Th2/Th1 cytokine imbalance is associated with higher COVID-19 risk mortality

ABSTRACT: In this study we profiled 288 new serum proteomics samples measured at admission from patients hospitalized within the Mount Sinai Health System with positive SARS-CoV-2 infection. We first computed Th1 and Th2 pathway enrichment scores by gene set variation analysis and then compared the differences in Th2 and Th1 pathway scores between patients that died compared to those that survived.

Proteomic analysis of T cell differentiation

Project description:T helper cells (Th) play an important role guarding, regulating and modulating immune responses. The different Th lineages fulfill different tasks in response to infections. The two best defined subtypes are Th1 (involved in cellular immunity) and Th2 (humoral immunity). The cytokine environment after activation determines the differentiation path of a Th cell. We defined a strategy to investigate the differentiation signature of T helper cells with a proteomics approach. Murine primary naïve CD4+ T-cells from spleen were differentiated and activated in vitro. After 3 days of differentiation proteins were analysed. The proteomic profile of the Th1 and Th2 cells will help understand these phenotypes better, which is important in finding therapeutic targets for disease and for the development of effective vaccines.

Whole blood gene expression profiles associated with mortality in community-acquired pneumonia

Project description:We aimed to examine the differences in the gene expression profile in peripheral blood at hospital admission between patients with community-acquired pneumonia (CAP) who died and survived during hospitalization. Whole blood samples for genome expression profile analysis were obtained within 24 hours of hospital admission. Gen set enrichment analysis (GSEA) identified gene sets positively enriched in the patients who survived, mainly related with interferon- alpha response, apoptosis, and sex hormones pathways. Similarly, GSEA identified gene sets positively enrichment for the patients who died (oxidative stress, endoplasmic reticulum stress, oxidative phosphorylation, and angiogenesis pathways).
