Time-course study of microarray transcriptome analysis in influenza virus-infected MDCK and TR7 cells.

ABSTRACT: Comparative transcriptome analysis indicated that expression of apoptosis-related MX1 and MMP9 was suppressed in TR7 cells both before and during viral infection. Meanwhile, CDKN2A was upregulated in TR7 cells, and CCND3 and CASP7 were upregulated in MDCK cells, which supported that cell cycle arrest in TR7cells benefited influenza virus production.

GFP-negative normal MDCK cells were then collected by FACS, and expression of various genes was compared between the two conditions.

Transcriptome analysis in MDCK cells on anti-influenza A activity of bakuchiol

Project description:Bakuchiol is a phenolic isoprenoid compound present in Babchi (Psoralea corylifolia Linn.) seeds. We examined the anti-influenza activity of synthetic bakuchiol using Madin-Darby canine kidney (MDCK) cells. We found that the naturally occurring form, (+)-(S)-bakuchiol, and its enantiomer, (-)-(R)-bakuchiol, inhibited influenza A infection and growth, and reduced the expression of viral mRNAs and proteins in MDCK cells. However, the molecular mechanism in anti-influenza activity of bakuchiol remains unclear. Thus, to evaluate its molecular mechanism in host cells, we performed the next generation sequencing analysis of the MDCK transcriptome in cells treated with bakuchiol and influenza A/PR/8/34. These results provide keys to elucidate the molecular mechnism of anti-influenza activity of bakuchiol for us, and these data will contribute to the development of novel approaches to anti-influenza treatments.

Transcriptome analysis of pathways and drug transporters after hyperosmotic stress of renal MDCK I cells

Project description:Purpose: To perform pathway analysis of MDCK I treated with hyperosmolality and to investigate how hyperosmolality affects the expression of membrane transporters. Method: MDCK I cells were treated for 24 h with medium supplemented with 200 mOsm raffinose, NaCl or urea (total of 500 mOsm). The control was regular medium (300 mOsm). RNA was isolated from 12 replicates from each condition and a cDNA library was prepared using TruSeq RNA Library Preparation Kit v2 from Illumina following the manufacturer's protocol. The sequencing was done with a HiSeq with single read and a length of 51. The alignment was done with Hisat 2, the count with FeatureCount and the statistical analysis with DESeq2. Results: The pathways the cell cycle, the cytokine-cytokine receptor interaction pathway, and the chemokine-signaling pathway were significantly activated in MDCK I cells after treatment of 200 mOsm raffinose or NaCl (total: 500 mOsm), and not 200 mOsm urea, for 24 hours. Several membrane transporters were regulated by hyperosmolality. Conclusion: Hyperosmolality has an inflammatory-like response in MDCK I cells and it also affects the expression of several membrane transporters.

Nfat5 is involved in the hyperosmotic regulation of Tmem184b, which could be involved in ibuprofen transport in renal MDCK I cells

Project description:Purpose: To investigate the involvement of Nfat5 in the regulation of an ibuprofen transporter Method: Reverse transfection with siRNA against Nfat5 was used to knockdown Nfat5 in MDCK I cells. The uptake of both radiolabelled taurine and ibuprofen was measured in MDCK I cells, first treated with siRNA against Nfat5 for 24 h and afterwards cultivated with raffinose-supplemented normal growth medium (500 mOsm) for 24 h. The knock down of NFat5 was investigated by qpCR and western blotting. A transcriptome analysis of MDCK I cells treated with siRNA against Nfat5 was performed. Validation of the transcriptome analysis was perform with qPCR and radiolabelled uptake of ibuprofen. Results: The siRNA transfection resulted in a knock down of Nfat5, and the uptake of both taurine and ibuprofen was significant decreased in the transfected MDCK I cells. The transcriptome analysis of MDCK I cells treated with siRNA against Nfat5 revealed genes regulated by Nfat5 during hyperosmotic exposure. qPCR and the transciptome analysis showed an involvement of Nfat5 in the hyperosmotic regulation of Tmem184 and uptake in Tmem184b-siRNA treated MDCK I cells showed that Tmem184b might be involved in uptake of ibuprofen. Conclusion: The regulation of Tmem184b was found regulated by Nfat5 and could be a potential possible candidate for the gene product responsible for the uptake of ibuprofen in MDCK I cells
