Saliva methylome analysis of ART children that underwent embryo culture in G3 or K-SICM medium

ABSTRACT: Follow-up study of 9 year old IVF children who underwent embryo culture in G3 (Vitrolife) or K-SICM (Cook) medium. Genome-wide DNA methylation profiling of 9 year old IVF children (saliva samples) who had undergone embryo culture in G3 medium (Vitrolife) or K-SICM medium (Lonza). The EPIC array was used to profile the methylome at approximately 850,000 CpG sites across the human genome.

Effect of embryo in vitro culture on mESC transcriptome.

Project description:Transcriptome analysis of mouse embryonic stem cell lines derived from embryos cultured in optimal and suboptimal conditions compared to cell lines derived from control embryos. The use of assisted reproductive technologies (ART) such as in vitro fertilization (IVF) has resulted in the birth of more than 5 million children. While children conceived by these technologies are generally healthy, there is conflicting evidence suggesting an increase in adult-onset complications like glucose intolerance and high blood pressure in IVF children. Animal models indicate similar potential risks. It remains unclear what molecular mechanisms may be operating during in vitro culture to predispose the embryo to these diseases. One of the limitations faced by investigators is the paucity of the material in the preimplantation embryo to test for molecular analysis. To address this problem, we generated mouse embryonic stem cells (mESC) from blastocysts conceived after natural mating (mESCFB) or after IVF, using optimal (KSOM + 5% O2; mESCKAA) and suboptimal (Whitten’s Medium, + 20% O2, mESCWM) conditions.

Project description:Assisted reproductive technologies, including in vitro fertilization (IVF), are now frequently used, and increasing evidence indicates that IVF causes gene expression changes in children and adolescents that increase the risk of metabolic diseases. Although such gene expression changes are thought to be due to IVF-induced epigenetic changes, the mechanism remains elusive. We tested whether the transcription factor ATF7, which mediates stress-induced changes in histone H3K9 tri- and di-methylation, the typical epigenetic silencing marks, is involved in the IVF-induced gene expression changes. We investigated the liver transcriptome in 3-week-old mice generated by IVF and normal mating.

Characterization and quantification of proteins secreted by single human embryos prior to implantation

Project description:The use of in vitro fertilization (IVF) has revolutionized the treatment of infertility and is now responsible for 1-5% of all births in industrialized countries. During IVF, it is typical for patients to generate multiple embryos. However, only a small proportion of them possess the genetic and metabolic requirements needed in order to produce a healthy pregnancy. The identification of the embryo with the greatest developmental capacity represents a major challenge for fertility clinics. Current methods for the assessment of embryo competence are proven inefficient and the inadvertent transfer of non-viable embryos is the principal reason why most IVF treatments (approximately two-thirds) end in failure. In this study, we investigate how the application of proteomic measurements could improve success rates in clinical embryology. We describe a procedure that allows the identification and quantification of proteins of embryonic origin, present in attomole concentrations in the blastocoel, the enclosed fluid filled cavity that forms within five-day old human embryos. By using targeted proteomics, we demonstrate the feasibility of quantifying multiple proteins in samples derived from single blastocoels, and that such measurements correlate with aspects of embryo viability, such as chromosomal (ploidy) status. This study illustrates the potential of high-sensitivity proteomics to measure clinically relevant biomarkers in minute samples and, more specifically, suggests that key aspects of embryo competence could be measured using a proteomic-based strategy, with negligible risk of harm to the living embryo. Our work paves the way for the development of “next-generation” embryo competence assessment strategies, based on functional proteomics.

Project description:The DKAT cell line is a novel model of triple-negative breast cancer that was isolated from the pleural effusion of a 35 year-old caucasian woman with triple-negative breast cancer. When cultured in serum-free media (MEGM, Lonza) this cell line exhibits an epithelial morphology and gene expression pattern. However, when cultured in the presence of serum (SCGM, Lonza) it undergoes a reversible EMT. We used microarrays to look at gene expression changes in the DKAT cell line when cultured in Mammary Epithelial Growth Media (MEGM, where DKAT cells have an epithelial morphology) vs Stromal Cell Growth Media (SCGM, where DKAT cells have a mesenchymal morphology).
