Project description:The mature mammalian retina results from a complex series of developmental events. A lot of work now exists on the organization, function, and development of mouse retina, and the high-throughput technologies for gene expression analyses have helped us to obtain deep insight into the mechanism about the genes that control the retinal neurogenesis and vasculogenesis. MicroRNAs (miRNAs) are a class of endogenous non-coding RNAs that inhibit protein translation through binding to target mRNAs. miRNAs have been reported to be involved in regulating multiple physiological and pathological activities, such as ontogenesis, organogenesis, immunoprotection, and tumorigenesis. To identify miRNAs that are specifically regulated in retinal development, total RNAs isolated from retinas isolated from mice in different developmental periods were used for high-throughput sequencing. The data presented here reveals the spatiotemporal miRNA expression patterns which occur during mice retina development and provides a foundation to further investigate how miRNAs contribute to retinal neurogenesis and vasculogenesis.

Project description:Mouse retina is heterogeneous, composed of multiple neuronal and non-neuronal cell types. Among them, bipolar cells (BC), which connect photoreceptors (cones and rods) to inner retina, are traditionally dissected into rare subtypes of subtle functional and morphological differences. While high-resolution single-cell transcriptomic profiles of BCs are availabl, little is known about the corresponding single-cell chromatin landscapes. Although it is now possible to directly generate multiome data, there are often restrictions on cost, feasibility, and data quality. Therefore, integrating single-cell ATAC and RNA profiles obtained independently from the same retina sample may provide an exciting opportunity to comprehensively characterize these rare cell subtypes and discover transcription factors (TFs) important in establishing or maintaining the cell identities

Project description:We investigated the gene expression profile changes after Ezh2 conditional knockout in the mouse retina at E16.5. Loss of Ezh2 leads to up-regulation of PRC2 targeted genes including cell cycle regulators and multiple genes which are not normally expressed in the retina, including many Hox genes. Loss of Ezh2 resulted in a dramatic decline in progenitor proliferation by postnatal day 3, such that there is an early end to neurogenesis, and disruption of laminar organization. Although there are only minor effects on embryonic retinal development, there is accelerated differentiation of several late born cell types postnatally, including photoreceptors and Mueller glia, which become reactive by postnatal day 14. Peripheral retina was dissected at E16.5 from Pax6alpha-Cre:Ezh2fl/+ and Pax6alpha-Cre:Ezh2fl/null mouse embryos. Total RNA was purified and RNA deep sequencing was done using 4 controls and 4 conditional knockout samples.

Project description:<p>The retina is a notable tissue with high metabolic needs which relies on specialized vascular networks to protect the neural retina while maintaining constant supplies of oxygen, nutrients and dietary essential fatty acids. Here we analyzed the lipidome of the mouse retina under healthy and pathological angiogenesis using the oxygen-induced retinopathy model. By matching lipid profiles to changes in mRNA transcriptome, we identified a lipid signature showing that pathological angiogenesis leads to intense lipid remodeling favoring pathways for neutral lipid synthesis, cholesterol import/export and lipid droplet formation. Noteworthy, it also shows profound changes in pathways for long-chain fatty acid production, vital for retina homeostasis. The net result is accumulation of large quantities of mead acid, a marker of essential fatty acid deficiency, and a potential marker for retinopathy severity. Thus, our lipid signature might contribute to better understand diseases of the retina that lead to vision impairment or blindness.</p>

Project description:Regeneration of neurons has enormous implications for human health and is a topic of interest both from the biological and translational perspective. The retina poses an excellent system to study the potential of replacing neurons following injury and the impact different lesions can have on this process. We have established mouse models where proneural transcription factors are expressed in Müller glia to stimulate neurogenesis, which is analogous to how zebrafish regenerate the retina after injury. Our previous studies have shown that Müller glia overexpressing either Ascl1 or Ascl1-Atoh1 will respond to inner retinal damage, caused by a neurotoxic dose of NMDA, by acquiring a progenitor-like state and giving rise to bipolar cells or retinal ganglion-like cells, respectively. It was not known however whether neurogenesis would still occur if the retina suffered outer retinal injury and if the timing of injury relative to the expression of proneural factors was critical for the process. To address these questions, we explored whether timing or mode of injury affect the induced neurogenesis from MG. We show that reprogramming MG with Ascl1 is not impacted by the mode or timing of injury, since all paradigms resulted in similar ratios of new bipolar neurons. By contrast, MG that express Ascl1-Atoh1 respond to outer retinal damage by producing a new type of retinal ganglion-like cell that is not generated after NMDA injury. Therefore, although the fate of reprogrammed neurons is mostly dictated by the proneural transcription factors, there is a context-dependent effect of the injured retinal microenvironment.

Project description:Degeneration of the retina is key to the pathogenesis of some of the most common causes of blindness, but there continues to be a gap in our understanding of the molecular mechanisms leading to retinal degeneration, which in turn has impeded progress towards effective treatments. PRDM13 has been associated with the development of the retina but little is known about the impact of elevated PRDM13 on the mammalian retina, which has been linked to human retinal disease. Here, we developed a mammalian model to induce PRDM13 expression outside of endogenous levels, in a controlled time-dependent manner. We discovered that systemic overexpression of PRDM13 in the adult mouse preferentially affects the retina and reduces photoreceptor function and survival over time. The removal of excess PRDM13 halted this degenerative phenotype and allowed for some restoration of photoreceptor function. Examination of the transcriptomic profile of the retinas prior to photoreceptor degeneration highlighted that genes involved in retinal development, phototransduction, and photoreceptor health were significantly altered by the overexpression of PRDM13. This model system can now be used to further elucidate the direct and indirect targets of PRDM13 that impact photoreceptor maturation, maintenance and function during mammalian retinal development and over adulthood.
