Effect of Meflin overexpression on the fibroblast transcriptome in a three-dimensional culture using cell accumulation technique

Project description:We previously reported that Meflin, encoded by the immunoglobulin superfamily containing leucine-rich repeat (ISLR) gene, is a marker for tumor-restraining cancer-associated fibroblasts. However, the functional roles of Meflin in fibroblasts remain unclear. To investigate the functional impact of Meflin expression in fibroblasts, RNA sequencing was performed on Meflin-overexpressing NHDF-Ad cells cultured under conditions that better mimic the in vivo tumor microenvironment, using a cell accumulation technique that accurately recapitulates the fibrotic stroma observed in aggressive human cancers. Meflin overexpression resulted in the downregulation of PDPN, a well-established marker of tumor-promoting CAFs, and upregulation of DPT and COL15A1, which are markers of universal fibroblasts that resemble normal fibroblasts and are considered a potential source of diverse fibroblast subtypes.These findings suggest that high-level expression of Meflin is crucial for skewing CAFs toward a more universal fibroblast–like state.

BMP9 Secretome of hHiPC LC-MS/MS

Project description:To investigate BMP9/ALK1 signaling in human highly proliferative cells (hHiPC), we selected one primary population of hHiPC from 3 individuals and studied the secretome responses to BMP9 treatment. Proteins were collected in hHiPC treated conditioned media (CM) for mass spectrometry analysis. Proteomic analysis of the hHiPC secretome following BMP9 treatment demonstrates that the secreted proteins, sclerostin (SOST), meflin/immunoglobulin superfamily containing leucine rich repeat (ISLR), and insulin-like growth factor binding protein-3 (IGFBP3), are novel downstream targets of BMP9/ALK1-mediated signaling. Consistent with this hypothesis, lentiviral shRNA and pharmacological inhibition of ALK1 in hHiPCs suppressed secretion of SOST, ISLR, and IGFBP3 following BMP9 treatment as measured by secretome LC-MS/MS.

Proteome profiling of breast cancer biopsies reveals a wound healing signature of cancer-associated fibroblasts - primary mammary fibroblasts treated with TGF-beta

Project description:Breast cancer is still the most common type of cancer in women; an important role in carcinogenesis is actually attributed to cancer-associated fibroblasts. In this study we investigated whether it is possible to assess the functional state of cancer-associated fibroblasts through tumor tissue proteome profiling. Tissue proteomics was performed on tumor-central, tumor-near and tumor-distant biopsy sections from breast adenocarcinoma patients, which allowed us to identify 2074 proteins. Data were interpreted referring to reference proteome profiles generated from primary human mammary fibroblasts comprising 4095 proteins. These cells were analyzed in quiescent cell state, as well as after in vitro treatment with TGFβ or IL-1β, stimulating wound healing or inflammatory processes, respectively. Representative for cancer cells, we investigated the mammary carcinoma cell line ZR-75-1, identifying 5212 proteins. Comparison of tissue proteomics data with all those reference profiles revealed predominance of cancer cell-derived proteins within the tumor and fibroblast-derived proteins in the tumor-distant tissue sections. Remarkably, proteins characteristic for acute inflammation were hardly identified in the tissue samples. In contrast, several proteins found by us to be induced by TGFβ in mammary fibroblasts, including fibulin-5, SLC2A1 and MUC18, were positively identified in all tissue samples, with relatively higher abundance in tumor neighboring tissue sections. These findings indicate a predominance of cancer-associated fibroblasts with wound healing activities localized around tumors.

Project description:Pulmonary fibrosis (PF) is both an independent disease and a pathologic basis for fibroproliferative lung diseases. Understanding of underlying mechanisms of PF is critical for developing effective therapeutics. Fibroblasts activation and extracellular matrix deposition are critical in pathogenesis of PF. Stimulation of certain factors can induce profibrotic changes in fibroblasts. Evaluation of associated genes in treated fibroblasts is helpfu to analyze the profibrotic changes of this critical effector cell for PF. We used microarrays to detail the global programme of gene expression following TGFβ and CCL1 stimulation of primary lung fibroblasts.
