Identification differentially expressed genes in the mouse testis caused by Prss37 deficiency

ABSTRACT: Testes from three wt and three Prss37-/- mice were used for gene expression analysis using whole mouse genome 4 × 44 k microarray (Agilent Technologies). A total of 31 genes were identified to be differentially expressed significantly (p < 0.05, fc ≥ 2). Fold changes of ten genes (seven up-regulated genes and three down-regulated genes) were examined by qRT-PCR for data validation.

Project description:The aim of the study was to elucidate the role of Alkbh1 by targeted deletion in C57/BL6 mice. Alkb deficiency results in sex-ratio distortion of offspring and apoptosis in adult testes, most likely caused by defects in the pachytene stage during spermatogenesis. Due to the pivotal role of Alkbh1 in mouse survival and potentially in germ cells, we searched for Alkbh1-regulated genes in adult testes.

Comparative analysis of testis transcriptome between a genetic male sterile line (GMS) and its wild-type 898WB in silkworm, Bombyx mori

Project description:Purpose:The purpose of study these processes, is helpful to understand the mechanisms of genetic sterility of lepidopteran insects and achieve sterile insect technique (SIT) for pest control. Methods:Intact testes were dissected on day 5 of fifth-instar larvae and immediately frozen in liquid nitrogen for RNA-Seq. The testes from three silkworms were taken as one biological sample, and three samples per strain were prepared as biological replicates. Results: A total of 1,872 up-regulated genes and 1,823 down-regulated genes were detected in the testis of the GMS mutant. Several genes play important roles in spermatogenesis and testicular development, such as “serine/threonine protein kinase”, “organic cation transporter protein”, “tyrosine protein kinase”,“lncRNAs” and “immune-associated genes”, and so on. GO (Gene ontology) annotation and KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway analysis was performed for functional annotation of the differentially expressed genes (DEGs). KEGG pathway analysis showed that the DEGs annotated to 123 pathways, and 10 pathways were enriched significantly, such as “metabolic pathway”, “biosynthesis of amino acids”, and “phagosome–lysosome pathway” associated with the testis development and spermatogenesis. The qPCR expression results were similar to the results obtained from the RNA-seq data, which showed the sequencing results were reliable. Conclusions:Differentially expressed genes of the testes between GMS mutant and 898WB were screened by RNA-Seq technology, which provide reliable reference for understanding the mechanism of male sterility in GMS mutant.

Project description:Transcriptional profiling of Drosophila melanogaster larval testes with and without the wMel strain of Wolbachia and found that 296 genes had at least a 1.5 fold change [q-value (%)<5%] in transcript levels, with 167 genes up-regulated and 129 genes down-regulated when comparing Wolbachia-infected flies to uninfected ones. Differential expression of genes related to metabolism, immunity, reproduction and other functions were observed. Two-condition experiment, Wolbachia-infected vs. Wolbachia-uninfected testes. Biological replicates: 3 control, 3 infected, independently grown and dissected. One replicate per array.

Comparative transcriptomic profiling between two radiologic phenotypes in lungs from patients with pulmonary disease caused by Mycobacterium avium complex

Project description:The incidence of Mycobacterium avium complex (MAC)-induced pulmonary disease (PD) has been increasing in South Korea, posing considerable challenges in treatment. Treatment guidelines recommend initiating therapy after serial computed tomography (CT) monitoring. While drug treatment often improves outcomes in patients with the nodular bronchiectatic (NB) form, those with the fibrocavitary (FC) form may continue to experience persistence despite treatment. Our objective was to classify type-specific and common genes based on radiological phenotypes (NB and FC) using RNA sequencing to analyze the cellular landscape of 11 NB sections, 10 FC sections, and 21 unaffected sections from 21 MAC-PD cases. Predictive in silico analysis indicated a significant role for genes related to B cell proliferation and regulation in both forms. Additionally, NB patients exhibited predominantly regulated antimicrobial immune responses, whereas FC patients showed enrichment in genes related to extracellular matrix structure. These target genes identified through genetic analysis could potentially serve as predictive biomarkers for type-specific biological processes and cellular pathways, necessitating further validation studies.

Transcription profiling of testis biopsies to investigate testis developmental gene expression in cryptorchid boys at risk of azoospermia

Project description:Despite timely and successful surgery, 32% of patients with bilateral and 10% with unilateral cryptorchidism will develop azoospermia. Cryptorchid boys at risk of azoospermia display a typical testicular histology of impaired mini-puberty at the time of the orchidopexy. During mini-puberty increased gonadotropin and testosterone secretion stimulate transformation of gonocytes into Ad spermatogonia. In azoospermia risk group this transformation is to a great extent impaired. This study aimed to analyze data on whole genome expression signatures of undescended testes at risk of developing azoospermia. Twenty-three testicular biopsies from 22 boys were analyzed (19 testes from 18 boys with cryptorchidism) and 4 contralateral descended testes from patients with testicular agenesis. Expression profiling identified 483 genes not or under-expressed in the azoospermia risk group compared with the control and LAZR groups. Annotated loci were associated with spermatogenesis. Other significant genes were cellular defense response genes and hormone controlled loci involved in spermatogenesis. Some genes transcribed in normal adult meiotic and post-meiotic germ cells are activated in healthy juvenile Ad spermatogonia. Thus, molecular events initiating the testicular expression program at the onset of puberty and maintaining it during adulthood occur very early in prepubertal testes. This molecular event is to a great extent impaired in HAZR group lacking Ad spermatogonia (stem cells for spermatozoa) indicating impaired mini puberty.
