RNA-seq of limmmunocompetent mice in response to Aspergillus fumigatus conidia and hyphae

ABSTRACT: We bulit a pulmonary aspergillosis model in immunocompetent mice. Mice were inoculated intratracheally with 50 μL suspension of conidia , hyphal fragments or normal saline, respectively, after anesthesia. Lungs from were harvested and ground for RNA extraction 24 hours after fungal inoculation. The result of RNA-seq indicated both conidia and hyphae can activate a series of immunologically relevant pathways and these pathways were much more strongly activated by hyphae. Hyphae could promote Th2 cell differentiation and suppress Th1 cell differentiation. Both hyphae and conidia could activate Th17 cell differentiation.

Project description:Hyphae and conidia, representing the two developmental stages of the fungus, were examined separately in this study. RNA from hyphae or conidia incubated in plasma in RPMI/HEPES without neutrophils was used as reference, and was co-hybridized with the query samples obtained from the corresponding sets incubated with neutrophils. Each biological replicate was carried out with neutrophils obtained from a different donor, and the RNA samples associated with each neutrophil source was paired separately with a corresponding reference sample prepared at the same time.

Ewald2021 - Innate immune response during invasive aspergillosis by dynamic optimization

Project description:Aspergillus fumigatus is an important human fungal pathogen and its conidia are constantly inhaled by humans. In immunocompromised individuals, conidia can grow out as hyphae that damage lung epithelium. The resulting invasive aspergillosis is associated with devastating mortality rates. Since infection is a race between the innate immune system and the outgrowth of A. fumigatus conidia, we use dynamic optimization to obtain insight into the recruitment and depletion of alveolar macrophages and neutrophils. We illustrate by modeling the active, but so far neglected, major role of alveolar epithelial cells in phagocytosis and cytokine release as well as the importance of fungal growth states for virulence. Hence, we discovered that germination speed is a key virulence trait of fungal pathogens due to the vulnerability of conidia against host defense. We proved this by linking measured germination kinetics of four Aspergillus spp. with their cytotoxicity against epithelial cells in silico and in vitro.Furthermore, we could reveal by modeling and ex vivo measurements, that epithelial cells are not only important phagocytes to clear conidia, but also potent mediators of cytokine release. In conclusion, our findings illustrate an underestimated role of epithelial cells in invasive aspergillosis. Further, our model affirms the importance of neutrophils and underlines that the role of macrophages in invasive aspergillosis remains elusive. We expect that our model will contribute to improvement of treatment protocols by focusing on the critical components of immune response to fungi but also fungal virulence.

Project description:The response of AM transcription to exposure to conidia is expected to provide information about the mechanism by which these cells prevent conidial germination and therefore invasive aspergillosis. Experiment Overall Design: Conidia from A. fumigatus or polystyrene beads were instilled into lungs of C57Bl/6 or gp91phox-/- mice to test the response of AM in a normal and immune compromised host to determine if different responses contributed to increased susceptibility of invasive aspergillosis. Following in vivo incubation for 0, 2, or 4 hours, AM RNA was collected and prepared for hybridization to mouse Affymetrix GeneChip arrays.

Project description:Four different developmental time points representing the Afu development from dormant conidia into young hyphae (0h, 4h, 6h, 8h) were chosen. Cell walls of the Afu samples were extracted using denaturing methods followed by isolation of solubilized proteins. Samples were processed using filter aided sample preparation (FASP) / trypsin digestion. The peptides were analyzed by one-dimensional LC (capillary C18 nano-Eflow mode) on-line with a nano-ESI source and tandem MS (LTQ ion trap).

RNA-Seq analysis of WT and BbWor1 disruption strain in beauveria bassiana

Project description:Wor1 is a conserved fungal regulatory protein that control the phenotypic switching and pathogenicity of Candida albicans.We simulated three models of Beauveria bassiana morphological transitions, including CTH (conidia to hyphae), HTC (hyphae to conidia) and BTB (blastospore to blastospore).We find that disruption of BbWor1 (an ortholog of Wor1) resulted in a distinct reduction in the time required for germination (CTH), a significant increase in hyphal growth and a decrease in the yield of conidia (HTC), indicating that BbWor1 positively controls conidia production and negatively regulates hyphal growth in conidia-hyphae switching. Meanwhile, the ΔBbWor1 prominently decreased blastospore yield, as well as shortening G0/G1 phase and prolonging G2/M phase, under the BTB model. Importantly, BbWor1 contributed to conidia-hyphae switching and blastospore propagation via different genetic pathways, and yeast one-hybrid testing demonstrated the necessity of BbWor1 to control the transcription of an allergen-like protein gene (BBA_02580), as well as a conidial wall protein gene (BBA_09998). Moreover, the dramatically weakened virulence of ΔBbWor1 was examined by immersion and injection methods.

Maturation of conidia on conidiophores of Aspergillus niger

Project description:An Aspergillus niger colony can produce a few billion conidia. Formation of conidia producing conidiophores starts in the centre of the colony and moves outwards ending a few millimetres from the edge of the radial extending mycelium. As a result, the conidia within the colony centre are older than those in the intermediate and sub-peripheral zones. Here, it was assessed whether this results in heterogeneity of conidia within the colony. Conidia were harvested from conidiophores of cultures resulting from a confluent inoculated plate. A total number of 645-2421 genes were differentially expressed (fold change ≥ 2) when expression profiles of 2-, 5-, and 8-day-old conidia freshly harvested from conidiophores were compared. Genes without a PFAM or KEGG annotation were enriched in the up-regulated genes.

Expression data from dormant and germinating conidia from Aspergillus niger.

Project description:Conidia of Aspergillus niger are characterized by a dormant state and are moderate stress-resistant. Upon contact with a moist substrate, germination of conidia occurs by changing from a dormant stabilized state towards a growing vegetative cell. The RNA expression levels of dormant conidia and conidia that were in various stages of germination were studied. The RNA composition of dormant conidia was substantially different than all the subsequent stages of germination. This indicates that the distinct morphological changes that occur during germination are not correlated with the highest change in the transcriptome.
