Gene expression in human keratinocytes cultivated with serum-free medium and serum containing medium

ABSTRACT: The objective of this study was thus to develop ab initio that chemically defined serum-free medium. We used a statistical design of experiment (DOE) approach to screen a large number of candidate factors, for both their potential individual and synergetic effects, as to identify a formulation which was then tested extensively. In this study, we present the properties of keratinocyte cultivated with serum-Free medium (Surge SFM) and complemented DME-Ham medium (with serum).

Characterization of human dental pulp cells grown in chemically defined serum-free medium

Project description:Dental pulp cells obtained from several donors proliferated actively in a serum-free medium STK2. The growth rate of dental pulp cells from most donors was higher in the serum-free medium than that in a medium containing 10% serum. DNA microarray analyses showed that gene expression profile of dental pulp cells grown in the serum-free medium was similar to that of cells grown in a medium containing 10% serum. However, several genes related to cell proliferation were up-regulated in dental pulp cells grown in the serum-free medium.

Enrichment and quantitation of newly secreted proteins under serum condition by combining BONCAT and pulsed SILAC

Project description:Despite the increasing interest of secretome involved in paracrine/autocrine mechanisms, the majority of the mass spectrometric cell secretome studies have been performed using serum-free medium (SFM), due to low-abundance of secreted proteins and high-abundance of contaminating fetal bovine serum (FBS) proteins in serum-containing medium (SCM). In this study, through the combination of bioorthogonal non-canonical amino acid tagging (BONCAT) and pulsed-SILAC (pSILAC), the low-abundant secreted proteins were captured selectively and quantified. We set up the combined BONCAT-pSILAC method by optimizing capturing condition and by using a composite human-FBS database, and applied it to the analysis of differentially secreted proteins between SFM and SCM. The amount of secreted proteins for 24 h in SCM was much bigger than that from SFM for U-87MG glioblastoma cells (1.28-fold) and mesenchymal stem cells from human umbilical cord blood (hUCB-MSCs; 2.01-fold). The proteins secreted more in SCM were predominantly predicted to be truly secretory. Therefore, this study suggests the analysis of the secretome should be processed in serum-containing medium that promotes cell proliferation and secretion.

Project description:Analysis of gene expression profile changes of immortalized human meibomian gland epithelial cells under differentiation. Results provide important information of the response of human meibomian gland epithelial cells to different differentiation media, such as serum containing medium, serum-free medium, serum-free medium with 1.2mM Ca2+. Primary cultured human meibomian gland epithelial cells and hTERT immortalized human meibomian gland epithelial cells (3 cells/condition/experiment) were grown in keratinocyte serum-free medium (SFM, Invitrogen-Gibco, Grand Island, NY) on 75cm2 to reach confluence, then change to SFM, SFM with 1.2mM Calcium, or serum containing medium (10% fetal bovine serum in an equal volume of DMEM and Ham’s F12 with 10ng/ml EGF) for 14 days. Total RNA was extracted from the above cultures. Gene expression was analyzed using Illumina HumanHT-12 v3 Expression Beadchips (San Diego, CA).

Epigenome and transcriptome of naive pluripotent mouse embryonic stem (ES) cells cultured in 2i serum-free medium

Project description:This study describes the epigenetic and transcriptome profiling of mouse ES cells cultures in 2i serum-free medium. The proteins for which ChIP-Seq profiles were generated in these cells, as well as for cells grown in serum conditions, include H3K4me3, H3K36me3, H3K27me3, H3K9me3, Suz12 and Ezh2. A sequence profile of genomic DNA is also included. Furthermore, we included RNA-Seq of ds_cDNA synthesized from double poly(A) selected mRNA and from rRNA depleted (strand specific) total RNA.

Corynebacterium diphtheriae proteome adaptation to cell culture medium and serum

Project description:ost-pathogen interactions are often studied in vitro using primary or immortal cell lines. This set-up avoids ethical problems of animal testing and has the additional advantage of lower costs. However, the influence of cell culture media on bacterial growth and metabolism is not considered or investigated in most cases. To address this question growth and proteome adaptation of Corynebacterium diphtheriae strain ISS3319 were investigated in this study. Bacteria were cultured in standard growth medium, cell culture medium and fetal calf serum. Mass spectrometric analyses and label-free protein quantification hint to an increased bacterial pathogenicity when grown in cell culture medium as well as an influence of the growth medium on the cell envelope.
