Project description:Isoprene is a C5 volatile organic compound, which can protect aboveground plant tissue from abiotic stress such as short-term high temperatures and accumulation of reactive oxygen species (ROS). Here, we uncover new roles for isoprene in the plant belowground tissues. By analyzing Populus x canescens isoprene synthase (PcISPS) promoter reporter plants, we discovered PcISPS promoter activity in certain regions of the roots including the vascular tissue, the differentiation zone and the root cap. Treatment of roots with auxin or salt increased PcISPS promoter activity at these sites, especially in the developing lateral roots (LR). Transgenic, isoprene non-emitting poplar roots revealed an accumulation of O2 - in the same root regions where PcISPS promoter activity was localized. Absence of isoprene emission, moreover, increased the formation of LRs. Inhibition of NAD(P)H oxidase activity suppressed LR development, suggesting the involvement of ROS in this process. The analysis of the fine root proteome revealed a constitutive shift in the amount of several redox balance, signaling and development related proteins, such as superoxide dismutase, various peroxidases and linoleate 9S-lipoxygenase, in isoprene non-emitting poplar roots. Together our results indicate for isoprene a ROS-related function, eventually co-regulating the plant-internal signaling network and development processes in root tissue. Project description:To further explore the molecular mechanisms of P regulation in banana plants, we used RNA sequencing-based transcriptomic analysis for banana plants subjected to Pi deficit stress for 60 days.We detected 1900 significantly differentially expressed genes (DEGs) in aboveground plant parts and 7398 DEGs in root parts under low P stress. Gene ontology (GO) classification analysis showed that 156,291 GO terms belonging to molecular functions, 53,114 GO terms belonging to cellular components, and 228,544 GO terms belonging to biological processes were enriched in the aboveground and root components. A number of DEGs involved in energy metabolism-related processes, signal transduction, control of rhizosphere P activation, and Pi mobilization were found, which were confirmed by quantitative reverse-transcription PCR (RT-PCR) analysis. At the transcriptomic level, we detected 13 DEGs from different organs and with different functions in the time-course response to phosphorus deficiency stress. These DEGs may include some key genes that regulate the phosphorus network, increasing our understanding of the molecular mechanism of Pi homeostasis in banana.

Project description:The primary objective of this project is to determine the physiological and developmental differences between photoperiod and root induced tuberization responses in potato using genomic techniques. These experiments will establish baseline genomic and proteomic response associated with a partially characterized root-produced compound (TIF) that either substitutes for, or over-rides, the primary leaf perceived photoperiod induction signal in potato. The proposed work will answer the following questions. Are photoperiod and root induced tuberization responses the same? Do photoperiod and root induced tuberization trigger the same developmental processes in the stolon? How are photoperiod and root induced signals expressed in the genome of the leaf, stolon and roots? The data from the proposed experiments will prove invaluable in identifying key pathways in specific tissues to target in subsequent experiments in order to determine identify of, and establish the mode of action of the TIF compound. Project description:In this experiment we measured the transcriptional response of tomato plants (cv “Money maker”) when attacked belowground by the nematode M. inognita and, subsequently, aboveground by different (and common for this crop) biotic agents. Three weeks-old plants were exposed to nematodes for 5 days. At the fifth day the terminal leaflet of one of the first two true leaves was infected with the Cauliflower Mosaic Virus, or with the pathogen, or with the potato aphid Macrosiphum euphorbiae, or with the CMV-infected M. euphorbiae. For each belowground/aboveground combination treatment a set of control plants that received only the aboveground treatment was prepared. The infected leaflets of 5 biological replicates, each consisting of 1 plant, were collected 1, 2, 3, 4, 5 and 6 days after the onset of the aboveground treatment and flash frozen in liquid nitrogen. A different set of plants was used for every time point. Corresponding leaves from plants that did not receive any aboveground treatment (control) were selected and sampled as described above. Three biological replicates were selected among the five for RNA isolation. Total RNA was sent for sequencing to BGI Hong Kong.

Project description:The intricate interactions between plants and microorganisms have garnered substantial scientific interest. While previous studies have highlighted the potential influence of various fungal volatile compounds(VCs) on plant growth and development, the precise mechanisms underlying this modulation still need to be discovered. In this study, we discovered that fungal volatile organic compounds from the soil-borne fungus Tolypocladium inflatum GT22 enhance the growth of Arabidopsis. Remarkably, following the priming of Arabidopsis with GT22 VC, it displayed an enhanced immune response, thereby mitigating the detrimental effects caused by both pathogenic infections and copper stress. Transcriptomic analyses of Arabidopsis seedlings treated with GT22 VCs revealed the differential expression of 90, 83, and 137 genes after 3, 24, and 48 hours of volatile exposure, respectively. These responsive genes are involved in growth, hormone regulation, defense mechanisms, and signaling pathways. Notably, the induction of genes related to innate immunity, hypoxia, salicylic acid (SA) biosynthesis and camalexin biosynthesis by GT22 VCs were reported. Among the VCs emitted by GT22, limonene is particularly noteworthy. Arabidopsis seedlings exposed to limonene exhibited not only growth promotion effects but also alleviation of copper stress, indicating that limonene may play a role in the interaction between GT22 and plants. Overall, the findings of this study provide evidence supporting that fungal VCs can promote plant growth and enhance both biotic and abiotic tolerance. Furthermore, our results suggest that seedlings exposed to T. inflatum GT22 VCs holds promising potential for harnessing beneficial effects to improve crop productivity.

Project description:Transciptomic changes in tobacco (Nicotiana tabacum cv. Samsun) following exposure to microbial volatile organic compounds (MVOCs) emitted by Cladosporium sphaerospermum isolate TC09 (NRRL 67603) were investigated. Germinated seedling plants were cultured in Magenta GA7 vessels containing Murashige and Skoog (MS) medium supplemented with 30 g/L sucrose with and without fungal cultures that were included in the vessel but placed in a separated container. Ten days after the onset of MVOC exposure, tissue samples including the apical meristem and terminal leaves were collected and subject to RNA sequencing and transcriptome analysis using available tomato genome as reference. Our study revealed massive number of differentially expressed genes (DEGs) associated with diverse biological processes including cell expansion and cell cycle, photosynthesis, phytohormone homeostasis and defense responses as a consequence of short-term exposure to potent MVOCs.
