Apoptosis as a host defense mechanism in golden pompano (Trachinotus blochii) and its regulatory role in Cryptocaryon irritans infection

Project description:Cryptocaryon irritans causes marine "white spot disease" in various fish hosts, including the golden pompano, a highly sensitive species with significant economic value in the coastal regions of South China. Therefore, exploring the molecular mechanisms that C. irritans infection is important for accelerating molecular breeding programs. In this study, we artificially infected golden pompano with C. irritans. Morphological observations of the gills using tissue sections revealed significant histological changes following infection. RNA-seq was employed to profile gene expression in the gills of golden pompano at different time points post-infection to investigate the molecular mechanisms of the disease. After filtering low-quality sequences, a total of 781,496,198 clean reads were obtained, with Q30 greater than 95%. Differentially expressed genes (DEGs) were identified at 24 hours, 3 days, and 5 days post-infection, with 800, 4208, and 2712 DEGs, respectively. After combining and removing false positives, a total of 2968 DEGs were analyzed for spatiotemporal expression patterns, and these DEGs could be categorized into two down-regulated expression patterns. KEGG enrichment analysis revealed that DEGs were associated with cell-cell junctions, cell-ECM interactions, signal transduction, and immune-related pathways. Further analysis suggested that gill tissue damage during infection led to reduced dissolved oxygen intake, increasing physiological and metabolic stress. This was accompanied by widespread downregulation of collagen genes and deprivation of growth factors, indicating impaired tissue repair. Additionally, as an apoptosis inhibitor, Bcl-2 was found to be downregulated in the infected fish. Therefore, C. irritans infection-induced gill damage leads to hypoxia and subsequently triggers cellular apoptosis.

Intermittent hypoxia: changing energy metabolism patterns of golden pompano is beneficial for coping with hypoxia

Project description:It is currently believed that one of the most severe abiotic stresses in the aquatic environment is hypoxia. However, dissolved oxygen in water is not always at the low concentrations that cause hypoxia, but it is possible to exhibit a state of intermittent fluctuation. The study used a low oxygen concentration (1.8 mg/L) to treat golden pompano (Trachinotus blochii) for 28 days (intermittent hypoxia, 2 hours per day) and systematically evaluated its metabolism indexes. RNA-seq was used to construct a regulatory network to further elucidate the transcriptional regulation of the golden pompano metabolism under intermittent hypoxia. We found that the liver lactic acid content, as well as pyruvate kinase, hexokinase, and phosphofructokinase activities were elevated during the first 7 days, suggesting that anaerobic glycolysis was enhanced during the preceding period. In addition, the triglyceride, lipoprotein lipase, and carnitine palmitoyltransferase-1 levels in the liver were elevated after 14 days, suggesting that lipid utilisation was activated after 14 days. At the same time, intermittent hypoxia increases the activity of aspartate aminotransferase and alanine aminotransferase in the liver, decreases total plasma protein and amino acid levels, and enhances the metabolism of protein and amino acids. Decreased levels of oxidative stress and LOEcrit (the O2 tension for loss of equilibrium) in the golden pompano after 28 days of intermittent hypoxia. Transcriptome analysis showed that the fatty acid metabolism, PPAR signalling pathway, fatty acid degradation, D-amino acid metabolism, and cholesterol metabolism pathway were activated. These results suggest that intermittent hypoxia improves the metabolic pattern of golden pompano, enhances its ability to tolerate hypoxia, and promotes its adaptation to hypoxia environment.

Project description:Maintaining marine salinities in inland aquaculture systems can be cost-prohibitive. As such, there has been an emphasis on the identification of marine species that can be reared at low salinities. The Florida pompano (Trachinotus carolinus) is a popular sportfish that can survive in salinities as low as 5 ppt and has been identified as a potential target species for commercial production. Vibrio spp., bacterial pathogens ubiquitous in aquaculture systems, are capable of quickly decimating aquaculture populations. However, there is limited data available on the effects of salinity influences on Vibrio susceptibility in juvenile Florida pompano. This study investigated the influence of lowered salinity on the immune response of juvenile Florida pompano exposed to Vibrio across a 14-day period through assessment of transcriptomic changes, fatty acid biosynthesis, and identification of internal and external signs of vibriosis. During the experimental period, there were no differences in osmoregulatory or immune related gene expression, no obvious signs of vibriosis, and no changes in fatty acid biosynthesis in fish exposed to 8, 12, 20, or 28 ppt seawater. These results suggest juvenile Florida pompano are resilient to salinity changes and exhibit baseline molecular machinery capable of responding to pathogenic challenges across a range of salinities, making them strong candidates for rearing in inland aquaculture.
