Regulation of adipogenesis via caveolin-1 in adipose derived mesenchymal stem cells

ABSTRACT: Using human adipose derived mesenchymal stem cells (ASCs) as a model system, we identified caveolae forming structural protein, caveolin-1 (CAV1) as a key component that limits adipogenic differentiation of ASCs. Using whole transcriptomic analysis by RNA sequencing, we show that lentiviral over expression of human CAV1 gene in ASCs significantly inhibits adipogenesis as evident by inhibition of adipogenic gene network, and conversely, RNAi driven knockdown of endogenous CAV1 significantly promoted adipogenesis.
