Comparison of rare vs macroscopic human prostate cancer cells in mouse bone

ABSTRACT: Luciferase and GFP labeled Du145 prostate cancer cells were injected into the left cardiac ventricle of CB17 SCID mice and allowed to grow until tumors were detectable in bones. FACS was used to isolate prostate cancer cells as described (Cackowski et al, J Cellular Biochemistry 2017) from bones with detectable tumor or the remaining pelvis, lumbar vertebrae, femorae, tibiae and humorae which had tumor cells present but below the limit of detection by imaging. After RNA isolation and cDNA synthesis, the samples were profiled by bulk RNA sequencing.

SWATH proteomic profiling of prostate cancer cells identifies NUSAP1 as a biomarker of response to Galiellalactone

Project description:Galiellalactone (GL) is a fungal metabolite that presents antitumor and anti-inflammatory activities in vitro and in vivo. Previous studies have shown that GL targets NF-KB and STAT3 pathways and induces G2/M cell cycle arrest in androgen-insensitive prostate cancer cells. In this study, we show that GL-induced cell cycle arrest is independent of the NF-KB and STAT3 pathways in DU145 and PC-3 cells, and also that GL did not affect cell cycling in androgen-sensitive prostate cancer cell such as LNCaP and 22Rv1 cells. In addition, we showed confluence resistance to GL in DU145 cells. Using a SWATH proteomic approach we identified the down-regulation of Nucleolar and spindle associated protein 1 (NUSAP1) under DU145 confluence and in LNCaP cells. Also, the inhibition of NUSAP1 by siRNAs induced resistance to GL in DU145 cells, suggesting that NUSAP1 may be a target for GL and could be useful as biomarker for responsiveness of the antitumor activity of GL.

Cellular and genetic mechanisms that shape the development and evolution of tail vertebral proportion in mice and jerboas

Project description:Limbs and vertebrae elongate by endochondral ossification, but local growth control is highly modular such that not all bones are the same length. Compared to limbs, which have a different evolutionary and developmental origin, far less is known about how individual vertebrae establish proportion. Using the jerboa and mouse tail skeleton, we find that cell number is a common driver of limb and vertebral proportion in both species. However, chondrocyte hypertrophy, which is a major driver proportion in all mammal limbs, is limited to the extreme disproportionate growth of jerboa mid-tail vertebrae. The genes associated with differential growth in the vertebral skeleton overlap significantly but not substantially with genes associated with limb proportion. Among shared candidates, loss of Natriuretic Peptide Receptor 3 in mice causes disproportionate elongation of the proximal and mid-tail vertebrae, in addition to the proximal limb. Our findings therefore reveal cellular and molecular processes tuning growth of individual vertebrae while also identifying natriuretic peptide signaling among genetic control mechanisms that shape the entire skeleton.

Experimental maceration of bovine bones and its effects on the bone proteome

Project description:This study is aimed at understanding how bone soft maceration procedures (e.g., soaking the bones in warm water with the aid of detergents) used to process bones for long term storage (e.g., in human osteological collections) affect the bone proteome. Three different maceration protocols have been tested in the study, in order to test different lenghts for the soaking in water, different temperatures and different detergents. Bovine tibiae (n=6, 3 left and 3 right from 3 animals) have been used in the experiment - in particular, the left ones have been used as non-macerated controls, and the right ones have been macerated with three protocols (one per protocol). Sampling was conducted on both fresh and macerated remains and used to conduct shotgun proteomics analyses.

To furhter understand the role of Lkb1 in this process, we performed microarrrays to compare the transcriptional profile between control and conditional Lkb1 mutant (Col2a1-Cre; Lkb1c/c) chondrocytes. Postnatal day 30 mouse growth plate chondorcytes from control and mutant mouse femurs and tibiae were isolated for RNA extraction and hybridization on Affymetrix microarrays.

Comparative transcriptomic analysis of the proximal tibial growth plates of 14-day old Longshanks and wildtype mice

Project description:This study was undertaken to uncover the molecular pathways that underlie differences among individuals in rates of longitudinal growth and mature length of the long bones in a rodent model. To do so, the transcriptomic profiles of the proximal epiphyses of 14-day old CD-1 wildtype mice and the Longshanks mice were compared. The Longshanks mouse was selectively bred for longer tibiae in relation to body mass (see Marchini et al 2014 – BMC Evol Biol 14:258). Over 13 generations, Longshanks tibiae were 11-12% longer compared to random-bred wildtype mice (hereafter Controls) from the same genetic background (ICR or CD-1), but body mass was unchanged. By comparing the transcriptomes of samples of Control and Longshanks proximal tibiae, we sought to identify differentially expressed genes that were associated with the faster growth and longer tibia of the Longshanks mouse, as a window into the pathways that may underlie intra- and interspecific differences in limb bone size and shape. Results were further validated using qPCR and cell and tissue culture assays.
