Expression data of wild-type and dcl1-15 torpedo-staged embryos

Project description:Severe loss-of-function alleles of DCL1 are embryonic lethal. Defects in cell division were seen as early as the globular stage in the strong loss-of-function allele dcl1-15. Phenotypic work with dcl1-15 and the null allele dcl1-5 suggested that, in addition to the severe patterning defects, the mutants were maturing earlier than wild-type embryos. We performed global gene expression analysis of dcl1-15 and wild-type torpedo staged embryos to examine this maturation phenotype further. The results suggested that DCL1 is a heterochronic gene. Comparisons to a time series of embryo development (http://www.seedgenenetwork.net/arabidopsis) showed that the genes differentially expressed in dcl1-15 embryos behaved more like green-cotyledon stage embryos than torpedo embryos. Seeds of a DCL1/dcl1-15 plant were sown. For each wild-type sample, 300 torpedo stage embryos were selected from wild-type siblings. For each mutant replicate, 300 dcl1-15/dcl1-15 embryos were selected from the siliques of DCL1/dcl1-15 where the wild-type embryos were at the torpedo stage.

Expression differences between zebrafish embryos lacking b3glct and wildtype sibs

Project description:Heads and trunk tissues were isolated from zebrafish embryos either wildtype or deficient for b3glct to identify differentially regulated genes which may explain lack of phenotype. Heads and trunks were dissected, solublized in trizol and purified RNA was sent to OakLabs for transcriptional profiling. The expression of mutant to normal tissues was compared to identify differentially regulated genes shared by both head and trunk tissue in mutant embryos.

Transcription profiling of D.melanogaster twist homozygous mutants vs wildtype embryos at developmental stages

Project description:Expression profiling of Drosophila twist homozygous mutant embryos in a timecourse of embryogenesis. Four one-hour timepoints were assayed (4-8 hrs after egg laying). Homozygous mutant embryos were collected using GFP-activated embryo sorting at the same time as stage-matched wildtype controls. Four independent collections were performed at each timepoint. Total RNA was extracted and amplified. Mutant and respective wildtype samples were hybridized together to INDAC oligonucleotide arrays.

Peroxide antimalarial treatment of K13-mutant and -wildtype P. falciparum parasites

Project description:Red blood cells (RBCs) infected with trophozoite stage P. falciparum parasites (Cam3.IIR539T or Cam3.IIrev lines) at 4% parasitaemia and 2% haematocrit were treated with 100 nM of DHA, OZ277 or OZ439 for a duration of 1, 3 and 5 h, respectively. The K13-mutant artemisinin resistant parasite line used was Cam3.IIR539T. The K13-wildtype artemisinin sensitive parasite line used was Cam3.IIrev. The Samples treated with vehicle (DMSO) acted as the untreated control.
