CD4+ T cells mRNA profiles of lean and obese mice

ABSTRACT: To figure out how obesity drives glycolysis in CD4+ T cells, we performed RNA-sequencing to analyze the transcriptome of lean and obese-derived splenic CD4+ T cells. Mice were fed with either a normal diet (referred to lean mice) or a high-fat diet (HFD, 60 kcal% fat, referred to obese mice) generally from 6-8 weeks of age, for up to 4 months.

Adipose tissue stromal cells from lean, obese, and formerly obese mice

Project description:Adipose tissue stromal cells contribute to the regulation of adipose tissue in lean and obese states. Myeloid cells such as adipose tissue macrophages (ATMs) and dendritic cells (ATDCs) undergo both quantitative and qualitative changes with obesity. Due to similarity in markers the identify of adipose tissue dendritic cells and macrophages has been elusive. We have refined prior protocols to unambiguously discern ATM and ATDC in mice. We used microarrays to compare the profiles of ATMs and ATDC from gonadal adipose tissue from lean, obese, and formerly obese mice. We also isolated preadipocytes (PA) from lean and obese mice for comparison. Male C57Bl/6 mice were fed normal diet (ND) or high fat diet (HFD) for 16 weeks. Weight loss (WL) mice were switched from the HFD to ND for 8 weeks. RNA was purified from FACS sorted cell populations (live cells only) obtained from gonadal/epididymal adipose tissue depots. ATMs were defined as CD11c+ (CD45+CD64+ CD11c+) or CD11c- (CD45+CD64+ CD11c-) ATMs. ATDC were defined as CD64- CD11c+. PA were defined as CD31- CD45- Sca1+ PDGFRA+.

Single-cell transcriptional profiles in the visceral adipose tissue from obese and lean C57BL6 mice

Project description:Cellular senescence has been reported to play an important role in insulin resistance. While the molecular mechanism remains unclear. Here we applied the scRNA-seq analysis for the visceral fat from the obese mice with 2 month of high-fat diet. Visceral fat was digested by type II collagenase. Stromal Vascular Fraction was collected, incubated with hashing antibody and then subjected to the 10X Chromium. 8180 cells were found from the obese mice and 14,502 cells were from the lean mice. 14 clusters and 9 cell types were identified. The key senescence marker P21 were found highly expressed in the obese group, when compared to the lean group. However, the other senescence marker P16 were unchanged. P21+ cells are mainly preadipocytes, macrophage and endothelial cells. More than 300 different expressed genes were found among the P21+ cells. The NF-kB pathway was activate according to the GO analysis, which is strongly associated with insulin resistance and diabetes

High fat diet mice intestinal epithelial exosomal proteins were analyzed by MSMS.

Project description:High-fat diet (HFD) decreases insulin sensitivity. How high-fat diet causes insulin resistance is largely unknown. Here, we show that lean mice become insulin resistant after being administered exosomes isolated from the feces of obese mice fed a high-fat diet (HFD) or from human type II diabetic patients with diabetes. HFD altered the lipid composition of exosomes from predominantly PE in exosomes from lean animals (L-Exo) to PC in exosomes from obese animals (H-Exo). Mechanistically, we show that intestinal H-Exo is taken up by macrophages and hepatocytes, leading to inhibition of the insulin signaling pathway. Moreover, exosome-derived PC binds to and activates AhR, leading to inhibition of the expression of genes essential for activation of the insulin signaling pathway, including IRS-2, and its downstream genes PI3K and Akt. Together, our results reveal HFD-induced exosomes as potential contributors to the development of insulin resistance. Intestinal exosomes thus have potential as broad therapeutic targets.
