Infiltration of monocyte-derived macrophages perturbs the CNS metabolic landscape, licensing arginine catabolism and augments neuro-inflammation

Project description:Inflammatory responses are associated with monocyte recruitment into inflamed tissues, where they differentiate into monocyte-derived macrophages (moMACs). While tissue-specific reprogramming of moMACs is well-established, how they alter tissue metabolism remains unclear. Here, using a murine neuro-inflammation model coupled with genetic fate-mapping, metabolomics and metabolite imaging, we identify that central nervous system (CNS) moMACs infiltration is associated with substantial metabolic changes, assigning disease-associated metabolites (DAMs) therein. Particularly, we found decreased arginine levels due to increased arginine catabolism driven by lesion-associated arginase 1 (Arg1) expressing moMACs. Arg1-expressing moMACs were linked to disease severity and via their impacts on arginine were engaged in lipid-metabolism and pro-inflammation. Genetic Arg1 deficiency within moMACs during neuro-inflammation increased extracellular arginine, improved outcome, and resulted in rewiring of the CNS metabolic landscape, associated with attenuated DAMs and regulatory T-cell expansion. Together, we demonstrate key roles for moMACs-driven arginine catabolism in neuro-inflammation.

Identification and characterization of alveolar and recruited lung macrophages during acute lung inflammation

Project description:To precisely identify mice resident alveolar macrophages (AMs) and bone marrow (BM)-derived macrophages, we developed a technique to separately label AMs and BM-derived macrophages with a fluorescent lipophilic dye followed by fluorescent-associated cell sorting (FACS). We utilized this approach to isolate BM-derived macrophages and AMs at different timepoints after intratracheal (IT) instillation of E.coli lipopolysaccharide (LPS) and performed bulk RNAseq on these cell populations to characterize the individual phenotypes of resident AMs and BM-derived macrophages present in the lungs.

The JMJD3-IRF4 axis regulates M2 macrophage polarization and host responses against helminth infection

Project description:Polarization of macrophages to M1 or M2 cells is important for mounting responses against bacterial and helminth infection respectively. Jumonji domain containing 3 (JMJD3), a histone 3 K27 demethylase, has been implicated in the activation of macrophages. Here we show that JMJD3 is essential for M2 macrophage polarization to helminth infection and chitin, though JMJD3 is dispensable for M1 responses. Furthermore, Jmjd3 is critical for proper bone marrow macrophage differentiation in a demethylase activity-dependent manner. Jmjd3 deficiency affected trimethylation of H3K27 in only a limited numbers of genes. Among them, we identified Irf4 as the target transcription factor critical for controlling M2 macrophage polarization. Collectively, these results show that JMJD3-mediated H3K27 demethylation is critical for regulating M2 macrophage development leading to anti-helminth host responses. Human resistin alters lung mRNA expression from helminth-infected lungs.

Project description:Goal: To examine the effects of human resistin during helminth infection. Methodology: To examine the function of human resistin (hResistin), we utilized transgenic mice expressing the human resistin gene along with its entire regulatory region (hRetnTg+). Following infection with the helminth Nippostrongylus brasiliensis, whole lung RNA was sequenced in hRetnTg+ mice, control hRetnTg- and naïve mice. Conclusion: In hRetnTg+ mice, many genes involved in inflammation and the immune system, specifically toll-like receptor signaling and chemokines, are significantly upregulated, suggesting that human resistin promotes TLR signaling and inflammation during helminth infection.
