The differential response of immunity and metabolism leads to differences in the resistance of Siganus oramin and Trachinotus blochii to Cryptocaryon irritans

ABSTRACT: Numerous studies have demonstrated that the C. irritans can be efficiently propagated in the animal model golden pompano (Trachinotus blochii), especially in the process of intensive high-density culture, which causes large-scale infection and triggers bacterial invasion is a devastating blow to the golden pompano industry. This is in sharp contrast to the low sensitivity of S. oramin to C. irritans.

SARS-CoV-2 infection induces transcriptomic alterations specific to activation of innate immunity and cellular distress.

Project description:In the grips of the 2019-2020 SARS-CoV-2 pandemic, the pathogenisis of the virus has been shown to induce sharp responses from the immune system of infected hosts and can result in severe/fatal pulmonary distress. To understand how the virus can provoke such responses in infected hosts we infected golden hamsters with validated SARS-CoV-2 clinical isolates from 5 patients to understand the dynamic changes and functionality of induced gene expression.

Quantitative proteome profiling of Coxiella burnetii reveals major metabolic and stress differences under axenic and cell culture cultivation – proteome dataset

Project description:Coxiella burnetii, a category B select agent, is endemic worldwide, except New Zealand. It causes annually several outbreaks of the zoonotic disease Q fever predominantly in small ruminants. To date, the lipopolysaccharide (LPS), besides a type IV secretion system (T4SS), is the only defined and characterized virulence determinant of C. burnetii. This surface molecule is used to distinguish between virulent (Ph I) and low-virulent (Ph II) organisms, the latter emerge only after frequent passaging in the laboratory. As an obligate intracellular pathogen, targeted genetic modification is still not a routine and labour intensive procedure. The deeper study of novel determinants is complicated and demands not only advanced techniques for axenic and for cell culture-based cultivation but also novel approaches in the high-resolution mass spectrometry. This work is the first proteomic study comparing. C. burnetii Ph I and Ph II propagated in different axenic media and in cell culture.

Comparative Transcriptome Analysis Reveals immunoregulation mechanism of lncRNA-mRNA in Large Yellow Croaker (Larimichthys crocea) in Response to Cryptocaryon irritans Infection

Project description:Cryptocaryonosis caused by Cryptocaryon irritans is one of the major diseases of large yellow croaker (Larimichthys crocea), which lead to massive economic losses annually. The pathogenesis for cryptocaryonosis has been researched by a series of transcriptome studies under different infection conditions. However, little is known about the roles of tissue-specifically expressed genes during the infection of C. irritans. In this study, we analyzed the tissue-specific expression of transcripts in the major infection organs including gill and skin of L. crocea after C. irritans infection. we constructed transcriptome expression profiles of L. crocea gill and skin, including 23,172 protein-coding genes and 7,503 long noncoding RNAs (lncRNAs). By comparing transcriptome data from different tissues of L. crocea, we observed tissue specificity of transcripts in gill and skin, including 3,003 protein coding genes and 639 lncRNAs. A total of 212 of the protein coding genes were involved in immune system. Further analysis revealed that the tissue-specific DEGs in gill and skin were mainly involved in HIF-1 signaling pathway and Complement and coagulation cascades, respectively. In addition, 9 non-tissue-specific hub genes, including CCL4, DDIT4, LEP, ect., which are highly associated with C. irritans infection were identified. To our knowledge, this is the first comparative transcriptome analysis of gill and skin after C. irritans infection. Our results are helpful to understand the molecular mechanism of pathogenesis for cryptocaryonosis, espec-tissue specificity of protein-coding genes and lncRNAs involved in immune regulation.

A single nucleotide polymorphism in the promoter of a hairpin RNA contributes to Alternaria leaf spot resistance in apple (Malus × domestica Borkh.)

Project description:Apple leaf spot caused by the Alternaria alternata f. sp. mali (ALT1) fungus is one of the most devastating diseases of apple (Malus × domestica). We identified a hairpin RNA (hpRNA)-mediated small RNAs, MdhpRNA277, from apple (cv. ‘Golden Delicious’) that is induced by infection with ALT1. MdhpRNA277 produces mdm-siR277-1 and mdm-siR277-2, which target five R genes, MdRNL1, MdRNL2, MdRNL3, MdRNL4, and MdRNL5, that are expressed at high levels in the resistant apple variety ‘Hanfu’ and at low levels in the susceptible variety ‘Golden Delicious’ following ALT1 infection. MdhpRNA277 is strongly induced in ‘Golden Delicious’ but was not induced in ‘Hanfu’ following ALT1 inoculation. The promoter activity of MdhpRNA277 was much stronger in ‘Golden Delicious’ than in ‘Hanfu’ after ALT1 inoculation. We identified a single nucleotide polymorphism (SNP) in the MdhpRNA277 promoter region between the susceptible variety ‘Golden Delicious’ (pMdhpRNA277-GD) and resistant variety ‘Hanfu’ (pMdhpRNA277-HF). The transcription factor MdWHy binds to pMdhpRNA277-GD, but not to pMdhpRNA277-HF. Transgenic ‘GL-3’ apple lines expressing pMdhpRNA277-GD: MdhpRNA277 were more susceptible to ALT1 infection than were those expressing pMdhpRNA277-HF:MdhpRNA277 due to induced mdm-siR277 accumulation and low levels of expression of the five target R genes. The failure of MdWHy to bind to pMdhpRNA277-HF might contribute to the low levels of MdhpRNA277 and mdm-siR277-1/-2 expression and the high levels of R gene expression and resistance to Alternaria leaf spot in resistant apple varieties. We confirmed that the SNP in pMdhpRNA277 is associated with Alternaria leaf spot resistance by analyzing the progeny of three additional crosses. The SNP identified in this study could be used as a marker to distinguish between apple varieties that are resistant or susceptible to Alternaria leaf spot.

Metabolomics of Solanum lycopersicum infected by Phytophthora infestans leads to Late blight detection and reveals metabolic differences in infection times

Project description:Tomato crops suffer attacks of various pathogens that cause large production losses. Late blight caused by Phytophthora infestans is a devastating disease in tomatoes because of its difficultly to control. Here, we applied metabolomics based on liquid chromatography–mass spectrometry (LC-MS) and metabolic profiling by matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) in combination with multivariate data analysis in the early detection of late blight on asymptomatic tomato plants and to discriminate infection times of 4, 12, 24, 36, 48, 60, 72 and 96 h after inoculation (hpi). MALDI-MS and LC-MS profiles of metabolites combined with multivariate data analysis are able to detect early-late blight-infected tomato plants, and metabolomics based on LC-MS discriminates infection times in asymptomatic plants. We found the metabolite tomatidine as an important biomarker of infection, saponins as early infection metabolite markers and isocoumarin as early and late asymptomatic infection marker along the post infection time. MALDI-MS and LC-MS analysis can therefore be used as a rapid and effective method for the early detection of late blight-infected tomato plants, offering a suitable tool to guide the correct management and application of sanitary defense approaches. LC-MS analysis also appears to be a suitable tool for identifying major metabolites of asymptomatic late blight-infected tomato plants.
