Cleavage Under Targets and Tagmentation（CUT&Tag）analysis of ovarian cancer cells treated by Paeonol

ABSTRACT: Cells were harvested and aliquoted (500,000 cells). Cell nucleus was prepared and resuspended. Incubation of the cells was performed with ACSS2 and IgG primary antibodies throughout the night at 4℃. The cells were centrifuged and washed using Dig-Wash buffer. A dilution of the pA-Tn5 adapter complex was introduced into the cells, followed by incubation at RT for 1 h. Subsequently, the cells were subjected to incubation in Tagmentation buffer for 1 hour at 37℃, and the STOP buffer was eventually added. The DNA library was purified and amplified for sequencing, and CUT&Tag data analyses were performed.
