Project description:Foliar analysis is very important for the nutritional management of crops and as a supplemental parameter for soil fertilizer recommendation. The elemental composition of plants is traditionally obtained by laboratory-based methods after acid digestion of ground and sieved leaf samples. This analysis is time-consuming and generates toxic waste. By comparison, portable X-ray fluorescence (pXRF) spectrometry is a promising technology for rapid characterization of plants, eliminating such constraints. This worked aimed to assess the pXRF performance for elemental quantification of leaf samples from important Brazilian crops. For that, 614 samples from 28 plant species were collected across different regions of Brazil. Ground and sieved samples were analyzed after acid digestion (AD), followed by quantification via inductively coupled plasma optical emission spectroscopy (ICP-OES) to determine the concentration of macronutrients (P, K, Ca, Mg, and S) and micronutrients (Fe, Zn, Mn, and Cu). The same plant nutrients were directly analyzed on ground leaf samples via pXRF. Four certified reference materials (CRMs) for plants were used for quality assurance control. Except for Mg, a very strong correlation was observed between pXRF and AD for all plant-nutrients and crops. The relationship between methods was nutrient- and crop-dependent. In particular, eucalyptus displayed optimal correlations for all elements, except for Mg. Opposite to eucalyptus, sugarcane showed the worst correlations for all the evaluated elements, except for S, which had a very strong correlation coefficient. Results demonstrate that for many crops, pXRF can reasonably quantify the concentration of macro- and micronutrients on ground and sieved leaf samples. Undoubtedly, this will contribute to enhance crop management strategies concomitant with increasing food quality and food security.

Project description:This study explores the use of short settling times as a strategy to enhance microbial selection and prevent bulking induced by nutrient shortage in an aerobic dynamic feeding (ADF) process using mixed microbial cultures (MMCs) for polyhydroxyalkanoate (PHA) production from fermentation digestate. A 5.0-L aerobic reactor was operated under three conditions: Long Settling (LS, 30 minutes), Short Settling (SS, 10 minutes), and Short Settling under Nutrient Shortage (SS-NS, 10 minutes with reduced nutrient load). Short settling significantly improved biomass settleability, reducing the Sludge Volume Index (SVI) from 126 to 25 mL g⁻¹, and promoted the formation of dense flocs enriched in PHA-accumulating bacteria, as confirmed by transmission electron microscopy. Process kinetics revealed enhanced substrate uptake rates and improved storage yields under SS conditions, despite an elevated C/P ratio. Notably, nutrient shortage conditions were inadvertently reached by the proliferation of Polytoma mirum in the feeding tank. Although Polytoma mirum did not affect VFA composition, its presence significantly reduced nitrogen and phosphorus concentrations. These nutrient shortage conditions were maintained for at least 30 days until the system began to exhibit stress, as demonstrated by an increased SVI (178.6 mL g⁻¹). Microbial community analyses indicated marked shifts: the eukaryotic assemblage transitioned from sessile to motile ciliates under SS, while bacterial diversity within the PHA‐accumulating fraction remained high, with key taxa such as Sphaerotilus and Neomegalonema becoming more prevalent under phosphorus-limited conditions. Overall, short settling not only improved microbial selection but also prevented bulking by retaining well-aggregated biomass, thereby mitigating nutrient shortage conditions.

Project description:Plasma-treated water (PTW) contains reactive oxygen and nitrogen species and can therefore induce mild oxidative stress in plants. In a previous study, PTW treatment of leaves resulted in both short- and long-term systemic signals. RNA-seq was applied to analyse the adaptive response triggered by PTW treatment and to further determine the mode of action of PTW in barley leaves and roots under short- and long-term conditions. The application of PTW to the leaves resulted in stronger long-term transcriptional changes in the roots. PTW treatment significantly increased root biomass, while it had no effect on shoot growth. In roots the signalling of auxin, ethylene, brassinosteroids and gibberellin was stimulated, while cytokinin signalling was impeded. Many genes encoding cell wall modifying enzymes, structural proteins and receptor kinases were upregulated in the roots in long-term response to PTW. This indicates that PTW treatment appears to affect root growth by promoting cell elongation. In addition, foliar treatment with PTW led to changes in root architecture: root length, surface area, diameter and number of forks increased in the long term, as did the content of soluble phenolic compounds. All these changes indicate that PTW could be a suitable stimulant for improving the root growth of plants.

Project description:The rapeseed crop (Brassica napus) is valued mainly for animal feed, food oil and biofuel production. This plant is particularly susceptible to many pathogens like parasitic plants, fungi or animals attacking aerial or root parts. Conventional plant protection products, used intensively in agriculture, have a negative impact on the environment (air and water quality, biodiversity, etc.) as well as on human health. There is therefore a growing demand for the development of new, more planet-friendly alternative protection methods such as biocontrol compounds. Natural rhamnolipids (RLs) can be used as elicitors of plant defense mechanisms. Indeed, these glycolipids, from bacteria secretome, are biodegradable, non-toxic and they are known for their stimulating and protective effects, in particular on rapeseed against filamentous fungi. Characterizing the organ responsiveness to defense stimulating compounds like RLs is missing. This analysis is crucial in the frame of optimizing the effectiveness of RLs against various diseases. A TMT labeling of the proteins extracted from the shoots and roots of B. napus has been performed and showed a differential pattern of protein abundance between them. These results have allowed identifying several protein families possibly involved in the differential responses observed in shoots and roots of rapeseed upon RLs treatment. In particular, quantitative proteomic analysis highlighted differential accumulation of parietal and cytoplasmic defense or stress proteins in response to treatments with RLs with a clear effect of the type of application (foliar spraying or root absorption). These results must be considered for further use of RLs to fight specific rapeseed pathogens.

Project description:We have found that MtFTb genes play a role in the response to LD conditions, both under vernalized and non-vernalized conditions. To explore the regulatory gene network downstream of FTb genes on a global scale, we performed RNA sequencing of the gene-edited Mtftb1/b2 vs WT plants. For this, we grew wild-type and gene-edited Mtftb1/2 plants under two different conditions: vernalised long days (VLD), where the plants were vernalized for 14 days and then grown for two weeks under LD photoperiod, with the aim of capture changes in expression profiles in the period in which the plant becomes physiologically committed to flower; the second condition was non-vernalised long days (NVLD), where non-vernalized plants were grown for 60 days, a crucial time point when wild-type plants typically undergo the transition to flowering. This approach allowed us to investigate the gene expression changes in the FTb1/2 mutant independent of vernalization effects.
