Blood epigenome analysis before and after acupuncture treatment in an outpatient cohort of patients suffering from burnout

ABSTRACT: Genome-wide characterization of epigenetic changes in blood DNA obtained before and after acupuncture treatment in a cohort of patients suffering from Burnout. CpG methylation was measured by using the Infinium MethylationEPIC BeadChip (Illumina) across 859'500 CpGs. Burnout was clinically assessed before and after acupuncture using the Maslach Burnout Inventory (MBI).

Circulating exosomal microRNAs contribute to the antimigraine effects of acupuncture

Project description:We conducted this study to determine whether exosome regulation underlies the antimigraine mechanisms of acupuncture. By comparing serum samples from patients with migraine and healthy controls using high-throughput small RNA sequencing technology , we identified 705 exosomal microRNAs that are differentially expressed in patients with migraine, and this set of 705 microRNAs included five that are particularly well characterised (hsa-miR-369-5p, hsa-miR-1268b, hsa-miR-145-5p, hsa-miR-222-5p, and hsa-miR-4488). By comparing serum samples collected from patients with migraine before and after acupuncture treatment, we showed that acupuncture normalised the expression levels of those five well-characterised exosomal microRNAs.

Gene expression data from thalamic regions of MPTP-intoxicated mouse brain by acupuncture

Project description:Acupuncture stimulations at GB34 and LR3 inhibit the reduction of tyrosine hydroxylase in the nigrostriatal dopaminergic neurons in the parkinsonism animal models. Especially, behavioral tests showed that acupuncture stimulations improved the motor dysfunction in a previous study. The thalamus is a crucial area for the motor circuit and has been identified as one of the most markedly damaged areas in Parkinson’s disease (PD), so acupuncture stimulations might also have an effect on the thalamic damage. We investigated gene expression profile changes in the thalamic region of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism models after acupuncture at the acupoints GB34 and LR3 C57BL/6 mice were divided into four experimental groups; ① C: Control, ② M: MPTP-treatment only, ③ MA: MPTP- and acupuncture-treatment at acupoints GB34 and LR3, ④ MNA: MPTP- and acupuncture-treatment at non-acupoints. Total RNA was isolated from two brains' thalamic regions of each experimental group (4 experimental group × 2 samples of each experimental group = total 8 samples).

Gene expression data from cervical spinal cord regions of MPTP-intoxicated mouse by acupuncture

Project description:It has been shown that acupuncture at acupoints GB34 and LR3 inhibits the degeneration of nigrostriatal neurons in an 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) mouse model of Parkinsonâ??s disease. This preventative effect of peripheral acupuncture stimulation is hypothesized to be transmitted through the spinal cord to the nigrostriatal neurons. The gene expression profile changes following acupuncture at the acupoints were investigated in the cervical spinal cord of an MPTP-induced parkinsonism model using an Affymetrix genechip mouse gene 1.0 ST array. C57BL/6 mice were divided into four experimental groups; â? Investigation of therapeutic response markers for acupuncture in Parkinson's disease: An exploratory pilot study

Project description:Introduction: In our previous study, we demonstrated that acupuncture improved gait disturbances, such as hypometric gait, in patients with Parkinson’s disease (PD). To investigate specific genes that indicate a therapeutic response to acupuncture, we analyzed transcriptome alterations following acupuncture using blood samples collected in our previous clinical trial. Methods: We reanalyzed the clinical data of patients and selected a representative patient with PD for transcriptomic analysis following acupuncture. We examined the expression changes of PD biomarker genes known to be consistently dysregulated in both the brain and blood in patients with PD. We validated these gene expression changes using quantitative real-time polymerase chain reaction (qPCR) in the blood of five other patients with PD who received acupuncture treatment. Results: After acupuncture treatment, the transcriptomic alterations in the representative patient were similar to those induced by dopaminergic therapy. Among PD biomarkers, ankyrin repeat domain 22 (ANKRD22), which is upregulated after dopaminergic therapy, and synapsin 1 (SYN1), which is the common gene to indicate synaptic dysfunction in PD, were upregulated following acupuncture. These alterations correlated with changes of gait parameters in patients with PD.alterations following acupuncture using blood samples collected in our previous clinical trial. Conclusion: This is the first report on gene expression changes and improvement of gait disturbance in patients with PD following acupuncture. Our data suggest that ANKRD22 and SYN1 can be used as biomarkers for therapeutic response to acupuncture in patients with PD, especially those with gait disturbance.alterations following acupuncture using blood samples collected in our previous clinical trial.
