Changes in miRNA expression profile upon treatment with plumbagin or menadione in SW-620 colorectal cancer cells

ABSTRACT: To elucidate the mode of actions of Plumbagin and Menadione, miRNA-profilling was performed after treating cells with vehicle (DMSO), Plumbagin (10uM), and Menadione (10uM) for 24 hours. Threel replicates for control and Menadione and two replicates for Plumbagin were subjected to miRNA-microarray.

Expression data from MCF-7 breast cancer cells treated with Plumbagin (PL) and its derivative Acetyl Plumbagin (AP)

Project description:The study aimed at finding the molecular mechanism of action of PL and AP in breast cancer cells. The dataset shows the comparison of gene expression in Plumbagin/Acetyl Plymbagin treated MCF-7 cells when compared with untreated samples. 11 samples were analyzed. MCF-7 cells treated with 10 uM PL or AP for 6 hours. Please note that three samples were done in triplicates and one sample MCF-7 treated with 10uM PL was done in duplicates as one of the replicate RNA didn't show up a good RIN number, SAMPLE1 &2 are replicates, SAMPLE 3,4 &5 are triplicates, SAMPLE 6,7 & 8 are triplicates and SAMPLE 9,10 & 11 are triplicates (SAMPLE numbers are indicated in the description field).

Differential gene expression induced by anti-cancer agent plumbagin is mediated by androgen receptor in prostate cancer cells

Project description:Plumbagin treatment of mice harboring PTEN-P2 tumors in the prostate or on prostate tissue maintained in vivo results in tumor regression when coupled with androgen deprivation therapy. This suggested that dihydrotestosterone (DHT) production in the testes prevents tumor regression due to plumbagin treatment by an unknown mechanism. We performed RNA-seq analysis on cells treated with plumbagin, DHT, or both, and analyzed differential gene expression using RNA-seq. DHT and plumbagin synergize to differentially regulate many genes that are not differentially regulated by either agent alone. For many genes, increases in mRNAs caused by DHT are sharply down-regulated by plumbagin, and some transcripts increase or decrease in response to plumbagin in a DHT-dependent manner. We also observed that plumbagin causes extensive RNA damage independently of DHT. We also include data from a simple time course experiment in which gene expression in response to DHT was studied using RNA-seq.

Time course of oxidative stress responses in yeast, caused by Menadione or hydrogen peroxide

Project description:Oxidative stress caused by Menadione or Hydrogen peroxide in synchronized Saccharomyces cerevisiae cultures. Alpha factor synchronized cultures (0.2-0.4 OD), treated at the beginning of S phase (25 min after release from G1 arrest) with either 2 mM Menadione (MD) or 0.24 mM Hydrogen peroxide (HP), show cell cycle effects. Cells treated with MD arrested at G1. Cells treated with HP delayed at S and then, after removal of HP at 135 minutes , continued the cell cycle, only to arrest at G2/M. Growth was carried out in 30C with shaking (295 rpm). Two time course experiments were performed with each oxidative stress agent, designated as H2O2 and H2O2_II, MD and MD_II. Project description:Investigation of whole genome gene expression changes at short (2 hours) and extended (24 hours) timepoints in wild-type Saccharomyces cerevisiae treated with 50 μM menadione during exponential growth compared to an rph1Δ strain Transient treatment with 50 μM menadione elevates mitochondrial ROS and extends chronological lifespan in yeast. Deletion of RPH1, a H3K36me3 histone demethylase, block chronological lifespan extension. This study aimed to identify Rph1p-dependent gene expression changes induced by menadione treatment that may support chronological lifespan extension.
