Project description:ITo derive a scaffold from human testis for the purpose of tissue engineering and regenerative medicine, we developed a method to produce a cytocompatible decellularized testicular matrix (DTM) while maintaining the native tissue-specific characteristics and components. The potential benefits of tissue-specific scaffolds consisting of naturally-derived extracellular matrix (ECM) have been demonstrated using a wide variety of animal and human tissue sources. However, so far, testis scaffolds have never been considered for constructive remodelling purposes. We have therefore developed a protocol for the preparation of cell-free extracellular matrix and characterized the material extensively using a combination of proteomics, immunohistochemsitry, and cell population assays. To prepare cell-free matrix, human cadaveric testicular tissue was exposed for 24 h or 48 h to 1% Triton X-100 and/or 1% sodium dodecyl sulfate (SDS). The extent of decellularization was evaluated by histology. Confirmation of cell removal in DTM was done by a DNA quantification technique. The protein composition was analysed by LC-MS/MS. The retention of testicular tissue-specific characteristics was evaluated by immunohistochemistry, Alcian blue staining and scanning electron microscopy. Soluble toxicity and testicular cell attachment was assessed to check the cytocompatibility of DTM scaffolds. Histological analysis showed that DTM could be obtained by mechanical agitation in 1% SDS for 24 h. The resulting DTM was found to be clear of cells while retaining the typical three-dimensional structure. Proteomics analysis revelaed the presence of the major components of the native tissue scaffold, including collagen type I and IV, fibronectin, laminin and glycosaminoglycans. In addition, numerous additional ECM proteins in DTM were detected, indicating its complex nature. Importantly, we demonstrated that DTM scaffolds are not cytotoxic, as evidenced by MTT assay showing a normal fibroblast proliferation activity after indirect exposure, and support testicular cell attachment and infiltration.

Project description:Male reproductive aging involves coordinated changes in germ cells, the testicular niche, mitochondrial homeostasis and the gut microbial–metabolic environment, yet the temporal relationships among these processes remain poorly understood. Here, we profiled male mice at 5, 10, 15 and 20 months using testicular transcriptomics, total and mitochondrial-enriched proteomics, gut 16S rRNA gene sequencing, untargeted gut metabolomics and immune-factor profiling. Across molecular layers, the most pronounced remodeling occurred during late life, particularly between 15 and 20 months, with coordinated attenuation of meiotic, spermiogenic, mitochondrial-translation and respiratory-chain programs. In parallel, senescence-associated, macrophage-associated and inflammatory signals increased within the testicular niche. Mitochondrial-enriched proteomics further uncovered compartment-selective changes that were not consistently reflected at the RNA or total-proteome level. The gut microbial community and metabolic modules also underwent extensive late-life remodeling, and their age-associated patterns covaried with testicular oxidative-phosphorylation, fatty-acid-oxidation and mitochondrial quality-control programs. Collectively, these findings identify late life as a convergence window for spermatogenic, niche, mitochondrial and gut microbial–metabolic aging. By revealing coordinated remodeling across the gut microbial–metabolic environment and testicular mitochondrial programs, this study provides an integrative framework for investigating gut–testis communication and identifying microbial–metabolic processes relevant to male reproductive aging.

Project description:Smooth-muscle-like peritubular cells make up the wall of semniferous tubules of men. These human testicular peritubular cells (HTPC) have been shown to fulfill different roles. They transport sperm, secrete various factors like GDNF (glial cell line derived neurotrophic factor) and are immunologically active. They might contribute to spermatogonial stem cell niche altering and testicular ageing. Previous studies were limited regarding heterogeneity (due to lifestyle, age, medical history etc.) and accessibility of HTPCs. To circumvent this problem a cellular primate model should be established. The proteome of 6 MKTPCs of individual healthy and young (2 or 3 years) donors was assessed to investigate the suitability as a model.

Here, we developed a formation-differentiation culture approach to generate optimized testicular organoids (O-Torgs) derived from neonatal mouse primary testicular cells. These O-Torgs could essentially recapitulate the seminiferous tubule morphogenesis, Sertoli cell maturation and testosterone secretion. Importantly, O-Torgs not only support spermatogenesis but also sustain proliferative undifferentiated spermatogonia and continuous generation of functional haploid cells for up to three months, with offspring derived from these haploid cells displaying normal growth and reproductive capability to F2 generation. Mechanistically, we found that the formation stage enhanced the reconstruction of tubule-like structures and subsequent spermatogenesis by providing a more conducive extracellular matrix niche. Finally, O-Torgs were proved to be effective in modeling male infertility and drug screening, with BTT-3033 acting as a potential drug in protecting busulfan-mediated germ cell loss. Overall, our work establishes a strategy to obtain functional testicular organoids, offering promising avenues for male infertility modeling and drug discovery.

Project description:Establishment and maintenance of spermatogenesis need a complex process and vast regulatory network. There is growing evidence reveals that long noncoding RNAs (lncRNA) plays important role in regulating testicular development and spermatogenesis in a stage-specific way. We here report the identification of lncRNA LOC108635509 as key lncRNA regulator in black goat spermatogenesis. In the current study, we screened the transcriptomes (lncRNA and mRNA) of testicular from Guangxi black goats before puberty (3 days old, D3; 30 days old, D30), puberty (90 days old, D90) and postpuberty (180 days old, D180), and found there were 1211, 12180, 834 differential lncRNAs and 1196, 8838,269 differential mRNAs at the ages of D30 vs D3, D90 vs D30, and D180 vs D90. The expression pattern of differentially expressed (DE) lncRNAs indicated that D90 was a key node of lncRNAs participated in the regulation of testicular development and spermatogenesis in black goat. The of GO and KEGG analyses identified that DE lncRNAs and their target genes regulated spermatogenesis through MAPK, Ras, and PI3K-Akt signal pathways. Using cis- and trans-acting, 39 DE lncRNA-targeted genes were found to be enriched for male reproduction. Of these, LOC108635509, which specific expressed in testis and upregulated the expression levels at D90, was found participated in the regulation of testicular development through promoting the proliferation of SCs. Overall, this study provides new insight into the regulatory mechanisms that support spermatogenesis and testicular development in black goats.
