Transcriptional profiles of liver in control and p21-AAV8 administered non-pregnant mice and pregnant mice at dpc8 and dpc16.

ABSTRACT: We report transcriptional profiles of liver from control and p21-AAV8 administered no-pregnant mice and pregnant mice at dpc8 and dpc16. We revealed that genes related to carbohydrate transport and regulators of glucose and glycogen metabolism were down regulated in AAV-p21 administered liver at dpc16.

Project description:Transcriptional profiling of microglia (seven biological replicates per experimental condition) or non-microglia (four biological replicates total). One thousand microglia (CD11b+ Ly6G/Ly6C-) or non-microglia cells (CD11b-) from P30 rd1 (FVB) retinas infected with either AAV8-GFP or AAV8-GFP + AAV8-TGFB1 were sorted into 10 µL of Buffer TCL (Qiagen) containing 1% beta-mercaptoethanol and immediately frozen on dry ice. Samples were subsequently sent to the Broad Institute Genomics Platform for cDNA library synthesis and sequencing using a modified Smart-Seq2 protocol with an expected coverage of ~6 million reads per sample. Prior to analysis, reads were subjected to quality control measures and mapped to the GRCm38.p6 reference genome. Reads assigned to each gene were then quantified using featureCounts and normalized and analyzed for differential expression using DESeq2.

Project description:PolyA-selected nuclear RNA isolated from livers of adult male mice administered AAV8-STAT5ca was analyzed by RNA-seq to determine the impact of constitutive STAT5 activation on the expression of sex-biased genes in male mouse liver. This is part of a study entitled 'Constitutively active STAT5b feminizes mouse liver gene expression', where delivery of constitutively active STAT5 (STAT5CA) to male mouse liver was shown to induce widespread feminization of the liver, with extensive induction of female-biased genes and repression of male-biased genes, largely mimicking results obtained when male mice are given GH as a continuous infusion. Many of the STAT5CA-responding genes were associated with nearby (< 50 kb) sites of STAT5 binding to liver chromatin, supporting the proposed direct role of persistently active STAT5 in continuous GH-induced liver feminization. The feminizing effects of STAT5CA were dose-dependent; moreover, at higher levels, STAT5CA overexpression resulted in some histopathology, including hepatocyte hyperplasia, and increased karyomegaly and multinuclear hepatocytes. Together, these findings establish that the persistent activation of STAT5 by GH that characterizes female liver is by itself sufficient to account for the sex-dependent expression of a majority of hepatic sex-biased genes.

Project description:Influence of ovarian stimulation with 200 IU of hCG, (administered in the late follicular phase among ICSI patients undergoing a GnRH-antagonist protocol), on the endometrium on the day of oocyte pick-up. The purpose of the present study is to assess the influence of the administration of low dose hCG on the endometrium. In addition, by analysing the correlation of the morphological pattern and gene expression profile of human endometrium on the day of oocyte retrieval in patients of both treatment groups, we want to study the implantation potential. Pregnant and non-pregnant patients were compared. In total 6 samples were analyzed for gene expression analysis with microarrays, 3 pregnant and 3 non-pregnant patients were compared.

Transcriptional profiles of liver lobular zones from non-pregnant mice and pregnant mice at dpc8 and dpc16.

Project description:We report transcriptional profiles of each liver lobular zone from non-pregnant mice and pregnant mice at dpc8 and dpc16. By RNA-sequencing of LCM-isolated each zonal cells, we revealed that genes related to actin cytoskeleton, extracellular matrix organization and vascularization were enriched in the PA (zone2) and CV (zone3) zones at dpc8 and cell division related genes were enriched in the PV (zone1) zone at dpc8. At dpc16, genes related to metabolism, catabolism and xenobiotic process were enriched in all zones.
