Comparison of the responses of Mycobacterium tuberculosis and Mycobacterium bovis to nutrient starvation

ABSTRACT: Transcriptional profiling of Mycobacterium tuberculosis H37Rv and Mycobacterium bovis Ravenel before (i.e., log-phase control) and at 24h and 96h after nutrient starvation. The transcriptional profile of the two strains were compared at log phase, and after 24h and 96h of starvation. In addition, the response of each strain to starvation was examined after 24h and 96h of starvation.

Transcriptional response of bovine moncocyte-derived macrophages to infection with strains of Mycobacterium bovis and Mycobacterium avium subspecies paratuberculosis

Project description:Mycobacterium bovis (M. bovis) and Mycobacterium avium subspecies paratuberculosis (MAP) are important pathogens of cattle, causing bovine tuberculosis and Johne’s disease respectively. M. bovis and MAP infect residential macrophages in the lung and intestines respectively and subvert the macrophage biology to create a survival niche. To investigate this interaction we simultaneously studied the transcriptional response of bovine monocyte-derived macrophages to infection with two strains of M. bovis (AF2122/97 and G18) and two strains of MAP (C & L1).

Gene Expression profiling of M. bovis BCG strain under hypoxia

Project description:Objective of the study is to find out the differentially regulated genes in Mycobacterium bovis BCG subjected to hypoxic condition. Gene expression profiling was carried out using Agilent microarray platform. Keywords: Hypoxia response Mycobacterium cells were lysed by bead beating in AL Buffer of Qiagenâs bacterial RNA kit. RNA was purified as per manufacturerâs instructions. RNA quality control was done using Agilent 2100 Bioanalyzer lab on a chip. Total RNA (500ng) of Mycobacterium under hypoxia and log phase were labelled with Cy3 and Cy5 (Perkin Elmer) dyes respectively using Ambion Message AmpII bacteria kit as per manufacturer recommended protocol. To eliminate dye bias, technical replicates were labelled in the reverse direction i.e log phase culture with Cy5 and hypoxia with Cy3. 825ng each of labeled control cRNA was mixed with 825ng of treated labelled cRNA, mixed with appropriate amount of blocking buffer and hybridization buffer (Agilent Technologies). The hybridization mix was applied on to the backings and hybridized to custom designed 60mer M.tuberculosis H37Rv microarray using sure hyb chambers at 65degC for 17 hours. Slides were washed with gene expression wash buffer 1 and 2 (Agilent Technologies) followed by Acetonitrile and scanned at 5 micron resolution using Agilent scanner. Automated feature extraction was done using Agilentâs Feature Extraction Software. Statistical analysis of microarray data was done using GeneSpring GX and Biological analysis of the differentially regulated genes were done using Genotypics Biointerpreter -Web based tool for biological interpretation of gene list in a click of mouse.

An important and understudied aspect, however, is the extent to which regulatory features affecting the expression of such shared genes are present in both species. Here we report on an analysis of whole transcriptome maps for an important member of the TB complex Mycobacterium bovis and a closely related model organism for studying mycobacterial pathogenicity Mycobacterium marinum.

Transcriptome analysis of VapC21 overexpression and deletion in Mycobacterium tuberculosis.

Project description:We report the effects of VapC21 overexpression on Mycobacterium tuberculosis H37Rv strain. The total RNA was isolated from M. tuberculosis harboring either pTetR-int or pTetR-Int-vapC21 the expression was induced with 50 ng/ml anhydrotetracycline for 24 hrs. We report the effect of deletion VapC21 on the transciptional profile of Mycobacterium tuberculosis Erdman strain. For RNA-seq analysis, total RNA was isolated from mid-log phase culture of either parental or △vapC21 mutant strain.
