Project description:Analysis of baseline gene expression in bone marrow derived dendritic cells (BMDC) from female CBA/J (CBA) and C57BL/6 (BL/6) mice. Results provide insight into strain-dependent differences in gene expression. CBA and BL/6 BMDCs prepared from individual mice were plated in replicate at 1x10^6 cells/ml in 48-well tissue culture plates (BD Falcon). Replicates were pooled after 4hr and total RNA was obtained by TrizolM-BM-. (Invitrogen) extraction according to the manufacturerM-bM-^@M-^Ys instructions.

Gene expression data in Bone Marrow Derived Dendritic Cells (BMDC) following nanoemulsion adjuvant exposure

Project description:Antigen uptake, processing and presentation by dendritic cells are regulated by complex intra- and inter-cellular signalling events. Typical vaccine adjuvants lead to the transcription of pro-inflammatory cytokines and chemokines which relate to immune induction. We used microarrays to detail the global expression of genes in BMDC underlying emulsion internalization and DC activation with a non-inflammatory nanoemulsion adjuvant. BMDC were incubated with 0.001% v/v naomeulsion (W805EC) or media alone for 6 hours. Total RNA was extracted per sample using RNA easy (Qiagen).

Project description:Purpose: To determine H3K4me3 chromatin profile in UBC9WT and UBC9KO BMDC before and after LPS stimulation. Methods: H3K4me3 chromatin profile was determined by sequencing UBC9 WT and UBC9 KO BMDC chromatin immunoprecipitated with antibody specific for H3K4me3. Results: We show differential chromatin profile for H3K4me3 on the ifnb1 locus between UBC9 KO cells and UBC9 WT. Conclusions: Loss of SUMOylation causes a deregulation of ifnb1 transcription activation mark.
