ScRNA-seq analysis of Msi2CreER-MycT58A Mouse Pre-cancer and Pancreas ASCP tumor

ABSTRACT: scRNA-seq analysis evaluating transcriptional heterogeneity in mouse pre-cancer and pancreas tumors in an effort to model the progression of pancreas tumors. Single cells were captured at t=0 (before tamoxifen induction), t=5 (5 weeks after tamoxifen induction) and t=12 (12 weeks after tamoxifen induction).

Project description:Identifying the cells from which cancers arise, and the paths they take towards malignancy is critical for understanding the molecular basis of tumor initiation and progression. To determine whether stem and progenitor cells can serve as cells of origin for cancer, we created a new Msi2-CreERT2 knock-in strain. When crossed to a conditional CAG-LSL-MycT58A model, Msi2-CreERT2 mice developed multiple pancreatic cancer subtypes: pancreatic ductal adenocarcinoma (PDAC), adenosquamous carcinoma of the pancreas (ASCP), acinar cell carcinoma (ACC), and rare anaplastic tumors. Combining single-cell genomics with computational analysis of developmental states and lineage trajectories, we demonstrate that oncogenic MYC preferentially triggers the transformation of the most immature subset of Msi2+ cells, leading to the rise of a common pool of pre-cancer cells with multi-lineage properties. These pre-cancer cells subsequently diverge to distinct pancreatic cancer subtypes by activation of distinct transcriptional programs and large-scale genomic changes. Importantly, the enforced expression of specific molecular signals can redirect cells toward specific subtypes. This study shows that multiple pancreatic cancer subtypes can arise from a common pool of Msi2+ cells and provides a powerful framework to understand and control the programs that shape divergent fates in pancreatic cancer.

RNAseq of cell line from pancreas from KRasG12D/RosaMycER mouse treated with Tamoxifen

Project description:Experiment designed to study the effect of deregulated myc on pancreatic cancer progression and regression. In the KRasG12D/RosaMycER mouse model KRas and MycER are expressed exclusively in the pancreas, but MycER activity depends on Tamoxifen presence. For this experiment a mouse was treated with Tamoxifen during 3 weeks, the pancreas was collected and the epithelial cells were grown in vitro in the presence of 4-OHT (4-Hydroxytamoxifen, active form of Tamoxifen). RNAseq were preformed on the cell line treated as follows: 1- 4-OHT all the time. 2- EtOH 6h. 3- EtOH 12h. 4- EtOH 18h.

Identification of Sox9-Regulated Pathways During the Secondary Transition Stage of Pancreas Development

Project description:Sox9 target genes were identified during the secondary transition stage of pancreas development by comparing gene expression in Sox9-ablated versus wild-type pancreata using microarray analysis. Sox9 was conditionally ablated during the secondary transition in the developing pancreas via recombination of a Sox9-flox allele (Kist et al., 2002) using the tamoxifen-inducible Rosa26-CreER allele (Vooijs et al., 2001). Dams were injected with 6 mg/40 g tamoxifen in corn oil at e12.5. Pancreata were manually microdissected at e15.5. Total RNA was isolated and pooled from pancreata of e15.5 Sox9fl/fl; Rosa26-CreER (mutant) versus Rosa26-CreER (wild-type) littermates for four biological replicates.
