Global gene expression profiling of M. tuberculosis H37Rv and the isogenic regX3-inactivated mutant (DregX3) grown in phosphate enriched or phosphate-limited medium

ABSTRACT: In order to understand the repertoire of genes regulated by RegX3, we performed microarray analysis of gene expression in M. tuberculosis (WT) or DregX3 grown in phosphate-enriched or phosphate limited medium. The differentially regulated genes provide insight into the role of RegX3 in gene regulation in Mtb.

Genome-wide association study of RegX3 in M. tuberculosis (Mtb) grown under phosphate limited conditions

Project description:In order to understand the role of the transcriptional regulator RegX3 in regulating gene expression under phosphate limitation, we performed chromatin immunoprecipitation with RegX3 antibody followed by next generation sequencing using the immunoprecipitate. We observed RegX3 binding at a number of loci including genes which were observed to be differentially regulated during growth in phosphate limited vs. phosphate enriched media, suggesting a global role of RegX3 in regulating gene expression under phosphate starvation.

Project description:We used the previously designed oligonucleotide microarrays (BM-CM-<rgmann et al., 2007, Environmental Microbiology, 9: 2742-2755) to detect the mRNA transcripts of R. pomeroyi DSS-3 when the cells were cultured under steady-state conditions limited with ammonium (NH4Cl, 0.26 mM) but with an excess of D-ribose-5-phosphate (C5H9Na2O8P*2H2O, 0.5 mM), methylphosphonic acid (CH5PO3, 0.5 mM), or potassium phosphate (KH2PO4, 0.5 mM), or during ammonium excess (NH4Cl, 2.8 mM) but were limited with potassium phosphate (KH2PO4, 9.2 M-NM-<M). Data for the technical replicates were averaged and combined, resulted in a total of 12 samples.

Time course of Saccharomyces cerevisiae Exposed to Arsenic under Phosphate Limited Conditions

Project description:Arsenic is known as a human carcinogen that easily be exposed by the living organisms through environment and food consumption. The arsenic is transport into the cells via phosphate transporters due to its structural similarity with phosphate in both prokaryotes and eukaryotes. We here evaluated and analyzed the toxicogenomic impacts of arsenate and the role of different phosphate concentrations on arsenic toxicity. Our results showed that arsenic uncoupled phosphate levels which eventually affected the growth rate of yeast cells. Analysis of arsenate levels in the medium over 4 to 10 h of its exposure clearly showed that arsenate was easily taken up by the cells in phosphate limited condition.
