Effect of Atm Inhibitor on Galactose-Induced Cataract in Rat Lens

ABSTRACT: Analysis of gene expression changes in galactose-induced cataracts when an Atm inhibitors is added. A total of six samples were analyzed: a sample without induced turbidity (control), three samples in which turbidity was induced by galactose (galactose), and a sample with ATM inhibitor added (KU55933 or AZD0156).

Project description:We investigated the therapeutic effects of histone acetyltransferase (HAT) inhibitors on cataracts using an ex vivo model of galactose-induced cataracts in the rat lens.Among the tested HAT inhibitors, TH1834 was the only inhibitor that could completely reverse white opacities once formed in the lens. Combination treatment with C646/CPTH2 and CBP30/CPTH2 also had therapeutic effects. To identify the genes regulated by HAT inhibitors, we conducted microarray analysis of treated and untreated cataract samples.

Pyruvate is effective in restoring cataracts formed by galactose-induced cataract models.

Project description:The addition of Pyruvate to the lens induced by galactose eliminated the white clouding formed and showed a therapeutic effect. Genes regulated by pyruvate addition were identified by microarray analysis. A total of seven samples were used in this analysis: two samples without galactose (control), three samples with galactose (galactose) and two samples with Pyruvate (Pyruvate).

Transcriptomic analysis of human lens and age-related cataract

Project description:As an ageing population trend, age-related cataracts (ARC) continue to be the primary cause of reversible severe vision impairment and blindness worldwide, with an increasing incidence. The primary disease mechanism of ARC formation is metabolic abnormalities and oxidative stress mainly caused by increasing age. Additionally, ultraviolet B exposure, systematic disease factors (diabetes and hypertension), and lifestyle factors (smoking, drinking, and malnutrition) are the risk factors for ARC formation. Although surgical removal of the onset lens and replacement of the intraocular lens is currently the only effective treatment method with a high success rate and a high-cost fee, cataracts have so far lacked effective therapeutic drugs leading to a heavy burden on the patient's family and society. Therefore, we performed transcriptomic analysis of human lens and ARC to investigate the possible pathogenesis of lens opacification in ageing people.

Response to LiCl of galactose grown cells

Project description:Wild type strain CEN.PK113-7D was grown in an aerobic batch cultivation with a start concentration of 20 g/L galactose. During exponential growth at a biomass concentration of 3 g dry weight/L LiCl was added to a concentration of 10 mM. Just before addition of LiCl (time 0) and 20, 40, 60 and 140 minutes after addition of LiCl samples were taken for transcription analysis. Transcriptome profiling from Wild Type and Lss mutant (heterozygous and homozygous) lens at E14.5 reveals the potential pathways involved in cataract formation

Project description:Congenital cataract is one of the leading causes of blindness in children worldwide. About one third of congenital cataracts are caused by genetic defects. Previous studies have identified LSS as a causal gene for congenital cataracts. However, its roles in lens development remains largely unknown. Here, we performed RNA-seq on the lens of WT and Lss mutant (heterozygous and homozygous )at E14.5, identified that cholesterol synthesis signaling pathways were significantly downregulated. Overall, our study points out that LSS functions as a critical determinant of lens development, which will contribute to better understand LSS defects in cataractogenesis and develop therapies for cataract.

Galactose is the next most abundant sugar in many hemicelluloses and it will be important to capture this carbon for an efficient bioconversion process of plant biomass. Thermophilic fugus Myceliophthora thermophila has been used as cell factory to produce biochemicals directly from renewable polysaccharides. This study determined the transcriptomic profiles of M. thermophila responding to galactose and identified the gene involved in the oxido-reductive pathway for galactose degradation, of which hexokinase might be the rate-limiting enzyme in this thermophilic fungus. Galactokinase was necessary for high induction of galactose transporter and disruption of galK resulted in decreased galactose utilization. Both of galactokinase deletion and sugar transporter overexpression could further activate the oxido-reductive pathway and led to improved galactose utilization. In addition, disruption of the Leloir pathway brought about reduced tolerance to cell wall perturbation, oxidative stress and high osmolality. In order to accelerate galactose consumption, the third galactose-degradation pathway- De Ley-Doudoroff pathway was successfully integrated into M. thermophila and the consumption rate of galactose was increased by 57%. This study will be benefit for the rational design of fungal strains to produce biofuels and biochemical from plant biomass, especially, marine plant biomass with the abundant of galactose, such as red seaweed and molasses.

FATC domain deletion compromises ATM protein stability, lymphocyte development, and promotes lymphomagenesis

Project description:ATM kinase is a master regulator of the DNA damage response and loss of ATM leads to primary immunodeficiency and greatly increased risk for lymphoid malignancies. The FATC domain is conserved in Phosphatidylinositol-3-kinase-related protein kinases (PIKKs). Truncation mutation in the FATC domain (R3047X) selectively compromised reactive oxygen species-induced ATM activation in cell-free assays. Here we show that in mouse models, knock-in ATM-R3057X (AtmRX, corresponding to R3047X in human ATM) mutation severely compromises ATM protein stability, and causes T cell development and B cell immunoglobulin class switch recombination defects and infertility resembling ATM-null. The residual ATM R3057X protein retains minimal, yet functional measurable, DNA damage-induced checkpoint activation and significantly delays lymphomagenesis in AtmRX/RX mice compared to Atm-/-. Together, these results support a physiological role of the FATC domain in ATM protein stability and show that minimal ATM activity can prevent growth retardation and delay tumorigenesis without restoring lymphocyte development and fertility.
