Reprogramming of human peripheral blood mononuclear cells into induced mesenchymal stromal cells

ABSTRACT: We reported direct reprogramming human PBMCs into induced MSCs using non-integrating episomal vectors, and performed epigenetic analysis, including chromatin openness and methylation status, along with transcriptomic analysis, on functional as well as non-functional induced MSCs from different reprogramming factors combinations, in order to better understand epigenetic signature of functional MSCs.

Project description:Human induced pluripotent stem (iPS) cells have previously been derived from somatic cells using viral vectors that integrate transgenes into the genome. Genomic integration, however, can allow persistent leaky expression of the transgenes and can create insertional mutations, thus limiting the utility of these cells for both research and clinical applications. Here, we describe the derivation of human iPS cells free of vector and transgene sequences using non-integrating oriP/EBNA1-based episomal vectors. The resulting iPS cells are similar to human embryonic stem (ES) cells in both proliferative and developmental potential. These results demonstrate that reprogramming of human somatic cells does not require genomic integration or the continued presence of exogenous reprogramming factors, and removes one important obstacle to the clinical applications of these cells. Project description:global gene expression were compared among human blood iPSC, human fibroblas iPSC, human embryonic stem cells, human bone marrow MNC and human forskin fibroblast Reprogramming blood cells to induced pluripotent stem cells (iPSCs) provides a novel tool for modeling blood diseases in vitro. we demonstrated that iPSCs free of transgene and vector sequences could be efficiently generated from human bone marrow and cord blood mononuclear cells using non-integrating episomal vectors. The reprogramming described here is up to 100 times more efficient, occurs 1 to 3 weeks faster as compared to the reprogramming of fibroblasts, and does not require isolation of progenitors or multiple rounds of transfection. This approach provides an opportunity to explore banked normal and diseased cord blood and bone marrow samples without the limitations associated with virus-based methods. Generation of Integration-free Induced Neural Stem Cells from Mouse Fibroblasts

Project description:Recent studies demonstrated that fibroblasts could be converted into induced neural stem cells (iNSCs). However, the insertional mutation caused by random integration of viral vectors has been a major limitation of iNSCs for the future clinical translation. Here we show that non-viral transfection of episomal vectors encoding Brn4/Pou3f4, Sox2, Klf4, and c-Myc sufficiently generates iNSCs. The episomal vector mediated iNSCs closely resemble brain-derived NSCs as well as iNSCs generated by retrovirus in morphology, gene expression profile, epigenetic status, self-renewal capacity and both in vitro and in vivo differentiation capacity. The novel conversion protocol defined in the current study offers a method for generating integration-free iNSCs for the clinical research we developed a novel method for generating integration-free iNSCs. Interrupted reprogramming into induced pluripotent stem cells does not rejuvenate human mesenchymal stromal cells

Project description:Replicative senescence hampers application of mesenchymal stromal cells (MSCs) because it limits culture expansion, impairs differentiation potential, and hinders reliable standardization of cell products. MSCs can be rejuvenated by reprogramming into induced pluripotent stem cells (iPSCs), which is associated with complete erasure of age- and senescence-associated DNA methylation (DNAm) patterns. However, this process is also associated with erasure of cell-type and tissue-specific epigenetic characteristics that are not recapitulated upon re-differentiation towards MSCs. In this study, we therefore followed the hypothesis that overexpression of pluripotency factors under culture conditions that do not allow full reprogramming might reset senescence-associated changes without entering a pluripotent state. MSCs were transfected with episomal plasmids and either successfully reprogrammed into iPSCs or cultured in different media with continuous passaging every week. Overexpression of pluripotency factors without reprogramming did neither prolong culture expansion nor ameliorate molecular and epigenetic hallmarks of senescence. Notably, transfection resulted in immortalization of one cell preparation with gain of large parts of the long arm of chromosome 1. Taken together, premature termination of reprogramming does not result in rejuvenation of MSCs and harbours the risk of transformation. This approach is therefore not suitable to rejuvenate cells for cellular therapy.

Characteristics between porcine bone marrow-derived mesenchymal stem and peripheral blood mononuclear cells

Project description:Mesenchymal stem cells (MSCs) have been isolated from various organs and extensively studied for their potential in regulating transplantation. MSCs from different mammalian species are well characterized; however, the properties and therapeutic potential of porcine bone marrow-derived MSCs (BM-MSCs) remain unclear. In this study, we aimed to profile the characteristics of porcine BM-MSCs by comparing their gene expression patterns and immunomodulatory properties with those of porcine peripheral blood mononuclear cells (PBMCs) and bone marrow-attached cells (BMACs). Using quantitative polymerase chain reaction, flow cytometry, immunocytochemistry, and RNA sequencing, we confirmed the expression of key MSC markers, including CD105, CD73, and CD90, in porcine BM-MSCs, and aligned them closely with human MSCs. We found significant differences in gene expression between BM-MSCs and PBMCs, with BM-MSCs exhibiting a distinct expression pattern similar to that of BMACs. Gene ontology enrichment analysis revealed the pathways involved in immune modulation and tissue repair, underscoring the potential of BM-MSCs to enhance immune regulation. Notably, BM-MSCs exhibited higher transforming growth factor-beta levels than PBMCs, suggesting a central role in their immunosuppressive function. These findings indicate the immunomodulatory capabilities of porcine BM-MSCs and support their application in xenotransplantation, where they may help mitigate graft rejection and promote tissue regeneration.
