Gene expression analysis of murine alveolar macrophages in the presence of lung cancer cells

ABSTRACT: To investigate the interaction between lung alveolar macrophages and lung cancer cells in vivo, gene expression analysis was performed using orthotopic tumor bearing animal model with C57BL/6 mice and Lewis Lung Carcinoma (LLC) cells. CD45+, F4/80+, Siglec-F+ population was sorted as alveolar macrophage population with fluorescence-activated cell sorting (FACS) technique.

Alveolar Macrophages Maintain Tissue Localization and Gain Enhanced Anti-Tumor Activity in Lewis Lung Carcinoma-Reprogrammed Lung Microenvironment

Project description:The role of alveolar macrophages (AMs) in lung carcinogenesis has been extensively studied, yielding significant insights. However, the status of AMs in tumor-bearing lungs remains incompletely characterized. Using orthotopic Lewis Lung Carcinoma (LLC) mouse models, we found that tumors induced an inflammatory extra-tumoral lung microenvironment (ETLME), distinct from the immunosuppressive tumor microenvironment (TME). T cells with an exhaustion phenotype and tumor-associated macrophages (TAMs) mainly accumulated in the TME rather than the ETLME. Surprisingly, AMs were absent from the tumor lesions and remained in the lung tissues, but they displayed a more active dynamic balance between proliferation and death in ETLME. Furthermore, AMs presented an activated phenotype characterized by upregulation of CD11b and downregulation of Siglec-F, elevated expression of inflammatory genes, and enhanced phagocytic and efferocytotic activity. Notably, AMs in ETLME retained their lipid metabolism capacity and responsiveness to external stimuli. More importantly, LLC-experienced AMs display enhanced anti-tumor ability. These findings indicate that AMs maintain their tissue localization and functional integrity within the ETLME.

Gene expression signatures for murine alveolar type II lung epithelial cells after tumor bearing

Project description:To determine the influence of primary tumors on pre-metastatic lungs, we have employed whole genome microarray expression profiling as a discovery platform to identify gene signatures of alveolar type II epithelial cells (AT-II) in TLR3 deficient mice (Tlr3-/-) and wide-type (WT) littermates with tumor bearing. We subcutaneously inoculated Tlr3-/- and WT mice with Lewis lung carcinoma (LLC). Two weeks later, lung tissues from Tlr3-/- and WT mice were dissociated and AT-II cells were sorted. AT-II cells from mice without tumor bearing were set as controls.

Project description:We performed the transcriptomic analysis of alveolar macrophage(AM) isolated from wild type and HDAC1-2 cKO mice. AM (CD45+CD11c+F4/80+) were sorted from the lung of 2-weeks-old mice. Total RNA from AM was extracted by Trizol reagent according to the manufacturer’s instructions. Library construction and data processing were performed by Beijing Genomics Institute. Libraries were sequenced on Illumina Hiseq4000.

Siglec-1−/FSClow alveolar macrophages exhibit pro-inflammatory phenotypes in chronic obstructive pulmonary disease

Project description:Alveolar macrophages (AMs) in patients with chronic obstructive pulmonary disease (COPD) orchestrate persistent inflammation in the airway, however, sub-populations of alveolar macrophages predominantly participating in chronic inflammation have been poorly characterised. We recently reported that Siglec-1 was involved in recognition and engulfment of non-typeable Haemophilus influenzae and that the expression of Siglec-1 on AMs was decreased in COPD lung tissues. Here, based on the expression of Siglec-1 and the intensity of forward scattergram (FSC) using flow cytometry, we identified three subsets present in alveolar macrophages: Siglec-1+/FSChi, Siglec-1−/FSChi, Siglec-1−/FSClow subsets. The Siglec-1−/FSClow subset increased in number in the lungs of COPD subjects. RNA-sequencing revealed that the Siglec-1−/FSClow subset exhibited pro-inflammatory and protease-producing phenotypes compared to other two subsets. Clinically, a decrease in the intensity of Siglec-1 expression in macrophages of induced sputum from COPD subjects was correlated with exacerbation rates in prospective four years. Collectively Siglec-1− AMs showed the pro-inflammatory properties and that their emergence in COPD airways was a risk factor in exacerbation. The Siglec-1−/FSClow subset might serve as “pathogenic macrophages” which could lead to poor clinical outcomes for patients with COPD

Single-cell RNA sequencing reveals Reg3g-dependent alteration of alveolar macrophage transcriptomes during LPS-induced acute lung injury

Project description:To investigate how Reg3g modulates alveolar macrophage (AM) transcriptional programs during acute lung injury, we performed single-cell RNA sequencing on fluorescence-activated cell sorting (FACS)-purified CD45⁺Ly6G⁻F4/80⁺CD11b/c⁺ lung macrophages from mice treated with PBS, LPS, or LPS + Reg3g for 24 hours. Libraries were generated using the 10x Genomics Chromium GEM-X Single Cell 3′ platform. Following quality control, approximately 29,000 cells were retained. Unsupervised clustering identified 22 distinct cell populations. Reclustering of AMs revealed multiple transcriptional states. LPS exposure shifted AMs toward a pro-inflammatory phenotype, whereas Reg3g treatment attenuated this inflammatory remodeling and restored homeostatic transcriptional signatures. These data identify Reg3g as a modulator of alveolar macrophage functional plasticity during acute lung injury.

To determine genes regulated by TDAG8 in vitro, mouse Lewis lung carcinoma (LLC) cells stably expressing TDAG8 (TDAG8-LLC cells) and control vector (control LLC cells) were cultured under acidic conditions. Transcriptome microarray analysis using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays was performed with total RNA prepared from these cells. To determine genes regulated by TDAG8 in vitro, mouse Lewis lung carcinoma (LLC) cells stably expressing TDAG8 (TDAG8-LLC cells) and control vector (control LLC cells) were cultured under acidic conditions. Transcriptome microarray analysis using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays was performed with a mixture of equal amounts of total RNA samples from four independent cell cultures.
