cases with complicated pregnancies to those from healthy pregnant controls. Cases included pregnant women with preeclampsia (PE) and/or fetal growth restriction (FGR). Decidual tissue was obtained by vacuum suction of the placental bed after the placenta was delivered. Women with PE and/or FGR were included as cases. PE was defined as persistent hypertension (blood pressure (BP) ¡Ý 140/90 mm Hg), plus proteinuria (¡Ý0.3g/24 h or ¡Ý2+ according to a dipstick test), developing after 20 weeks of pregnancy. FGR implied birth weight under 2 standard deviations (SD) below the expected birth weight, as related to gestational age (GA) and sex. Due to tissue sampling procedures, only cases delivered by cesarean section (CS) were included. Healthy women with normal pregnancies, undergoing CS for various reasons considered irrelevant to the aim of this study (e. g. breech presentation and previous CS), served as controls. Tissue was immediately submerged in a RNA stabilisation solution (RNAlater, Ambion, Huntingdon, U.K.), incubated at 4¡ãC overnight and stored at -80¡ãC

MeDIP and hMeDIP assays using human placentas of obese and lean (healthy weight) pregnancies

Project description:Maternal obesity has both a direct effect on the outcome of pregnancy but also programs the fetus for obesity and metabolic syndrome in later life. Due to its roles at the interface between mother and fetus, the placenta transduces and mediates the effects of the obese intrauterine environment on the fetus. Here we have generated genome-scale high-resolution maps of 5mC and 5hmC in human placentas of obese and healthy weight pregnancies. Widespread alterations to the placental epigenome identified across the genome in the setting of maternal adiposity are primarily characterized by increase in 5mC and reciprocal decrease in 5hmC. This is also evident at the two pregnancy-associated gene clusters on chromosomes 17 and 19. In addition, maternal adiposity is associated with downregulation of ten-eleven translocation (TET) and isocitrate dehydrogenase (IDH) family genes and is accompanied by decreased levels of Î±-ketoglutarate (Î±KG) in the placenta. Of note, Î±KG levels exhibit a close positive correlation with both ascorbate and aconitate levels and less substantial but significant association with global 5hmC levels and placenta-specific gene expression. These observations suggest a mechanistic linkage between obesogenic environment and conversion efficiency of 5mC to 5hmC and highlight the idea that TET-mediated active DNA demethylation pathway can sense and respond to metabolic derangements and plays a role in subsequent cellular adaptation. Collectively, this study demonstrates that altered DNA demethylation process is likely one of the mechanisms underlying metabolic modulation of the placental epigenome with maternal adiposity. Project description:IUGR (Intra-Uterine Growth Restriction) refers to a condition where the foetus does not reach its growth potential in utero. It is supposed to be often linked with placental dysfunction, especially of vascular origin. In this study, 4 pools of three placentas from human normal pregnancies and 4 pools of three placentas from IUGR human pregnancies, obtained after caesarean section near normal term , were used to prepare RNA. The cDNAs prepared from these RNA were hybridized to a Nimblegen expression array in order to detect differences in gene expression between normal and pathological placentas.

Single-cell resolution of longitudinal blood transcriptome profiles in rheumatoid arthritis, systemic lupus erythematosus and healthy control pregnancies

Project description:Rheumatoid arthritis (RA), Systemic Lupus Erythematosus (SLE) are genetically the most closely related of the major rheumatic diseases as they share components of the type I IFN signalling pathway linked to treatment response and disease activity. One difference between the two is how they respond to pregnancy; while RA often improves, SLE is prone to flare. Multiple mechanisms contribute to immunological tolerance of the foetus during pregnancy. More research involving comparative analyses is needed, however, as understanding how pregnancy-related immune-modulation differs in RA and SLE can potentially lead to better treatment options. We provide data from a cohort of women with universal accessibility to health care that have been followed closely throughout their pregnancies, are well regulated and have low disease activity. We analysed samples both by bulk- and scRNA-seq, and ran a cell-type estimation, validated by flow cytometry, before combining this in a cell-type adjusted analysis. This novel comparative approach gives an improved resolution of expression profiles in RA and SLE pregnancies.
