Project description:It has been established that the frequently reported early flowering phenotype of the phyB mutant is very sensitive to small changes in ambient temperature (Halliday et al., 2003). A 6oC drop in temperature to 16oC is sufficient to abolish the pronounced early flowering phyB phenotype observed at 22oC. Using Affymetrix technology we will identify genes that are differentially regulated at 16oC and 22oC in a phyB-specific manner using phyB mutant and wild type seedlings. 16 samples were used in this experiment.

Project description:In this study we used genetic approaches and transcriptome profiling to unravel the complex interaction of different developmental pathways required for chloroplast development in plants. The recently described snowy cotyledon 3 (sco3) mutant as well as the Phytochrome B (phyb) mutant revealed, in the double mutant, a complex suppressive or additive genetically linked regulation of chloroplast development, flowering time and transcription regulation. Project description:The goal of this study was to capture the transcriptional response regulated by the GATA transcription factor encoded by SREB during the first 48-hours of the temperature-dependent shift from yeast (37oC) to mycelia (22oC). Gene expression microarrays were used to compare an SREB null mutant (SREBΔ) to an isogenic wild type strain (ATCC 26199). SREB null mutants fail to fully complete the conversion from yeast to sporulating mycelia after a drop in temperature from 37oC to 22oC, and under iron-replete conditions cannot properly repress siderophore biosynthesis. Deletion of SREB in ATCC 26199 produced SREBΔ. The isogenic wild-type strain is ATCC 26199.

Project description:Plant growth and development are strongly affected by light signals perceived by phytochromes (phy) and cryptochromes (cry). The physical interaction between photoreceptors and the cross-talk among downstream signalling steps creates a network of interactions in which the action of one photoreceptor depends on the status of the others. The processes modulated by phy and or cry include seed germination, seedling de-etiolation, plant body formation and flowering. Compared to the wild type the leaves of the phyB mutant are characterised by their extended petioles and pale colour. These features resemble the phenotype of normal plants grown under the low red to far-red ratios typical of dense plant canopies thus providing support to the idea that phyB plays a key role in the perception of high red to far-red ratios. The triple mutant lacking phyA cry1 and cry2 shows little differences in leaf size and shape with the wild typebut the quadruple phyA phyB cry1 cry2 mutant shows severely impaired leaf expansion. In addition to the specific effects on leaf sizeshape and pigmentation the phyB mutation accelerates flowering in the wild type background. This effect can be observed as a reduced number of days between sowing and visible flower buds (time scale) and as a reduced final number of leaves (developmental scale). However in the phyA cry1 cry2 triple mutant background the phyB mutation accelerates flowering on a developmental scale but severely delays flowering on a time scale. The long photoperiods inducing flowering are perceived by the leaves. Flowering signals that could include hormones and sugars migrate from the leaves to the apex in response to the light stimulus. We propose to compare the following mRNA samples of expanding leaves: 1) wild type 2) phyB mutant 3) phyA cry1 cry2 triple mutant 4) phyA phyB cry1 cry2 quadruple mutant. The comparison between the wild type and the phyB mutant will help to uncover the changes in leaf mRNA patterns underlying enhanced petiole growthreduced pigmentation and early flowering in response to the absence of phyB activity. The comparison between the phyA cry1 cry2 triple mutant and the phyA phyB cry1 cry2 quadruple mutant should reveal the mRNA differences between photosynthetic leaves with dramatically different abilities to expand. The added value of the simultaneous analysis of the two comparisons is that effect of phyB is different in both genetic backgrounds and this should provide information on the mechanisms and significance of photoreceptor interactions.

Project description:Sensing environmental changes is important for survival of plants, as sessile organisms, in habi-tat. Recently, latitudinal clines in leaf senescence have been reported to be influenced by tem-perature and photoperiod. However, the relationship of light quality with latitudinal response of leaf senescence is still unclear. Here, we found that Arabidopsis accessions in far-red (FR) showed a strong negative correlation of leaf senescence with the latitude of their origin. phyto-chrome A (phyA) and B (phyB) mutants showed early and delayed senescence in FR, respec-tively, suggesting that PHYA and PHYB are involved in FR-dependent leaf senescence. Tran-scriptomic analysis identified genes oppositely regulated by PHYA and PHYB, and they were mainly associated with phytohormone, temperature, and defense, suggesting their associations with FR-dependent antagonistic effects of PHYA and PHYB on leaf senescence. Among these genes, the expression of WRKY6 in Arabidopsis accessions showed the highest correlation with latitude of their origin. Consistently, wrky6 mutant exhibited delayed senescence phenotype in FR. In conclusion, we propose that PHYs-regulated genes, including WRKY6, are involved in latitudinal leaf senescence in FR and this regulatory mode may contribute to latitudinal adapta-tion by responding FR, as a piece of geographical information.

Project description:Here we report that FERONIA (FER) integrates phyB-mediated light signaling pathway to control salt stress response. Mutation in phytochrome B (phyB) largely suppresses the dwarfism and leaf bleaching phenotype of fer-4 mutant under salt stress. FER interacts with and phosphorylates the N-terminal domain of phyB. Mutation in fer-4 or disruption of the FER-mediated phosphorylation sites of phyB at Ser106 and Ser227 leads to a retention of phyB in the photobodies under dark conditions, suggesting that FER-mediated phosphorylation accelerates the dark reversion of phyB. Interestingly, salt stress slows down the dark reversion of phyB via the inhibition of the kinase activity of FER, and mutation of phyB or overexpression of PIF5 attenuates the salt-induced growth inhibition. Together, our study indicates that the plasma membrane-localized FER determines the dark reversion rate of phyB in the nucleus via a signature of phosphorylation and thus coordinates the extracellular stress signals and nuclear outputs to dynamically control plant growth and survival under stress conditions.

Project description:Seedling hypocotyls display negative gravitropism in the dark but agravitropism in the light. The Arabidopsis thaliana pif quadruple mutant (pifQ), which lacks four PHYTOCHROME-INTERACTING FACTORS (PIFs), is agravitropic in the dark. Endodermis-specific expression of PIF1 rescues gravitropism in pifQ mutant seedlings. Since phytochromes induce light responses by inhibiting PIFs and the COP1-SPA ubiquitin E3 ligase complex in the nucleus, we asked whether phyB can cell autonomously inhibit hypocotyl negative gravitropism in the endodermis. We found that while epidermis-specific expression of PHYB rescues hypocotyl negative gravitropism and all other phyB mutant phenotypes, endodermis-specific expression of PHYB does not. Epidermal phyB induces the phosphorylation and degradation of endodermal PIFs in response to red light. This induces a global gene expression pattern similar to that induced by red light treatment of seedlings expressing PHYB under the control of its own endogenous promoter. Our results imply that epidermal phyB generates an unidentified mobile signal that travels to the endodermis where it promotes PIF degradation and inhibits hypocotyl negative gravitropism.

Suboptimal, but not near-freezing, temperatures regulate flowering through the thermosensory pathway, which overlaps with the autonomous pathway. Here we show that ambient temperature regulates flowering by two genetically distinguishable pathways, one that requires TFL1 and another that requires ELF3. The delay in flowering time observed at lower temperatures was partially suppressed in single elf3 and tfl1 mutants, whereas double elf3 tfl1 mutants were insensitive to temperature. tfl1 mutations abolished the temperature response in cryptochrome mutants that are deficient in photoperiod perception, but not in phyB mutants that have a constitutive photoperiodic response. Contrary to tfl1, elf3 mutations were able to suppress the temperature response in phyB mutants, but not in cryptochrome mutants. The gene expression profile revealed that the tfl1 and elf3 effects are due to the activation of different sets of genes and identified CCA1 and SOC1/AGL20 as being important cross talk points. Finally, genome-wide gene expression analysis strongly suggests a general and complementary role for ELF3 and TFL1 in temperature signalling.
