Characterization and MicroRNA Levels of Serum-Derived Extracellular Vesicles in Hepatitis B Virus-Related diseases

ABSTRACT: Serum samples were collected from patients with CHB, DeCi and normal controls. We separated EVs from serum and identified them. MiRNAs from EVs were sequenced by NGS and analyzed to obtain differentially expressed miRNAs. Then we detected the expression level of candidate miRNAs from every single sample by qRT-PCR. The differences of serum EVs in patients with liver disease and 4 miRNAs with significantly different expression levels in EVs were determined. These serum EVs and miRNAs in EVs may be candidates for diagnosing and monitoring liver disease, or important therapeutic targets for the treatment of liver disease.

Macrophage-derived extracellular vesicles regulate concanavalin A-induced hepatitis by suppressing macrophage cytokine production.

Project description:Acute liver failure is a clinical syndrome of severe hepatic dysfunction. Immune cells play an important role in the failure. In recent years, the immunoregulatory function of extracellular vesicles (EVs) has been reported; therefore, it is inferred that EVs have some role in an immune mediated liver injury. In this study, we investigated the immunoregulatory function of EVs in a concanavalin A (Con A)-induced liver injury. The mouse model was prepared by a single intravenous administration of 15 mg/kg Con A, in which there was a significant increase in serum EVs number. In in vitro study, the secreted EVs number was also significantly increased in Con A-treated RAW264.7, a mouse macrophage cell line, but not Hepa1-6, a mouse hepatoma cell line. In in vitro EVs treatment study, the EVs from Con A-treated mice serum and Con A-treated RAW264.7 suppressed the inflammatory cytokines production in Con A-stimulated RAW264.7. miRNA sequencing analysis showed that mmu-miR-122-5p and mmu-miR-148a-3p were commonly increased in these EVs and the EVs-treated cells. The enriched pathways in the miRNAs predicted target genes included the inflammatory response pathways. mRNA levels of the target genes in the pathways (mitogen-activated protein kinase, Phosphoinositide 3-kinase/Akt and Rho/Rho associated coiled-coil containing protein kinase pathways) were decreased in the EVs-treated cells. In in vivo RNA interference study, knock down of liver RAB27A, an EVs secretion regulator, significantly exacerbated the Con A-induced liver injury. These data suggest that macrophage-derived EVs play an important role in a Con A-induced liver injury through the immunoregulation.

Identification of the serum microRNAs (miRNAs) displaying differential levels between hepatocellular carcinoma (HCC) patients and chronic hepatitis B (CHB) controls

Project description:To identify serum miRNAs for the diagnosis of HCC, the profiling of 754 human miRNAs was analyzed in sera from six histopathologically confirmed HCC patients and eight CHB controls, using TaqMan Array Human MicroRNA A+B Cards Set v3.0 (Applied Biosystems, Foster City, CA). Serum miRNAs with differential levels between HCC and CHB were identified by comparing serum miRNA profiling of HCC patients with that of CHB controls.

Identification of miRNAs that regulate hepatitis B virus replication and are potential biomarkers for therapeutic outcome

Project description:Background: Chronic hepatitis B (CHB) infection is a serious health problem worldwide and with the current treatment a functional cure is only achieved in 3-15% of the patients. Therefore it is important to investigate novel biomarkers for treatment outcome and to develop new treatment options for CHB. miRNAs are small, non-coding RNA molecules that are involved in various cellular processes by regulating gene expression at the posttranscriptional level and also play an important role in a variety of infectious diseases. In this study we want to identify novel miRNAs that are involved in hepatitis B replication and could therefore be used as biomarkers and targeted as a potential new therapeutic option. Methods: miRNA/small RNA NGS was performed on total RNA isolated from 10 matching plasma and liver biopsy of an identification cohort of CHB patients and 10 plasma and liver controls. Results: We identified 23 and 18 miRNAs that were differentially expressed between controls and CHB patients in plasma and liver respectively, of which five were differentially expressed in both liver and plasma. Of these candidate miRNAs 3 were upregulated in liver and 12 in plasma from CHB and 15 were downregulated in liver and 11 in plasma.

Identification of differentially expressed microRNAs in liver tissue of Hepatitis B virus (HBV) infected hepatocellular carcinoma (HCC)

Project description:Global expression profiling of miRNAs in liver tissue of HBV infected HCC and chronic hepatitis B (CHB) with no fibrosis was evaluated. A total of 40 differentially expressed miRNAs were identified in HCC. Top 10 miRNAs are validated in more numbers of HCC tisuues by qRT PCR. Finally six miRNAs (miR-15a, miR-16, miR-21,miR-29b-3p, miR-126, miR-142-3p, miR-193a-5p) showed similar expression pattern in both microarray and qRT PCR Differential expression analysis of microRNAs in HBV infected HCC (n=4) compared to CHB patients with no fibrosis (n=8) as control.

Project description:miRNAs have been found to circulate in the blood in a cell-free form; their potential as readily accessible disease markers is currently evaluated. Here, we investigated if miRNA expression profile in peripheral blood can be useful as disease parameter in patients with chronic hepatitis C (CHC), chronic hepatitis B (CHB), non-alcoholic steatohepatitis (NASH), and normal liver (NL). RNA from exosome rich fraction was extracted from serum of patients with 64 CHC, 4 CHB, 7 NASH, and 12 healthy volunteers. miRNA expression profile was analyzed by microarray. Histological evaluation for the degree of inflammation and fibrosis in liver disease patients was also performed. miRNA expression profile according to the degree of liver fibrosis and inflammation was established. Diagnosis of fibrosis, inflammation by using miRNA expression pattern was shown with accuracy 67.2~89.1%, 67.2~76.5%, respectively. Moreover, miRNA expression profile in several liver diseases was also established. Classification among CHC, CHB, NASH, and NL was well tolerated and reproducible. The expression pattern of miRNA in peripheral blood showed not only the useful parameter of the grade and stage of liver disease, but also diagnostic tool for liver disease.

Acute and chronic liver injury change the small RNA cargoes carried by serum small extracellular vesicles in mice

Project description:Compared to whole serum miRNAs, miRNAs in serum small extracellular vesicles (sEVs) are well protected form RNA enzymes, thus provide a consistent source of miRNA for disease biomarker detection. Serum sEVs and their miRNA cargos released by injured liver cells could be promising biomarkers for diagnosis of liver diseases. We were very interested to find out the effects of liver injury on serum extracellular vesicles as well as the small RNA components they transported, if there is any difference between acute and chronic injury. Study in this regard will help us to identify new serum biomarkers for liver injury, and to find out if there are specific markers for acute or chronic liver injury. To identify potential biomarker for liver injury based on serum sEVs miRNAs, we established the carbon tetrachloride (CCL4) induced acute and chronic liver injury mice model, and examined the dynamic changes of small RNA components, especially miRNAs, in serum sEVs.

Project description:With the goal to identify miRNAs and other small RNAs involved in EV-mediated oligodendrocyte-neuron communication, we performed deep sequencing (RNA-seq) of small RNAs derived from EVs isolated from culture supernatants of primary mouse oligodendrocytes by differential centrifugation. Oligodendrocytes were cultured and EVs harvested under serum-free conditions in presence of B27 supplement. We found that RNA isolated from EVs harvested from cells under serum-replacement conditions contains miRNA contaminants carried into the sample by defined media components.

Surgical trauma in mice modifies the content of circulating extracellular vesicles

Project description:Purpose: Aseptic tissue injuries including major surgeries are associated with structural and functional changes in the brain suggesting the existence of periphery-to-brain communication. However, the nature of the carriers of such signaling remains largely unknown. Analyzis of the miRNAome and proteome of circulating extracellular vesicles (EVs) to evaluate their potential contribution to the periphery-to-brain signaling. Methods: EVs were isolated from the serum of the mice subjected to a tibia surgery after 6, 24, and 72 h, using Exoquick ULTRA commercial ki. Proteome and miRNAome were mapped using mass spectrometry and RNA-seq approaches. Results: Next-generation sequencing of small RNA libraries prepared from the total RNA isolated from the mouse serum EVs detected 252 miRNAs. Differential expression analysis using DESeq2 statistical package revealed 50 dysregulated miRNAs in the 24 h post-surgery EVs. The surgery effect declines after 72 h as judged by the lower number of dysregulated miRNAs. The differentially expressed miRNAs were examined by the DIANA-miRPath online tool that predicted dysregulation of several KEGG pathways including pathways involved in the lipid metabolism, ECM-receptor interaction, gap junction, various signal transduction pathways. Conclusions: The main question of the current project, is the surgery capable of modifying the expression of cargo molecules in circulating EVs, can be answered positively. Moreover, differential expression of a specific set of miRNAs in the “surgical” EVs suggests that they can be involved in the regulation of several metabolic pathways, such as ECM-receptor interaction and lipid metabolism in the recipient tissues, which is consistent with our data on hippocampal metabolic and functional dysregulation after surgery We conclude that surgery alters the EV composition in the blood, which paves the way for further studies on the functional effects of these EVs on the brain.
