Transcriptional expression of different grafting combinations during healing

ABSTRACT: Grafting was widely used in pear cultivation as an ancient cultivation technique, the use of rootstocks could improve the resistance of pear trees, make pear trees early fruit. The healing process of plant grafting involves complex signal transduction and material accumulation.We used two different grafting combinations, include ‘Qingzhen D1’ on ‘QAUP-1’ and ‘Qingzhen D1’ on ‘Qingzhen D1’.The samples which are the part of graft union of those two combinations were collected om 1 day, 5 days , 9 days and 30 days after grafting. We performed an in-depth RNA-seq analysis to observe the transcriptional changes that occur during the development of two combinations.

Epigenetic bases of grafting-induced vigour in eggplant [RNA-Seq]

Project description:In horticulture, grafting is a popular technique used to combine positive features of two different plants, obtained by joining a scion (generally the part above the ground) onto a rootstock (constituted by the stem and roots). Despite its wide-use, the biological mechanisms driving rootstock-induced alterations of the scion phenotype are not fully understood. Given that epigenetics is an important component of distance signalling in plants, we investigated the genome wide changes in the DNA methylation induced in eggplants grafted onto two interspecific rootstocks commercially used to induce scion vigour, compared to self-grafted plants. We found that vigour was associated to a specific change in scion gene expression and a genome wide hypo-methylation in CHH context. Interestingly, this hypomethylation correlated with downregulation of younger and potentially more active LTR retrotransposons. Our data indicate that graft-induced epigenetics modifications are associated to both physiological and molecular phenotypes in plants, and suggest that rootstocks can induce vigour by reducing DNA methylation in the scion genome, similarly to what observed in some heterotic hybrids.

Rootstock-dependent gene expression profiling in apple scions

Project description:The purpose of this project is to examine the effects of rootstocks on the gene expression patterns in scions of apple trees. Gene expression patterns were examined in the Gala variety grafted onto seven different, commonly used rootstocks. These trees were grown in the greenhouse to limit environmental effects. Also, gene expression profiles were examined in three different varieties (Ambrosia, Melrose,and Gala) grafted onto B.9 rootstocks grown in the field. Each sample is a pool of RNA from two different trees. RNA samples were isolated from 0.5 g of actively growing shoot tips, including leaf and stem tissues.

Oligoarray analysis of semi-global gene expression in ripening Japanese pear fruit

Project description:A custom oligoarray of Japanese pear (Pyrus pyrifolia) based on 9,812 independent ESTs from different tissues (fruits at various growth stages, vegetative and flower tissues) was designed and used for comprehensive investigation of gene expression before and during ripening (105 to 147 days after full bloom). Gene expression in fruit development of Japanese pear was measured from 105 to 147 days after full bloom (DAFB). 147 DAFB is the optimum maturity for eating. Two to three independent experiments were performed at each time (105 to 147 DAFB) using different trees for each experiment.

Grapevine grafting: scion transcript profiling and defense-related metabolites induced by different rootstocks.

Project description:Genotype, climate, soil qualities, and rootstocks are among the main factors that influence grape development as well as fruit and wine composition. In this work, the rootstock/scion interactions were studied using transcriptomic and metabolic approaches on leaves of Gaglioppo variety, grafted onto thirteen different rootstocks growing in the same locations. The experiments were performed in a vineyard in the south of Italy, characterized by the Mediterranean climate with warm and dry summers. Whole leaf transcriptome of ‘Gaglioppo’ grafted on five selected rootstocks showed high variability in gene expression. In particular, significant modulation of transcripts linked to primary (e.g. carbohydrate and transport) and secondary metabolism (e.g. phenylpropanoids pathways and response to stress) was observed. Interestingly, genes involved in the priming defense responses (e.g stilbenes and defense genes) were strongly activated in some graft combinations. These results were further confirmed by the quantification of stilbene contents. According with these observations, the leaves of ‘Gaglioppo’ grafted on 1103 Paulsen showing the lowest levels of stilbene synthase transcripts and stilbene metabolites, reveal the greater sensitivity to downy mildew in in vitro assay. This study carrying out an extensive analysis of rootstock effects to scion leaves open a useful way to unravel this complex interaction

Transcription changes at sub-optimal root temperature in different grafting combinations of tomato rootstocks and scions

Project description:The commercial tomato cultivar âKommeetâ was grafted either onto the tomato cultivar âMoneymakerâ (sub-optimal temperature sensitive) or onto the line accession âLA 1777â of the wild tomato species S. habrochaites (sub-optimal temperature tolerant) in a heated glasshouse. Grafted tomato plants were grown in a nutrient film technique system with re-circulating nutrient solution resulting in different root temperature (T), which was either optimal (day and night 25Â±0.6Â°C) or sub-optimal (day and night 15Â±0.4Â°C) while the air T was optimal (day and night 25Â±0.6Â°C) throughout the experimental procedure. After 30 days of differential treatment, differences in growth, physiology, and global gene expression in roots and leaves of all grafting and T combinations were investigated.
