A week after final treatment CD4+CD25- Teff cells and CD4+CD25hi Treg cells were sorted by FACS. Genomic DNA isolated from sorted cells and differences in DNA methylation pattern in these cell types between PBS and OX40L-Jagged-1 treated mice was analyzed by whole genome bi-sulfite sequencing.

OX40L protects against inflammation-driven fibrosis

Project description:Fibrosis is a leading cause of deaths in industrialized countries and has no effective therapy. We demonstrated that blockade of OX40L prevented inflammation-driven fibrosis affecting the skin and the lungs and promotes regression of established dermal fibrosis in different murine models. To characterize the pathways involved in the protection of skin fibrosis and affected by OX40L blocking, we used microarrays and identified distinct genes differentially expressed between ox40l+/+ and ox40l-/- in the bleomycin-induced dermal fibrosis mouse model. Total RNA were extracted from lesional skin samples of 3 ox40l+/+ and 4 ox40l-/- male mice aged 9 weeks treated with bleomycin for 3 weeks, and were hybridized on Affymetrix microarrays.

Single cell RNA sequencing of femur bone marrow cells from the NZBWF1 systemic lupus erythematosus mouse model and its progenitors.

Project description:Systemic lupus erythematosus (SLE) is an autoimmune disorder characterized by failure of self-tolerance mechanisms and the resultant production of autoreactive antibodies. The NZBWF1 mouse spontaneously develops a lupus-like syndrome and is used as model of SLE. The NZBWF1 model represents the F1 generation of a cross between New Zealand Black (NZB) and New Zealand White (NZW) mice. In this study we investigated the model and its progenitors (NZB, NZW) gene expression with single cell RNA sequencing on cells isolated from bone marrow and processed with the 10X Chromium.

Exosomal OX40L elicits positive T cell co-stimulation and promotes anti-tumor immunity through engagement of cytotoxic T lymphocytes

Project description:Exosomes play a role in immune regulation associated with cancer and other diseases. We generated exosomes that harbor the ligand of the co-stimulatory OX40 pathway, and demonstrated abundant levels of OX40L on the surface of exosomes, without inducing morphological and molecular alterations. Functionally, exosomal OX40L efficiently elicits positive co-stimulation of both human and murine T cells. The systemic administration of exosomal OX40L promotes anti-tumor immunity in an orthotopic melanoma mouse model, and the depletion of T cell subsets revealed that exosomal OX40L acts through CD8+ T cells. Mechanistically, exosomal OX40L enhances the proportion and the levels of cytotoxic markers in tumor-infiltrating CD8+ T lymphocytes. Taken together, using the OX40 pathway as proxy, we provide proof-of-principle evidence that the presence of positive co-stimulatory molecules on exosomes can functionally engage T cells to elicit anti-tumor immunity, paving the way to understand the biology of exosome-mediate T cell signaling and for further developing exosome-based reagents that engage T cell co-stimulation.
