Genome-wide chromatin binding of YBX1 in nasopharyngeal carcinoma (NPC) cell line S18 (ChIP-Seq)

ABSTRACT: The development of therapeutic resistance and metastatic dissemination takes place in 20-30% patients with nasopharyngeal carcinoma (NPC), resulting in poor survival. Hence, a better understanding of the underlying molecular mechanisms will help improve clinical outcome. We have identified a novel circRNA, circIPO7, as a promoter of metastasis and cisplatin chemoresistance in NPC cells. circIPO7 was found to bind to YBX1. Genome binding/occupancy profiling by high throughput sequencing were performed to explore the underlying molecular mechanisms.

Project description:Circular RNAs have been demonstrated to play significant roles in multiple biological processes, such as immune responses and tumorigenesis, suggesting it might be potential therapeutic targets. However, the function and mechanisms of circRNAs in nasopharyngeal carcinoma (NPC) metastasis remain largely unknown. In this study, we identified circWDR37 as a metastasis related circRNA. To further explore the underlying mechanism of circWDR37 promoting NPC metastasis, we performed RNA sequencing analysis of NPC cell line S18 upon circWDR37 knockdown.

Microarray expression profiles in S18 and S26 human nasopharyngeal carcinoma cell lines

Project description:Metastasis accounts for the leading cause of treatment failure in nasopharyngeal carcinoma (NPC). However, the exact mechanisms underlying metastasis remain elusive. S18 and S26 are a pair of NPC cell lines with high and low metastasis abilities, respectively. To evaluate the expression profiles of mRNA in S18 and S26 cells will help to demonstrate genes that contribute the metastasis of NPC. We performed microarray sequencing to clarify the genes that differentially expressed in S18 and S26 cells.

NESG1 cDNA was transfected into NESG1-negative NPC 5-8F cells and affymetrix microarrays HG-U133_Plus_2 were used to analyze the differential genes in NESG1-overexpressed NPC cells and their control NESG1-negative NPC cells.

Project description:Epstein-Barr virus (EBV) is a ubiquitous human herpesvirus associated with nasopharyngeal carcinoma (NPC). EBV encodes two groups of microRNAs (miRNAs) which are divided into BamHI fragment H rightward open reading frame 1 (BHRF1) and BamHI-A rightward transcripts (BART) microRNAs. EBV miR-BARTs have been found to be involved in the development and progression of NPC. However, the role of EBV-miR-BARTs in NPC progression remains illusive. This study aimed to investigate the role of EBV-miR-BARTs in NPC and explore the underlying mechanisms.

Gene expression data of nasopharyngeal carcinoma (NPC) cell and Thiostrepton treatment

Project description:Thiostrepton, a natural antibiotic, has recently been shown to be a potential anticancer drug for certain cancers, but its study in nasopharyngeal carcinoma (NPC) is still limited. The aims of this study were to investigate the anticancer effect of thiostrepton on NPC cells and to explore its underlying mechanism. Our results showed that thiostrepton reduced NPC cell viability in a dose-dependent manner. Thiostrepton inhibited the migration and invasion of NPC cells in wound healing and cell invasion assays. Thiostrepton effectively suppressed NPC cell proliferation, migration, and invasion, likely by several mechanisms. Thiostrepton may be a potential therapeutic agent for treating NPC in the future.

Project description:Nasopharyngeal carcinoma (NPC) is a common cancer in southern China and South East Asia where more than 50,000 new cases are diagnosed each year. We used microarrays to identify down or upregulated genes in NPC compared with non-malignant controls. Experiment Overall Design: Snap frozen nasopharyngeal biopsies from 25 patients with histologically confirmed undifferentiated NPC were included in the microarray analysis. Controls were obtained from 3 patients with no evidence of malignancy.

microRNA profile of human nasopharyngeal tissues: nasopharyngeal carcinoma tissues vs. normal nasopharyngeal tissues

Project description:MicroRNAs are biomarkers of prognosis and survival for many types of cancer. We evaluated whether microRNAs can predict the survival and efficacy of concurrent chemotherapy in nasopharyngeal carcinoma (NPC) patients. We retrospectively analyzed microRNA expression in 312 paraffin-embedded NPC specimens and 18 normal nasopharyngeal tissues using microarray. We found Forty-one microRNAs are differentially expressed between NPC and normal tissues, and a five-microRNA signature can predict survival independent of stage.
