Project description:Mitosis segregates into each daughter cell two centrioles, the older of which is uniquely capable of generating a cilium. How this older centriole, called the mother centriole, initiates ciliogenesis remains poorly understood. We have identified an evolutionarily conserved complex comprised of CEP90, OFD1, MNR and FOPNL. Human mutations in CEP90, MNR and OFD1 cause ciliopathies. Super-resolution microscopy revealed that this complex forms a ring at the distal centriole. Centrioles of cells lacking MNR or CEP90 failed to assemble distal appendages and cannot generate cilia. In addition to the centrioles, complex members localized to centriolar satellites, proteinaceous granules surrounding the centrioles. Disruption of satellites did not affect distal appendage assembly, indicating that the centriolar pool is required for ciliogenesis. Consistent with an essential role in ciliogenesis, mice lacking MNR or CEP90 did not assemble cilia, did not survive beyond embryonic day 9.5, and did not transduce Hedgehog signals. In addition to ciliogenesis, MNR, but not CEP90, restrained centriolar length. MNR recruited both OFD1, required for centriolar length control, and CEP90, which recruits CEP83 to root distal appendages. Thus, an evolutionarily conserved ciliopathy-associated complex functions at the distal centriole to control centriole length and assemble distal appendages.

Project description:Control of the number of centrosomes is critical for cell division, trafficking and cilia. Regulation of centrosome number occurs through the precise duplication of centrioles that reside in the center of centrosomes. Here we explored transcriptional control of centriole assembly by focusing on alternative splicing factors in isolation from other cell cycle processes. Of five splicing factors originally identified as required for centriole assembly, only SON is specifically required. Procentriole assembly is severely disrupted when SON is reduced but early centriole assembly events still occur. Whole genome mRNA sequencing identified thousands of genes whose splicing and expression are affected by the reduction of SON, with an enrichment of genes involved in the microtubule cytoskeleton. SON is required for the proper splicing and expression of the centriolar satellite protein, CEP131. Fluorescence microscopy and electron tomography establish key differences to the trafficking and microtubule network around the centrosomes that contribute to the centriole assembly defects. This establishes SON as required for centriole assembly, partially through its activity in splicing CEP131 and through control of microtubules organized by the centrosome.

Project description:Multiciliated cells (MCCs) harbour dozens to hundreds of motile cilia, which beat in a synchronized and directional manner, thus generating hydrodynamic forces important in animal physiology. In vertebrates, MCC differentiation critically depends on the synthesis and release of numerous centrioles by specialized structures called deuterosomes. Little is known about the composition, organization and regulation of deuterosomes. Here, single-cell RNA sequencing reveals that human deuterosome-stage MCCs are characterized by the expression of many cell cycle-related genes. We further investigated the uncharacterized vertebrate specific cell division cycle 20B (CDC20B) gene, the host gene of microRNA-449abc. We show that the CDC20B protein associates to deuterosomes and is required for the release of centrioles and the subsequent production of cilia in mouse and Xenopus MCCs. CDC20B interacts with PLK1, which has been shown to coordinate centriole disengagement with the protease Separase in mitotic cells. Strikingly, over-expression of Separase rescued centriole disengagement and cilia production in CDC20B-deficient MCCs. This work reveals the shaping of a new biological function, deuterosome-mediated centriole production in vertebrate MCCs, by adaptation of canonical and recently evolved cell cycle-related molecules. A specific aim of this mass spectrometry experiment was to verify on immunoprecipitated protein complexes from CDC20 or CDC20B transfected HEK cells that CDC20, but not CDC20B, interacts with multiple APC/C components.

Four total samples were analyzed. Two biological replicates of proliferative zone chondrocytes microdissected from control mice and two biological replicates from Ift88 deleted (Col2aCre;Ift88fl/fl) mice. Project description:Centriole biogenesis and maintenance are crucial for cells to generate cilia and assemble centrosomes that function as microtubule organizing centers (MTOCs). Centriole biogenesis and MTOC function both require the microtubule nucleator -tubulin ring complex (TuRC). It is widely accepted that TuRC nucleates microtubules from the pericentriolar material (PCM) that is associated with the proximal part of centrioles. However, TuRC also localizes more distally and in the centriole lumen, but the significance of these findings is unclear. Here we identify spatially and functionally distinct sub-populations of centrosomal TuRC. Luminal localization is mediated by augmin, which is linked to the centriole inner scaffold through POC5. Disruption of luminal localization impairs centriole integrity and interferes with cilium assembly. Defective ciliogenesis is also observed in TuRC mutant fibroblasts from a patient suffering from microcephaly with chorioretinopathy. These results identify a novel, non-canonical role of augmin-γTuRC in the centriole lumen that is linked to human disease.

Project description:Multiciliated cells (MCCs) form when progenitors massively expand centriole number, yielding the hundreds of basal bodies required to extend multiple motile cilia. This organelle biogenesis is promoted transcriptionally by Multicilin acting in a complex with the E2F transcription factors, which activates the expression of genes known to be involved required to form centrioles during the cell cycle, but also requires a cell cycle state required for the centrosome cycle. Multicilin also activates the expression of Emi2 (fbxo43) an inhibitor of the APC/C ubiquitin ligase. In Emi2 mutants, basal body formation during MCCl differentiation is blocked. RNAseq analysis is used to show that gene expression associated with MCC differentiation is upregulated normally in Emi2 mutants, but surprisingly is not downregulated when MCC differentiation is complete. Emi2 is required to promote the centrosomal cycle during MCC differentiation, but then also acts to turn off gene expression associated with centriole biogenesis in differentiated cells.

Project description:Primary cilium serves as a cellular M-bM-^@M-^\antennaM-bM-^@M-^] to sense environmental signals. Ciliogenesis requires the removal of CP110 to convert the mother centriole into the basal body. Actin dynamics is also critical for cilia formation. How these distinct processes are properly regulated remains unknown. Here we show that miR-129-3p, a microRNA conserved in the vertebrates, controlled cilia assembly by down-regulating both CP110 and four proteins critical for actin dynamics, Arp2, Toca1, abLIM1, and abLIM3. Consistently, blocking miR-129-3p repressed cilia formation in cultured mammalian cells, whereas its overexpression potently induced ciliogenesis in proliferating cells and extraordinary cilia elongation. Moreover, inhibition of miR-129-3p in zebrafish embryos suppressed cilia assembly in the KupfferM-bM-^@M-^Ys vesicle and pronephric duct, leading to developmental abnormalities including curved body, pericardial oedema, and randomised left-right patterning. Our results thus unravel a novel mechanism that orchestrates both the centriole-to-basal body transition and subsequent cilia assembly via microRNA-mediated posttranscriptional regulations. We want to find the targets of miR-129-3p by overexpressing miR-129-3p oligo or control oligo in hTERT-RPE1 cells. Through microarray analysis we could check the downregulated genes and these genes might be the targets of miR-129-3p. Project description:Centrioles are evolutionary conserved macromolecular structures that are fundamental for templating the formation of cilia and flagella, as well as for assembling centrosomes. Centrioles are nine-fold symmetrical microtubule-based cylindrical structures, which have three regions that can be clearly distinguished in the Chlamydomonas reinhardtii organelle: a ~100nm-long proximal region harboring a cartwheel, a ~250nm-long central core region containing the Y-shaped linker and a ~150nm-long distal region ending at the transitional plate. Despite the discovery of a growing number of centriolar components in many systems, no protein has been attributed to the central core region of the centriole thus far. Here, combining relative quantitative mass spectrometry and super-resolution microscopy on purified Chlamydomonas centrioles, we identified POB15 and POC16 as two proteins of the central core region. We also uncovered that POB15 and POC16 distribution correlates with that of centriolar microtubule glutamylation. Moreover, we developed an assay to uncover temporal relationships between centriolar proteins during organelle assembly, and thus established that POB15 is recruited after the cartwheel protein CrSAS-6, but before microtubule glutamylation takes place. Furthermore, we uncovered that WDR90, the human homolog of POC16, localizes to the distal lumen of human centrioles and is required for proper ciliogenesis. Overall, our work provides the first insights into the molecular identity and the function of components of the central core region of centrioles.

Project description:It is unclear that osteoclast lineages can sense mechanical stimuli, mainly by lack of information about the presence or absence of mechano-sensing organelles in these cells. Primary cilium is a microtubule-based organelle, which can translate mechanical loading signals into biochemical and transcriptional responses. We first identified the presence of primary cilia in tartrate-resistant acid phosphatase (TRAP)-positive mononuclear cells (preosteoclasts) prior to form TRAP-positive multinuclear cells (osteoclasts), suggesting that preosteoclasts can sense mechanical stimuli. To understand a role of primary cilia in preosteoclasts, we generated osteoclast-specific Ift88 or Kif3a knockout Cathepsin K-Cre transgenic mice. Deletion of Ift88 or Kif3a reduced the number of cilia in preosteoclasts. Micro-CT analysis display that mice with conditional deletion of Ift88 or Kif3a in osteoclast lineages led to a decrease in femoral cortical bone tissue area and bone marrow area under exercise conditions, implying that disruption of primary cilia in preosteoclasts/osteoclasts narrowed the femoral cortical bone shape. In contrast, the trabecular and cortical bone mass was not altered in these mice. Mechanistic studies showed that shear stress suppressed osteoclastogenesis by c-fos degradation. Also, shear stress increased periostin levels in conditioned media from preosteoclasts, resulting in increased differentiation and bone nodule formation of osteoblasts through activation of LRP6/-catenin. All shear stress-induced changes were near completely blocked by knock-downs of Ift88 and Kif3a in preosteoclasts. These findings first suggest that preosteoclasts may sense mechanical stimuli with a primary cilium, and that mechanical stress-mediated primary cilia activation of preosteoclasts may play an important role in controlling bone modeling and shaping by modulating both bone resorption and formation.
