Project description:To study the ontogeny of pharmacological relevant genes, such as, drug targets, transporters and metabolic enzymes, six wild-type C57BL/6JCrl female mice were sacrified at ten different age points, in order to capture potential transcriptomic switches as the mice progresses from newborn to infant, from infant to teenager, from teenager to adult, and finally from adult to old-adult (P1 - 1 day old; 1W - 1 week old; 2W - 2 weeks old; 3W - 3 weeks old; 1M - 1 month old; 2M - 2 months old; 3M - 3 months old; 6M - 6 months old; 1Y - 1 year old; and 2Y - 2 years old). Liver, heart, kidney, lungs and brain were extracted from all 60 females in the study, and liver samples were processed for RNA extraction. Overall, 8.3% of pharmacological relevant genes changed their expression with age, of which 57% were metabolic enzymes, like Cyp2c29 and Fmo3, and 22% were transporters.

Project description:Although in-hospital mortality rates for sepsis have decreased, survivors often experience lasting physical and cognitive deficits. Moreover, older adults are more vulnerable to long-term complications associated with sepsis. We employed a murine model to examine the influence of age and sex on the brain’s response and recovery following sepsis. Young (~4 months) and old (~20 months) mice (C57BL/6) of both sexes underwent cecal ligation and puncture (CLP) with restraint stress. The hippocampal transcriptome was examined in age and sex-matched controls at 1 and 4 days post-CLP. In general, immune and stress-related genes increased while neuronal, synaptic, and glial genes decreased one day after CLP-induced sepsis. However, specific age and sex differences were observed for the initial responsiveness to sepsis as well as the rate of recovery examined on day 4. Young females exhibited a muted transcriptional response relative to young males and old females. Old females exhibited a robust shift in gene transcription on day 1 and, while most genes recovered, genes linked to neurogenesis and myelination continued to be downregulated by day 4. In contrast, old males exhibited a more delayed or prolonged response to sepsis, such that neuronal and synaptic genes continued to decrease while immune response genes continued to increase on day 4. These results suggest that aging is associated with delayed recovery from sepsis, which is particularly evident in males.

Project description:Aging is associated with major nuclear changes affecting genomic integrity and gene expression. Here we compare the gene expression profiles in the neocortex of young (5 months old) and old (30 months old) B6xC3 F1 mice. In a related study, we compare genes that change expression with age to SIRT1 associated genes identified by ChIP on chip (NimbleGen) and find a significant overlap, indicating that SIRT1 target genes are preferentially deregulated with age. Project description:Analyses of the age-related changes that occur at a gene expression level and transcriptional profile have not been elucidated in depth. To determine the changes of the vascular transcriptome, we conducted gene expression microarray experiments on aortas of adult and old mice, in which age-related vascular dysfunction was confirmed by increased stiffness and associated systolic hypertension C57BL/6 mice obtained from Janvier (Saint Berthevin, France) were aged 6 months (n=6) and 20 (n=6) months and water and standard rodent chow were fed ad libitum. Briefly, the vessel was flushed thoroughly with ice-cold phosphate-buffered saline (PBS), through the left ventricle of the heart, cleaned of periadventitial fat and connective tissues, and snap-frozen in liquid nitrogen and stored at -80 °C.

Project description:We report the mRNA profile of aged mice (24 months old) fed either a control diet or a diet containing Rapamycin (14 ppm) for 3 months. After drug treatement, the hearts of the mice were removed and total mRNA was removed from the tissue. Analysis revealed that there were 700 significantly differentially expressed genes between the control fed group and the Rapamycin diet group by our analysis. Heart tissue samples from age-matched control mice (n=10) and rapamycin fed mice (n=10) were extracted for total RNA. The samples were sequenced using Illumina HiSeq 2000 (50 basepair paired-end sequencing). The sequencing yielded quality scores greater than 30 with an average of 10 million reads per sample. 34,293 genes were mapped back to the MGSCv37 C57BL/6J mouse genome (maximum paired distance=300 and minimum=130, minimum number of reads per mapping = 5, maximum number of mismatches= 2, with the reads being mapped to unique sites in the genome).
