Transcriptome profiles of colonic stem cells of WT and AHR deficient mice treated with either DMSO or high affinity AHR ligand, FICZ for 4 hours

ABSTRACT: To identify aryl hydrocarbon receptor (AHR) dependent transcriptional changes in mouse colon stem cells, we performed RNA sequencing of colon organoids from wildtype and AhR deficient mice stimulated with an high affinity AHR ligand, 5nM FICZ for 4 hours.

miRNA expression in Th17 cells after AhR activation

Project description:Th17 cells play a major role in the pathogenesis of Rheumatoid Arthritis, and it is well known the involvement of the Aryl Hydrocarbon Receptor (AhR) during the differentiation and activation of these cells. Due to its function as a transcription factor, we tested wether AhR would mediate part of its function in Th17 cells through the transcription of microRNAs (miRNAs). Therefore, we first perform a microarray experiment to identify the miRNAs induced after AhR activation in Th17 cells. Under a supervised hierarchical clustering, we identify 224 miRNAs differentially expressed (fold-change ³ 2.0 and FDR < 5%). Among them, 22 miRNAs were up regulated exclusively in Th17 cells in the presence of FICZ.

AhR activation by TCDD in the ET, cortical epithelial cell line

Project description:Over activation of the aryl hydrocarbon receptor (AhR) by TCDD results ampng other phenotypes in severe thymic atrophy accompanied by immunosuppression. The link between thymic atrophy, skewed thymocyte differntiation and immunosuppression is still not fully resolved. This study investigates the TCDD elicted exprssion changes in the ET, cortical thymus epithelial cell line. Keywords: TCDD, AhR, thymic epithelial cells, thymic involution, thymus atrophy ET cells were grown to 80-90% confluence and treated with 5nM TCDD or solvent control (1,4-Dioxane, 0,05%V). 2h,4h and 6h after treatment, RNA was isolated using TRIzol and gene expression measured. All experiments wer done in biological duplicates.

Transcription profiling of a frog cell line in response to the aryl hydrocarbon receptor (AHR) ligands FICZ and TCDD

Project description:The aryl hydrocarbon receptor (AHR) mediates the toxic effects of environmental contaminants, such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Frogs are very insensitive to the toxic effects of TCDD. Experiment Overall Design: XLK-WG cells (ATCC) were grown to ~90% confluence in RPMI-1640 media supplemented with 20% FBS at 29 degrees and 5% CO2. Cells were subsequently exposed to 100 nM TCDD, FICZ, or DMSO vehicle (0.25%) for 3 hr prior to extraction of total RNA. Experiment Overall Design: Study included three complete biological replicates of each treatment.

Proteome and Phosphoproteome of AhR-dependent non-genomic signaling in endotoxin-activated macrophages

Project description:Emerging studies revealed an immunomodulatory role of the Aryl hydrocarbon receptor (AhR), a receptor sensing environmental contaminants, and involved in their detoxification. Besides its function as a transcription factor, AhR can participate in non-genomic signaling through ubiquitination and phosphorylation-dependent processes. In this study, a multi-PTM-omics approach, including proteome, ubiquitome, and phosphoproteome, was utilized to examine mechanisms of non-genomic AhR-signaling in endotoxin-activated monocyte-derived macrophages. This dataset entails proteome and phosphoproteome data.

AhR interactome in early embryonic development

Project description:The transcriptional program of early embryonic development is tightly regulated by a set of well-defined transcription factors that suppress premature expression of differentiation genes and sustain the pluripotent identity. It is generally accepted that this program can be perturbed by environmental factors such as chemical pollutants, however the precise molecular mechanisms remain unknown. The Aryl Hydrocarbon Receptor (AHR) is a widely expressed nuclear receptor that senses environmental stimuli and modulates target gene expression. Here, we show that ectopic activation of AHR during early differentiation disrupts the differentiation program via the chromatin remodeling complex NuRD. The activated AHR/NuRD complex altered the expression of differentiation-specific genes that control the first two developmental decisions without affecting the pluripotency program. These findings identify a novel mechanism that allows environmental stimuli to disrupt embryonic development through AHR signaling.
