ABSTRACT: Genotype dependent gene expression changes in the leaves of Illinois protein lines at 8 days after pollination (DAP). Illinois protein lines (IPL) comprise lines known as Illinois high protein (IHP), Illinois low protein (ILP), Illinois reverse-low protein (IRLP) and Illinois reverse-high protein (IRHP) lines. These lines represent over a century old experiment containing lines that represent various levels of nitrogen metabolism potential as a function of their grain protein concentration. Project description:According to the physiological change of the ear leaf, the developing ear leaf after pollination can be divided into three classes: mature leaves [ML, 0–14 days after pollination (DAP)]; early senescent leaves (ESL, 15–24 DAP); and later senescent leaves (LSL, 25–30 DAP). We harvested ear leaves at 12 DAP, 20 DAP, and 28 DAP, representing ML, ESL, and LSL, respectively. To identify genes involved in the leaf senescence process, we sequenced three cDNA libraries, ML (12 DAP), ESL (20 DAP), and LSL (28 DAP) using an Illumina HiSeqTM 2000.

iTRAQ of Wucai (Brassica campestris L.)leaf color

Project description:Chlorophyll plays critical roles in photosynthetic light harvesting, energy transduction and plant development. In this study, a novel wucai (Brassica campestris L.) germplasm with green outer leaves and yellow inner leaves at the adult stage (W7-2) was used to examine chlorophyll metabolism response to cold acclimation. A green leaf wucai genotype without leaf color changes named W7-1 was selected as the control to evaluate the chlorophyll metabolism changes of W7-2. Compared to W7-1, the contents of chlorophyll a (Chl a) and chlorophyll b (Chl b) in W7-2 were significantly reduced at five developmental stages (13, 21, 29, 37 and 45 days after planting (DAP). An iTRAQ-based quantitative proteomic analysis was carried out at 21 and 29 DAP according to the leaf color changes in both of genotypes. A total of 1409 proteins were identified, of which 218 showed differential accumulations between W7-2 and W7-1 during the two developmental stages.

Transcriptomic analysis of maize developing earshoot, IBMRIL X IHP1 hybrid

Project description:Maize earshoot is a metabolic sink espcially related to nitrogen metabolism. Studies on the transcriptomic and metabolic changes occuring in earshoot can provide interesting answers about the nitrogen metabolic potential of the maize variety under study. Intermated B73 X Mo17 recombinant inbred lines (IBMRIL) belong to a predominantly used mapping population. Illinois high protein (IHP) line is a metabolic extreme with respect to nitrogen mebolism with around 30% seed protein. Developing earshoots from this genotype grown at nitrogen-deficient and nitrogen-sufficient conditions were sampled, processed and analyzed through microarray technology. IBMRIL population was crossed to IHP to find the allelic interaction related to nitrogen metabolism. Microarray experiment was conducted using the developing earshoots from IBMRIL X IHP1 lines that deffered in their grain yield significantly. Keywords: Yield response
