Gene expression profiling in melanised sites of Atlantic salmon fillets

ABSTRACT: Black spots, which deteriorate quality of Atlantic salmon fillets represent a significant problem for commercial aquaculture. These areas are characterized with accumulation of melanomacrophages, occasional formation of granulomas and substitution of skeletal muscle with connective tissue. Transcriptome analysis was performed to elucidate the origin and the character of inflammation in the melanized foci of skeletal muscle.

The farmed Atlantic salmon (Salmo salar) skin-mucus proteome

Project description:Norway is the largest producer and exporter of farmed Atlantic salmon (Salmo salar) worldwide. Skin disorders correlated with bacterial infections represent an important challenge for fish farmers due to the economic losses caused. Little is known about this topic, thus studying the skin-mucus of Salmo salar and its bacterial community depict a step forward in understanding fish welfare in aquaculture. In this study, we used label free quantitative mass spectrometry to investigate the skin-mucus proteins associated with both Atlantic salmon and bacteria. In addition, the microbial temporal proteome dynamics during 9 days of mucus incubation with sterilized seawater was investigated, in order to evaluate their capacity to utilize mucus components for growth in this environment.

Gill and liver transcript expression changes associated with gill damage in Atlantic salmon (Salmo salar)

Project description:Gills of teleost fish represent a vital multifunctional organ; however, they are subjected to environmental stressors, causing gill damage. Gill damage is associated with significant losses in the Atlantic salmon aquaculture industry. Gill disorders due to environmental stressors are exacerbated by global environmental changes, especially with open-net pen aquaculture (as farmed fish lack the ability to escape those events). The local and systemic response to gill damage, concurrent with several environmental insults, are not well investigated. We performed field sampling to collect gill and liver tissue after several environmental insults. Using a 44K salmonid microarray platform, we aimed to compare the transcriptomes of pristine and moderately damaged gill tissue. The gill damage-associated biomarker genes and associated qPCR assays arising from this study will be valuable in future research aimed at developing therapeutic diets to improve farmed salmon gill health.

Susceptibilities of Atlantic salmon to Piscirickettsia salmonis

Project description:The aquaculture industry has confronted severe economic losses due to infectious diseases in the last years. Piscirickettsiosis or Salmonid Rickettsial Septicaemia (SRS) is the bacterial disease caused by Piscirickettsia salmonis. This Gram-negative, non-motile, cellular pathogen has the ability to infect, survive, replicate, and propagate in salmonid monocytes/macrophages generating a systemic infection characterized by the colonization of several organs including kidney, liver, spleen, intestine, brain, ovary and gills. In this study, we attempted to determine whether global gene expression differences can be detected in different genetic groups of Atlantic salmon as a result of Piscirickettsia salmonis infection. Moreover, we sought to characterize the fish transcriptional response in order to reveal the mechanisms that might confer resistance in Atlantic salmon to an infection with Piscirickettsia salmonis. In doing so, after challenging with Piscirickettsia salmonis, we selected the families with the highest (HS) and the lowest (LS) recorded susceptibility for gene expression analysis using 32K cGRASP microarrays. Our results revealed in LS families expression changes are linked to iron depletion, as well as, low contents of iron in kidney cells and low bacterial load, indicated that the iron-withholding strategy of innate immunity is part of the mechanism of resistance against Piscirickettsia salmonis. This information contributes to elucidate the underlying mechanisms of resistance to Piscirickettsia salmonis infection in Atlantic salmon and to identify new candidate genes for selective breeding programmes.

Nutrigenomic effects of glucosinolates on liver, muscle and distal kidney of Atlantic salmon

Project description:Glucosinolates (GLs) present in plants from the Brassica family hold promise in the biological control of the skin parasite salmon louse (Lepeophtheirus salmonis) in farmed Atlantic salmon. We recently reported significantly reduced lice infestation in fish fed GLs. However, their wider application requires better knowledge of GLs’ actions, including positive and adverse effects that may occur under the overexposure. Microarray analyses performed in the liver, muscle and distal kidney of salmon under high dose of GLs suggested massive tissue remodeling and reduction of cellular proliferation in skeletal muscle and liver. In the distal kidney, gene expression profiles induced under the high dose of GLs pointed to activation of anti-fibrotic responses. At the same time, prevalent activation of genes from the Phase-2 detoxification pathways could be considered part of beneficial effects. Multiple gene expression evidence suggested GLs-mediated iron/heme withdrawal response, including increase in heme degradation in muscle (up-regulation of heme oxygenase-1), decrease of its synthesis in liver (down-regulation of porphobilinogen deaminase) and increase in iron sequestration from blood (hepatic induction of hepcidin-1 and renal induction of intracellular storage protein ferritin). This could be beneficial upon encounter with the parasite, which depends on the provision of iron/heme by the host.
