Project description:As muscle function is relevant for the stress response, and as gene array and proteomics analyses have not been conducted to investigate muscle response in stress paradigm in birds, we developed generic approaches to assess molecular bases of muscle response to stress in chicken. Restraint test was chosen as stress situation because this treatment has previously been shown to be a high stressful situation in birds and mammals. Project description:The process of commercial catching, transport and slaughter (CTS) is known to be an acute stressful event in broiler chickens. Corticosteroid concentrations increase, impacting measures of IGF-1, growth hormone and metabolites of the immune system from blood plasma samples. We used ARK-Genomics chicken 20K oligo array, a two channel DNA microarray, to investigate the significantly differentially expressed genes in the livers of chickens during CTS. We investigate the differences of gene expression profiles in hepatic tissues between control birds (n=10) and birds experiencing CTS (n=10) using an ARK-Genomics chicken 20K oligo array, a two channel DNA array (http://www.ark-genomics.orgmicroarray) with full dye swap.

Project description:By using a 44k chicken Agilent microarray, we systematically analyzed the chicken hypothalamus transcriptome response to thermal stress. Twelve hypothalamus samples were chosen from three groups (four samples per group) to be used in chicken genome microarray to examine differential gene expression.We compared the expression profiles between each pairs of the three groups using the microarray data. Totally, 2474 genes were found to be differentially expressed in the three comparisons with p＜0.05 and fold change (FC) higher than 1.5 and the genes were mainly involved in self-regulation and compensation required to maintain homeostasis, including heat shock protein family, enzyme and the hormone, neurotransmitter, cell-cell signaling, metabolism and cytokines. The transcripts of heat shock protein including Hsp 40 and Hsp 90 were significantly changed respond to thermal stress and genes involved in regulation of cell morphogenesis were significantly upregulated in heat stressed group with comparison to control and temperature recovery group. Additionally, the down-regulated genes in both heat stress and temperature recovery groups compared to control group were enriched in muscle organ development, striated muscle tissue development, cardiac muscle tissue development and muscle tissue development, which indicates that muscle development was inhibited during and in short-term after heat treatment. Most of genes dysregulated in heat stress group were found to be recovered in temperature recovery group, which confirmed their roles they could play in coping with heat stress. The present study provides a broader understanding of molecular mechanisms underlying the stress response in chicken and discovery of novel genes that are regulated in a thermal stress specific manner.

Project description:Heat stress is a very complex physiological process. At present, there are some reports about physiological function and pathological changed on chicken heat stress. We use DNA microarrays to detail the global programme of gene expression underlying heat stress and identified distinct classes of differentially expressed (DE) genes during this process. Chicken were randomly divided into two groups to study the effects of heat stress on gene expression of brain, liver and leg muscle. Twenty-four birds (12 male and 12 female) were allocated to heat stress or post-heat stress groupM-oM-<M-^H40M-BM-11M-BM-0C,3hM-oM-<M-^I and control or pre-heat stress groupM-oM-<M-^H28M-BM-11M-BM-0CM-oM-<M-^I.

Project description:As an attempt to assess physio-psychological effects elicited in odorant-inhaled rats, gene expression profiling in the central nervous system was carried out with rats housed under stressful conditions. (R)-(-)-linalool inhalation to rats during 2 h restraint significantly up-regulated the expression of genes in hypothalamus, which were found to be related to neuron differentiation and regulation of transcription as well as immediate early genes. The expressions of 104 among focused stress-regulated genes were significantly altered by the inhalation. The (R)-(-)-linalool inhalation significantly repressed the restraint-induced changes in the expression levels of 77 of the 104. It also promoted the expression of the remaining 27 genes including those related to anti-apoptotic heat shock proteins. The differences in their hypothalamic gene expressions revealed that the inhaled odorants actually influenced stress responses, based on the restraint-induced hypothalamic gene expressions related to apoptosis. These results indicate that the analysis of gene expression profiles in rats subjected to a stressful condition is useful to evaluate odorant-induced effects as shown by the particular results that (R)-(-)-linalool inhalation under only 2 h restraint- stressed condition induces neuron differentiation against apoptosis.

Project description:Domesticated animals share a unique set of morphological and behavioural traits, jointly referred to as the domesticated phenotype. These include modified growth, reproduction, metabolism, pubertal development and stress response. Striking similarities amongst a range of unrelated domesticated species suggest that similar regulatory mechanisms may underlie the domesticated phenotype. Several previous studies have focused on the brain to find mechanisms underlying domestication effects on the stress response, whereas the potential role of the pituitary gland as a target of domestication is highly overlooked. Here, we study gene expression in the pituitary gland of the domesticated White Leghorn chicken and its ancestor, the Red Junglefowl. We exposed chicken of both breeds to 15 minutes of restraint stress. We then culled the animals and dissected out the pituitary gland and snap froze them. RNA was later extracted from the pituitaries and gene expression was measured using Agilent microarray.
