Single-cell sequencing of mouse peripheral blood mononuclear cells

ABSTRACT: Systemic immune cells dysregulation, especially the inflammatory factors over-production plays the pivotal role in sepsis induced abnormalities. We further analyzed how HDCA affects the systemic immune cells during sepsis by employing scRNA-seq for PBMCs.

Project description:This study focuses on analyzing the gene expression profiles of peripheral blood mononuclear cells (PBMCs) in mice following the induction of sepsis, using RNA sequencing (RNA-seq). PBMCs play a central role in the immune response, and their transcriptional changes can provide critical insights into the systemic immune dysregulation that occurs during sepsis. By identifying key genes and pathways that are altered in response to sepsis, this research aims to uncover molecular mechanisms driving disease progression and immune dysfunction. The findings could pave the way for the development of targeted therapies or diagnostic biomarkers to improve the management and treatment of sepsis.

Project description:To identify signature genes that help distinguish (1) sepsis from non-infectious causes of systemic inflammatory response syndrome, (2) between Gram-positive and Gram-negative sepsis. Experiment Overall Design: A total of 70 critically ill patients (46 sepsis and 24 control) were enrolled in a single-centre observational study. Gene-expression profiling was performed using Affymetrix microarray (U133plus2) with 54,675 transcript. Data was divided into a training set (n=35) and a validation set (n=35). A molecular signature was developed in the training set and was then validated in the validation set.

Neddylation tunes peripheral blood mononuclear cells immune response in COVID-19 patients

Project description:The COVID-19 pandemic caused by SARS-CoV-2 has reached 5.5 million deaths worldwide, causing a huge impact globally. This highly contagious viral infection produces a severe acute respiratory syndrome that includes cough, mucus, fever and pneumonia. Likewise, many hospitalized patients develop severe pneumonia associated with acute respiratory distress syndrome (ARDS), besides an exacerbated and uncontrolled systemic inflammation which in some cases induce lethal cytokine storm. Although vaccines have clearly had a beneficial effect on disease development, there is still a high percentage of patients who develop pathology related to ineffective immune system response. Therefore, a thorough understanding of the modulatory mechanisms that regulate the response to SARS-CoV-2 is crucial to find effective therapeutic alternatives. Previous studies describe the relevance of Neddylation in immune system activation further its implications in viral infection. In this context, the present study postulates Neddylation, a reversible ubiquitin-like post-translational modification of proteins and controls their stability, localization and activity, as a key regulator in the immune response against SARS-CoV-2. For the first time, we describe an increase of serum global neddylation levels of COVID-19 patients particularly associated in the early response of the infection. In addition, the results showed that overactivation of neddylation control activation, proliferation, and response of peripheral blood mononuclear cells (PBMCs) derived from COVID-19 patients. Inhibition of neddylation and subsequent avoidance of activation of PBMCs, which reduces cytokine production and proteome modulation, may therefore be a critical mechanism and an efficient therapeutic approach to immunomodulate COVID -19 patients and avoid the much-feared cytokine storm.

Microarray profiling peripheral blood mononuclear cells

Project description:We performed RNA-Seq transcriptome profiling on 29 immune cell types consituting peripheral blood mononuclear cells (PBMCs) sorted from 4 Singaporean-Chinese individuals (S4 cohort). We also performed RNA-Seq and microarray transcriptome profiling of PBMCs from an extended cohort of 13 individuals (S13 cohort). The data was used first to characterize the transcriptomic signatures and relationships among the 29 immune cell types. Then we explored the difference in mRNA composition in terms of transcripts proportions and abundance. Lastly, we performed deep deconvolution for both microarray and RNA-Seq technologies.

Obesity regulates the fate of peripheral blood mononuclear cells through FoxM1

Project description:Obesity is associated with various diseases such as metabolic disorders, joint problems, and inflammations. Over 50% of pets, including dogs and cats, are obese, making a comparative study necessary at gene level. Since circulating peripheral blood mononuclear cells (PBMCs) reflect the organs related to obesity, we compared the mRNA expression in normal and obese PBMCs of dogs. Our findings show that there are 425 differentially expressed genes (DEGs) (p < 0.05, |log2 fold| ≥ 1) that exhibit distinct gene expression between normal and obese PBMCs. In Kyoto encyclopedia of genes and genomes (KEGG) and Ingenuity Pathway Analysis (IPA) indicate that obesity affect the PBMC cell cycle through FoxM1. These results suggest that obesity causes functional change of PBMCs through cell cycle regulation. In conclusion, obesity suppresses the PBMCs-mediated immune response through FoxM1.

Goat Peripheral Blood Mononuclear Cells (PBMCs) responses to infection vary with Peste des Petits Ruminants Virus virulence

Project description:Peste des Petits Ruminants (PPR) is a highly infectious disease caused by a virus of the genus Morbillivirus (PPRV), infecting mainly sheep and goat. Susceptibility of host can vary widely with host breed and virus strain. The mechanisms underlying this variability are not well understood. Here, we carried out the first comparative in vitro study on goat peripheral blood mononuclear cells (PBMCs) infected with PPRV strains of different virulence, vaccine strain (N75/1), low- (IC89), and high-virulent strain (MA08). Goat PBMCs were infected by the different strains and stimulated with Concanavalin A (Con A), and proteome changes of these cells were evaluated during the infection. The level of PPRV replication was assessed first by RT-qPCR quantification of viral N mRNA and by labelling the viral N protein inside the cells with specific antibodies and analysed by flow cytometry. The dynamics of PBMC sub-populations were also evaluated by flow cytometry. Our results showed that viral replication is critical for PPRV inhibitory effect on PBMCs proliferation. Highly virulent MA08 strain reached a higher level of replication in ConA-stimulated PBMCs and induced higher mortality compared to other strains. Low-virulent IC89 strain showed the lower replication level and cell proliferative inhibitory capacities. Differences in immune genes expression levels were assessed using RNAseq analysis and protein expression was compared using mass spectrometry. Analysis of these data provided the transcriptional and proteome landscape of PBMCs infected with these strains. The possible association between the transcriptional and proteome landscape and changes in immune cell subpopulations dynamics was explored.
