MRNA profiling of schwann cells cultured with monocytes derived macrophages and PDAC cell lines supernants-induced tumor-associated macrophages (TAMs)

ABSTRACT: Tumor associated macrophages(TAMs) have been demonstrated to promote tumor progression and perineural invasion in PDAC, however the underlying machanism of TAMs interacting with Schwann cells is still unclear. We found the gene expression of Schwann cell is different from TAMs-induced Schwann cells by comparing mRNA profiling.

Project description:Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease with high resistance to therapy. Inflammatory and immunomodulatory signals co-exist in the tumor microenvironment (TME), leading to dysregulated reparative and cytotoxic responses. Tumor-associated macrophages (TAMs) control immune dynamics in the TME, but their heterogeneity and plasticity have hampered understanding of the underlying mechanisms. Here, we combined single-cell and spatial genomics with functional experiments to elucidate macrophage functions in PDAC. We uncovered an inflammatory loop between tumor cells and interleukin (IL)-1b+ TAMs, a subset of macrophages elicited by a local synergy between prostaglandin E2 (PGE2) and tumor necrosis factor (TNF)-a. Interfering with the PGE2-IL-1b axis elicited TAM reprogramming and antagonized tumor-intrinsic inflammation, leading to PDAC control in vivo. IL-1b+ TAMs are conserved across human cancers and correlate with inflammation and patient prognosis in a context-dependent manner. Our study highlights how TAMs govern between inflammation and immune suppression in cancer, with significant therapeutic implications.

Schwann Cells Shape Tumor Cells and Cancer-Associated Fibroblasts in the Pancreatic Ductal Adenocarcinoma Microenvironment

Project description:Neuropathy is a feature more frequently observed in pancreatic ductal adenocarcinoma (PDAC) than other tumors. Schwann cells, the most prevalent cell in peripheral nerves, migrate toward tumor cells and associate with poor prognosis in PDAC. To unveil the effects of Schwann cells on the neuro-stroma niche, single-cell RNA-sequencing and microarray-based spatial transcriptome analysis of PDAC tissues were performed. Results suggested that Schwann cells may drive tumor cells and cancer-associated fibroblasts (CAFs) to more malignant subtypes: basal-like and inflammatory CAFs (iCAFs), respectively. Moreover, in vitro and in vivo assays demonstrated that Schwann cells enhanced the proliferation and migration of PDAC cells via Midkine and promoted the switch of CAFs to iCAFs via interleukin-1a. By Schwann cells condition medium, tumor cells together with iCAFs accelerate PDAC progression. Thus, we revealed, for the first time, that Schwann cells induce malignant subtypes of tumor cells and CAFs in the PDAC milieu.

Expression data from in vitro-induced non-hypoxic tumour associated macrophages (TAMs)

Project description:Active immunotherapy is a promising strategy for anti-angiogenic cancer therapy. Recently, we have reported that a vaccine using human umbilical vein endothelial cells (HUVECs) induced specific anti-endothelial immune responses in the most of immunized patients, and resulted in tumor regression in some patients with recurrent malignant brain tumors, whereas not in colorectal cancer patients. In this study, we hypothesized that non-hypoxic perivascular tumor associated macrophages (TAMs) in colorectal cancer, but not in glioblastoma, might negatively alter the therapeutic efficacy of anti-angiogenic active immunotherapy. To test this hypothesis, we examined global gene expression profiles of non-hypoxic macrophages stimulated in vitro by soluble factors released from tumor cells of human glioblastoma U-87MG (‘brain TAMs’) or colorectal adenocarcinoma HT-29 (‘colon TAMs’). Murine non-hypoxic TAMs were induced in vitro by incubation with soluble factors released from human cancer cell lines U-87MG ('brain TAMs') or HT-29 ('colon TAMs'), for RNA extraction and subsequent hybridization on Affymetrix microarrays. To evaluate homogeneous macrophage populations at different tumour developmental stages, RNA aliquots of control macrophages and TAMs obtained at five different time-points, i.e. 8h, 16h, 24h, 32h and 40h, were pooled and used for screening of differentially expressed genes. The experiments for TAMs as well as for control unstimulated macrophages were performed in triplicates.
